Screening for low grain cadmium phenotypes in sunflower, durum wheat and flax   总被引:10,自引:0,他引:10  
Cadmium (Cd) level in nonoilseed sunflower (Helianthus annuus L.), flax (Linum usitatissimum L.), and durum wheat (Triticum turgidum L. var. durum) grown on uncontaminated, alkaline soils has exceeded limits established in Northern Europe. Separate field experiments were conducted to investigate variability of grain Cd levels among sunflower, durum wheat and flax germplasm, and to seek an efficient screening method for future breeding. There were large variations in leaf Cd concentration among 200 sunflower lines. These lines performed more consistently for Cd uptake at the R5 stage than at the V8 stage across 4 locations with markedly differing soils. Cd concentration in V8 leaves was not related to Cd in grain. The positive correlation between R5 leaf Cd and kernel Cd level was obtained from nonoilseed hybrid (Sigco 954) (R2; = 0.74**), and 200 lines (R2 = 0.44**) tested over 4 locations in 2 field trials, respectively. This indicates that an efficient and low cost screening method can be developed for genotype selection, but plants must be grown to the R5 stage. A preliminary evaluation of 30 durum wheat and 74 flax lines indicated large variations in grain Cd level of durum wheat and flax. Grain Cd concentration ranged from 0.11 to 0.34 mg Cd kg-1 DW for durum wheat, and 0.14 to 1.37 mg Cd kg-1 DW for flax, respectively. This variability indicates that breeding for low grain Cd in durum wheat and flax should be feasible. The mechanism that controls the proportion of cannabichromene (CBC), a potential pharmaceutical, in the cannabinoid fraction of Cannabis sativa L. is explored. As with tetrahydrocannabinol (THC) and cannabidiol (CBD), CBC is an enzymatic conversion product of the precursor cannabigerol (CBG). CBC is reported to dominate the cannabinoid fraction of juveniles and to decline with maturation. This ontogeny was confirmed in inbred lines with different mature chemotypes. A consistent CBC presence was found in early leaves from a diverse clone collection, suggesting that CBC synthase is encoded by a fixed locus. Morphological variants possessing a ‘prolonged juvenile chemotype’ (PJC), a substantial proportion of CBC persisting up to maturity, are presented. PJC is associated with a reduced presence of floral bracts, bracteoles, and capitate-stalked trichomes. Genetic factors causing these features were independent of the allelic chemotype locus B that was previously postulated and regulates THC and CBD synthesis and CBG accumulation. In contrast to previously described Cannabis chemotypes, the cannabinoid composition of PJCs showed plasticity in that reduced light levels increased the CBC proportion. The ability of PJC plants to enable the production of pharmaceutical raw material with high CBC purity is demonstrated.  相似文献

The equilibrium method is introduced for the detection of invert sugar addition to apple juice. The method consists of a pre-equilibration of the sample with dry pyridine at 50 degrees C for 20 min followed by the addition of trimethylsilylimidazole and heating at 75 degrees C for 40 min. The resulting derivatized carbohydrates are then analyzed by capillary gas chromatography. This method was successfully used by independent laboratories to distinguish heated pure, intentionally adulterated (with invert sugar), and intentionally adulterated and then heated apple juice concentrates. The equilibrium method was shown to give significantly lower coefficients of variation for this sample set when compared to the original capillary gas chromatographic method. In addition, these results indicate that it may also be an effective method for the detection of medium invert sugar, depending on the level of the fingerprint oligosaccharides in this sweetener.  相似文献

Fluid within the tympanic bulla is an indication of middle ear disease. Radiography has a relatively low accuracy for the detection of soft tissue opacification in the tympanic bulla, and the most useful radiographic projection, the rostrocaudal open mouth (RCd (open mouth)), is technically difficult to perform in dogs and cats. An alternative projection for the feline tympanic bulla, the rostro 10 degrees ventro-caudodorsal oblique (R10 degrees V-CdDO), was compared to the RCd (open mouth) in 41 feline cadaver heads with the tympanic bullae randomly filled with KY jelly. Computed tomography was used as the gold standard. Each tympanic bulla was recorded as being positive or negative for soft tissue opacification. Although there was no significant difference between the accuracy of the two views, the R10 degrees V-CdDO was subjectively more accurate and easier to perform, and in a live patient may be performed without the need for general anesthesia. The R10 degrees V-CdDO projection is a good alternative to the RCd (open mouth) projection for detecting otitis media in the cat.  相似文献

The effects of frame size (FS) and body condition score (BCS) on performance of Brahman cows were evaluated using records collected from 1984 to 1994 at the Subtropical Agricultural Research Station, Brooksville, Florida. Age at puberty (AP), calving rate (CR), calving date (CD), survival rate (SR), weaning rate (WR), birth weight (BWT), weaning weight (WWT), preweaning ADG, and kilograms of calf produced per cow exposed (PPC) were obtained from first- (n = 215), second- (n = 130), and third or greater-parity (n = 267) dams. Based on hip height at 18 mo of age, heifers were assigned to three FS groups: small (115 to 126 cm), medium (127 to 133 cm), or large (134 to 145 cm). Small and medium FS heifers attained puberty at younger (P<.05) ages (633.2+/-12.3 and 626.4+/-12.0 d) than large FS heifers (672.3+/-17.1 d). Calving rate in large FS second-parity dams was 27% less (P<.05) than in small and medium FS dams. In third or greater-parity dams, CR was greater (P<.05) for small FS cows than for medium and large FS cows. Across the three parity groups, CR improved with increasing BCS. Except for the first-parity dams, animals with better fall BCS calved earlier (P<.05). In first-parity dams, SR was less (P<.01) in large (47.9+/-11.0%) than in small (80.7 +/-5.2%) and medium (83.4+/-4.7%) FS groups. Weaning rates of large FS first- and second-parity dams were less (P<.05) than those of small and medium FS dams. Second-parity dams with BCS 3 had lower (P<.05) WR than dams with BCS 4 and 5. Within first- and third or greater-parity dams, BWT of calves born to small FS cows were the lightest, and those born to large FS dams were the heaviest; those born to medium FS dams were intermediate (P<.05). In second-parity dams, BWT of calves of large FS dams were greater (P<.05) than those of small and medium FS dams. In first-parity dams, calves weaned by small FS cows had lower (P<.05) WWT than those weaned by higher FS cows. In the third or greater-parity group, large FS dams weaned heavier calves (P<.05) than other dams. In all parity groups of dams, calves out of large FS cows had greater ADG (P<.05) than those from small and medium FS cows. In first-parity dams, PPC was comparable between small and medium FS dams, but both tended to be greater (P<.10) than PPC of large FS dams. Small and medium FS females reached puberty at an earlier age, calved earlier, and had greater calving, survival, and weaning rates, as well as greater kilograms of calf produced per cow exposed than the large FS females. As the large FS cows matured, they seemed to have overcome the negative effects imposed by FS that were observed at younger ages. Their performance traits were generally all comparable to those of smaller cows once they had reached maturity.  相似文献
