The infection process ofRhizoctonia solani AG-3 was studied on potato sprouts, cv. Bintje, in growth chamber trials at 15 °C. Initially hyphae ofR. solani grew predominantly in the longitudinal direction of the sprouts (runner hyphae). They tended to follow the junctions between epidermis cells as was observed by SEM. The hyphae formed side-branches mainly half-way of the subterranean parts of the sprouts. They branched several times with short swollen cells to form infection cushions. Lesions developed only underneath the infection cushions and were first observed five days after inoculation. The necrotic area was proportional to the area covered with infection cushions on the sprouts. Depth of the lesions could extend up to the vascular bundle. Sprouts were colonized only in healthy tissue in the epidermal layer underneath the infection cushion and in necrotic tissue. A few days after appearance of the lesions,R. solani formed brown, uninfective mycelium on and in the circumference of these lesions.Aldicarb did not influence any part of the infection process. Ethoprophos delayed the emergence of sprouts, but increased the number of sprouts per tuber. As soon as sprouts had emerged, growth was considerably promoted by ethoprophos. Ethoprophos delayed the appearance of lesions and reduced their size. Oxamyl showed the same effects to a smaller extent.As the size of lesions appears to be proportional to the size of the infection cushions, any agents that change the size of the infection cushions, such as pesticides or antagonists, may alter the severity of the disease.Samenvatting Het infectieproces vanRhizoctonia solani AG-3 werd bestudeerd op aardappelspruiten, cv. Bintje, in een klimaatcel bij 15C. Aanvankelijk groeide de schimmel met runnerhyfen voornamelijk in de lengterichting van de spruit. Via SEM kon waargenomen worden, dat de hyfen hierbij vooral over de begrenzingen van de epidermiscellen groeiden. Het mycelium vormde veel zijvertakkingen, bestaande uit iets gezwollen korte cellen, welke voornamelijk halverwege op het ondergrondse deel van de spruit gevormd werden. Een dichte massa van deze cellen vormde een infectiekussentje. Lesies, welke vanaf vijf dagen na inoculatie werden waargenomen, bevonden zich slechts onder spruitoppervlak bezet met infectiekussentjes. De lesiegrootte was recht evenredig met het spruitoppervlak dat bezet was met infectiekussentjes. De diepte van de lesies reikte tot aan de vaatbundels. De spruit werd alleen door de schimmel gekoloniseerd in gezond epidermisweefsel onder het infectiekussentje en in necrotisch weefsel. Enkele dagen na verschijning van lesies vormde R.solani bruin, niet infectieus, mycelium op en rondom de lesies.Aldicarb had geen effect op het infectieproces. Ethoprophos vertraagde de opkomst en verhoogde het aantal tot ontwikkeling gekomen spruiten per knol in gestoomd zand. Direct na opkomst had ethoprophos echter een sterk groeistimulerend effect. Ethoprophos vertraagde de lesievorming en reduceerde de lesiegrootte, vergeleken met onbehandelde planten. Oxamyl vertoonde deze effecten in geringere mate.Daar de lesiegrootte direct gecorreleerd blijkt met de grootte van het infectiekussentje, mag verwacht worden dat elke beïnvloeding van de ontwikkeling van het mycelium van R.solani, bijvoorbeeld door pesticiden of antagonisten, een verandering van de lesiegrootte ten gevolge heeft.  相似文献

The aim of our study was to examine effects of the length of semen equilibration as well as two freezing techniques on ram sperm post-thaw quality. The ejaculates of Wallachian sheep rams (n = 12) were collected by an electro-ejaculation, equilibrated in a Triladyl® (0, 2, 4, 6, and 8 h) containing glycerol and egg yolk and frozen by programmable freezing (PF) or manual freezing (MF). After thawing, sperm samples were subjected to the motility (computer-assisted sperm analysis [CASA]), viability (SYBR-14/PI), and fertilizing ability (FA) (in vitro penetration/fertilization test on bovine oocytes) assays. It was found that the equilibration of 6 h (E-6) ensured higher post-thaw sperm motility and progressive movement compared with other lengths tested, irrespective of a freezing technique. The E-6 sperm viability did not differ between PF and MF but was lower (P < 0.05) than control. Sperm FA (E-6) was similar in PF (60.44%) and MF (62%) but slightly lower than in fresh (72.8%). Our data demonstrate that the use of MF was comparable with PF, which can be applied in the field conditions without need in a piece of cost-expensive equipment, which can greatly benefit the gene bank of animal genetic resources.  相似文献

On the basis of 268 direct sika deer observations and 212 ratio locations of three individuals, the influence of snow on deer distribution was studied. During the period of snow absence or its shallow cover, the deer did not show any preferences towards slope aspect. However, when the snow coverage became deeper, the deer used westerly exposures (having shallower snow cover) more intensively. Later on, when the snow started to melt on the easterly slope aspects deer mostly used those exposures due to relatively abundant food resources.  相似文献

Dulling of high-speed steel (HSS) cutting tools after milling five wood species has been analyzed. The wood investigated varied in mineral contamination, high-temperature corrosivity (HTC) and density (D). Experiments performed proved that up to date methods for evaluation of mineral contamination in wood should be improved. A new pipe-like form of mineral contamination particles was evidenced. The correlation between the presence of 3D aggregates of silica in wood and high content of K and Ca in the ash was established. Employing multivariable theoretical simulation method, the combined effect of mineral contamination, HTC and wood density on the tool dullness appeared to be a reasonable explanation of the results obtained.  相似文献

This paper describes a sensitive isocratic HPLC/ECD method developed for the determination of rosmarinic acid (RA) in plant material, animal feed, and pig plasma. The plasma sample preparation only includes protein precipitation and adjustment of the pH. The applicability of the method was tested on plasma samples of pigs that were exposed to a 91-day oral intake of RA via feed enriched by aerial parts of Prunella vulgaris. The plasma was directly analyzed using the method described as well as after enzymatic hydrolysis. When no hydrolysis step was included, RA and caffeic acid (CA) were quantified in the plasma. In hydrolyzed plasma samples, several other metabolites were determined, including dihydrocaffeic, ferulic, and dihydroferulic acid. The dual-channel coulometric detection employed, as an alternative to mass spectrometry, offers good selectivity and sensitivity owing to the electrochemical properties of the phenolic constituents.  相似文献

Soil variability was assessed in a 74.2-ha area within the Žofínsky prales natural forest. Parameters evaluated for 1765 soil profiles inside 353 graticule plots were as follows: (1) thickness of organic horizons, (2) thickness and form of mineral horizons, (3) humus form (HF), (4) soil taxonomic unit (STU) and (5) anomalies. In addition, soil reaction (pHKCl) and oxidizable carbon content (Cox) were measured in the laboratory for 734 samples from the upper mineral (A) and lower mineral (B) horizons. The most frequently occurring humus form was mor followed by moder, hydromor and peaty T-horizon. Entic Podzols, Dystric Cambisols, Haplic Cambisols, Albic Podzols, Histic (or Haplic) Gleysols, Endogleyic Stagnosols, Fibric or Hemic or Sapric Histosols and Stagnic Gleysols were all present at the site despite its homogeneous geological bedrock. Overall coefficient of variance (CV) was lower in terrestrial soils compared with (semi-)hydromorphic soils. Overall variance decreased in both soil groups with increasing depth, as did CV differences between the fine (up to 10 m) and the locality scales. The lowest CV values occurred for Cox and pHKCl. The CV values differed between STUs as well. Compared to lower horizons, variograms of upper horizons showed greater autocorrelation at the intermediate spatial scale (10–320 m)—ranging from 50 to 150 m. Semivariance values, however, reached 70–80% of sill already at a distance of 10 m. The most significant factor of variability at all studied spatial scales is presumably the soil disturbance regime, followed by terrain micro-topography and the effect of tree species.  相似文献

Root growth of winter barley was studied on 2 Belgian silt loam soils after subsoiling to 60 cm depth. In the first field (well-drained) rooting depth increased significantly after removal of a plough pan. At the flowering stage, total root weight in the soil layers between 25–50 cm and 50–75 cm was 125% and 325%, respectively, higher on the subsoiled plots than on the control. The P2O5 and K2O concentrations in the roots were significantly increased by 20% and 40%, respectively. In the second field (imperfectly drained) with initially relatively high bulk densities in the subsoil, the influence of subsoiling was negligible. In this study, the clearly-shown root-growth retardation that resulted in a lower yield on the control, seemed to be caused by the high penetration resistance in the plough sole (> 3MPA) rather than poor aeration status induced by the high bulk densities.  相似文献

The aim of this laboratory study was to investigate the effect of straw and vinasses on the nitrogen (N) mineralization-immobilization turnover of celery residues during two periods (each simulating a time period from autumn till spring) under laboratory conditions. During the first period (1-198 d), 15N-labelled celery residues (1.1 g dry matter (DM) kg−1 soil) were incubated together with straw (8.1 g DM kg−1 soil), aiming to immobilize the N released from celery residues, followed by an incorporation of vinasses (1.9 g DM kg−1 soil) after 84 d, with a view to remineralizing the immobilized celery-N. During the second period (198-380 d), the experimental set-up was repeated, except that non-labelled celery residues were used. Total N, mineral N and their 15N enrichments as well as microbial biomass N were determined at regular time intervals. During both periods, mixing celery residues with straw significantly increased microbial biomass N (90.5 and 40.5 mg N kg−1 extra compared to celery only treatment) and decreased the amount of mineral N (reduction of 56.1 and 45.9 mg N kg−1 soil compared to celery only treatment) and the celery-derived mineral 15N (0% of mineral celery-derived 15N in straw treatment compared to 35% of mineral celery-derived 15N in celery only treatment). After maximum immobilization, a natural remineralization (without addition of vinasses) of 32.2 (at day 198) and 11.1 mg N kg−1 soil (at day 380) occurred in the straw treatment, but the mineral N content remained significantly lower than in the celery only treatment during the complete experiment, and the amount of remineralized celery-15N was very low (5.4% of celery-derived 15N after 380 d). Vinasses caused no real priming effect, although it did slightly increase the amount of remineralized celery-15N (+6.4% of celery-derived 15N at day 380 compared to the straw treatment), probably due an apparent added N interaction caused by displacement reactions with the soil microbial biomass.  相似文献
