OBJECTIVE: To determine the effects of porcine mammary secretions on polymorphonuclear (PMN) leukocyte function and to relate concentrations of estradiol-17beta and cortisol in mammary secretions to PMN cell function. SAMPLE POPULATION: Mammary secretions from 10 healthy sows and blood PMN leukocytes from 27 healthy sows. PROCEDURE: Mammary secretions were collected within 24 hours after parturition (colostrum) and 12 to 13 days later (milk). Chemoattractant properties were assessed by use of a cell migration assay. Phagocytic capacity of PMN cells in colostrum and milk was assessed by recording chemiluminescence following phagocytosis of Escherichia coli or zymosan. Estradiol-17beta and cortisol concentrations were determined by use of radioimmunoassays. RESULTS: Chemoattractant properties of colostrum and milk were significantly greater than that of zymosan-activated serum. However, chemoattractant properties did not differ significantly between the 2 types of secretions. The capacity of PMN cells in colostrum to phagocytose either zymosan or E. coli was less, compared with cells in milk, and the ability of cells in either type of mammary secretion to phagocytose E. coli was greater than the ability to phagocytose zymosan. Concentrations of estradiol-17beta and cortisol were greater in colostrum, compared with milk. No clear relation was evident between PMN cell activity and hormone concentrations in mammary secretions. CONCLUSIONS AND CLINICAL RELEVANCE: Although chemoattractant properties of colostrum and milk did not differ, the phagocytic capacity of PMN cells in colostrum was significantly less than that of cells in milk. This may predispose sows to coliform mastitis during the early postparturient period.  相似文献

Fetal protein restriction is potentially associated with organ dysfunctions after birth (e.g. impaired gut growth, glucose tolerance and pancreatic β-cell function). Just after birth, gut growth and maturation is stimulated by enteral food intake, and inhibited by total parenteral nutrition (TPN), in part mediated via differential release of insulino- and intestino-tropic hormones like the Glucagon-Like Peptides 1 and 2 (GLP-1, GLP-2). We hypothesized that short-term co-infusion of GLP-1 and GLP-2 would stimulate pancreatic and intestinal growth in newborn TPN-fed pigs subjected to prenatal protein restriction. Two sows were fed a protein-restricted diet (PR: 8% crude protein during last 50% of gestation) while a third sow was fed a control diet (C: 15% crude protein). PR pigs were killed either at birth (n = 7) or after 3 days TPN with (n = 6) or without (n = 4) intravenous infusion of a mixture of synthetic human GLP-17–37 and GLP-21–33 (each 50 μg/kg/d). At birth, PR piglets did not show reduced body weight, relative to controls (1.45 vs. 1.50 kg), but significantly reduced weight of the small intestine (18.0 ± 0.6 vs. 21.9 ± 0.5 g/kg, P < 0.001) and a marginally reduced pancreas weight (0.85 ± 0.02 vs. 0.93 ± 0.04 g/kg, P = 0.10). Co-infusion GLP-1 and GLP-2 into PR pigs resulted in increased basal glucose levels (5.3 vs. 4.0 mM), and glucose-stimulated insulin release, but did not have any significant effect on body weight, or weight of internal organs (heart, lungs, spleen, kidneys, adrenals, stomach, colon, liver, intestine, pancreas). We conclude that short-term (3 days) infusion of native GLP-1 and GLP-2 does not stimulate gut growth or glucose tolerance in TPN-fed piglets born from protein-restricted mothers. Moderate maternal protein restriction does however cause significant reduction in intestinal growth in newborn piglets which may decrease the neonatal digestive capacity.  相似文献

The commercialization of animal feeds infected by prions proved to be the main cause of transmission of bovine spongiform encephalopathy (BSE). Therefore, feed bans were enforced, initially for ruminant feeds, and later for all feeds for farmed animals. The development and validation of analytical methods for the species-specific detection of animal proteins in animal feed has been indicated in the TSE (Transmissible Spongiform Encephalopathies) Roadmap (European Commission. Methods based on polymerase chain reaction (PCR) seem to be a promising solution for this aim. The main objective of this study was to determine the applicability of four different real-time PCR methods, developed by three National expert laboratories from the European Union (EU), for the detection and identification of cattle or ruminant species in typical compound feeds, fortified with meat and bone meals (MBM) from different animal species at different concentration levels. The MBM samples utilized in this study have been treated using the sterilization condition mandatory within the European Union (steam pressure sterilization at 133 degrees C, 3 bar, and 20 min), which is an additional challenge to the PCR methods evaluated in this study. The results indicate that the three labs applying their PCR methods were able to detect 0.1% of cattle MBM, either alone or in mixtures with different materials such as fishmeal, which demonstrates the improvement made by this technique, especially when compared with results from former interlaboratory studies.  相似文献

Abstract. Wet and droughty soils and those on steep slopes are now marginal for agriculture in Denmark. A nation-wide map of these soil types has therefore been made to show their distributions. OBJECTIVE: To examine to what extent the obesity epidemic is a general phenomenon in adults by assessing the secular change, by birth cohort and age, in the prevalence of obesity and median body mass index (BMI) in Danish men and women measured between 1964 and 1994. DESIGN: Multiple cross-sectional population surveys. SETTING: The greater Copenhagen area of Denmark. SUBJECTS: The study included 17,065 men (30 336 observations) and 13,417 women (24,065 observations), aged 20-84 years. MAIN OUTCOME MEASURES: Trends in median BMI and prevalence of obesity estimated from measured height and weight in 10-year age groups. RESULTS: In general the prevalence of obesity was increasing, although in an irregular way: among men in two phases, during the 1970s and 1990s and among women only during the 1990s. Great heterogeneity was observed between birth cohorts and age groups. There was only little indication of an increasing trend in obesity prevalence for women, except for the 55-64-year-olds. In men, the prevalence of obesity was increasing in all age groups except in the youngest ones, and it was statistically significant only for men aged 35-74 years. There was no significant linear change in median BMI in any group, except for an increasing trend among men aged 50 years and above. CONCLUSION: Although the overall Danish trend for obesity prevalence, similar to trends world-wide, showed a marked increase, the trend was very heterogeneous and generally neither uniform nor significantly positive; the changes were irregular, different among men and women, and different in the different age and birth cohorts. The obesity problem in middle-aged and older men of certain birth cohorts poses a specific public health challenge. Future studies of determinants behind the heterogeneity in the development of the obesity epidemic may provide clues to its causes.  相似文献

The carbon (δ(13)C) and oxygen (δ(18)O) stable isotope composition is widely used to obtain information on the linkages between environmental drivers and tree physiology over various time scales. The tree-ring archive can especially be exploited to reconstruct inter- and intra-annual variation of both climate and physiology. There is, however, a lack of information on the processes potentially affecting δ(13)C and δ(18)O on their way from assimilation in the leaf to the tree ring. As a consequence, the aim of this study was to trace the isotope signals in European beech (Fagus sylvatica L.) from leaf water (δ(18)O) and leaf assimilates (δ(13)C and δ(18)O) to tree-ring wood via phloem-transported compounds over a whole growing season. Phloem and leaf samples for δ(13)C and δ(18)O analyses as well as soil water, xylem water, leaf water and atmospheric water vapour samples for δ(18)O analysis were taken approximately every 2 weeks during the growing season of 2007. The δ(13)C and δ(18)O samples from the tree rings were dated intra-annually by monitoring the tree growth with dendrometers. δ(18)O in the phloem organic matter and tree-ring whole wood was not positively related to leaf water evaporative enrichment and δ(18)O of canopy organic matter pools. This finding implies a partial uncoupling of the tree-ring oxygen isotopic signal from canopy physiology. At the same time, internal carbon storage and remobilization physiology most likely prevented δ(13)C in tree-ring whole wood from being closely related to intra-annual variation in environmental drivers. Taking into account the post-photosynthetic isotope fractionation processes resulting in alterations of δ(13)C and δ(18)O not only in the tree ring but also in phloem carbohydrates, as well as the intra-annual timing of changes in the tree internal physiology, might help to better understand the meaning of the tree-ring isotope signal not only intra- but also inter-annually.  相似文献

Berry shrivel and bunch stem necrosis are the economically most important physiological disorders of grapevine. The nutritional status of the plant is considered as one of the main elicitors for the occurrence of these disorders. For foliar fertilization trials, two vineyards that had previously been affected by both diseases were selected in Southwest Germany, one with the variety ‘Zweigelt’ and one with Pinot Blanc. By the use of potassium, magnesium, and calcium foliar fertilizers, a rapid increase of different nutrients in the plants should be caused and the effects on the diseases observed. In 2010, no significant differences in the fertilization treatments were found for both varieties. In ‘Pinot Blanc’, significantly more berries were affected by berry shrivel in 2011 in plots treated with calcium fertilizer compared to those treated with potassium and magnesium. Plots with magnesium fertilizer showed significantly less berries affected by berry shrivel than the untreated control.  相似文献
