This study investigated the overwintering survival and infectivity of free-living gastrointestinal nematode (GIN) stages on pasture. The presence of GIN larvae was assessed on 3 sheep farms in Ontario with a reported history of clinical haemonchosis, by collecting monthly pasture samples over the winter months of 2009/2010. The infectivity of GIN larvae on spring pastures was evaluated using 16 tracer lambs. Air and soil temperature and moisture were recorded hourly. Free-living stages of Trichostrongylus spp. and Nematodirus spp. were isolated from herbage samples. Gastrointestinal nematodes were recovered from all tracer lambs on all farms; Teladorsagia sp. was the predominant species. Very low levels of Haemonchus contortus were recovered from 1 animal on 1 farm. The results suggest that Haemonchus larvae do not survive well on pasture, while Teladorsagia sp., Trichostrongylus spp. and Nematodirus spp. are able to overwinter on pasture in Ontario and are still infective for sheep in the spring.  相似文献

In vitro study with chicken bursal organ culture was attempted to assess the pathogenicity of locally isolated infectious bursal disease virus (IBDV) initially isolated from the bursa of naturally infected birds. In bursal organ culture, lymphoblastic transformation was noticed as early as 24 hr postinoculation and reached maximum at 72 hr postinoculation. The other microscopic changes were increased number of macrophages and formation of plasma cells. The IBDV antigen was detected 24 hr onward by coagglutination test with antibody coated Staphylococcus aureus strain Cowan I. On the basis of lesion score, the three isolates of IBDV (A, B, and C) were graded as virulent (B isolate) and moderately virulent (A and C isolates). A similar pattern of pathogenicity was also observed in the in vivo pathogenicity studies in chicken based on bursa: body weight ratio and histopathologic lesion score. The bursal organ culture thus provides a useful experimental model to differentiate the IBDV isolates on the basis of their virulence.  相似文献

Sustaining soil fertility is essential to the prosperity of many households in the mid-hills of Nepal, but there are concerns that the breakdown of the traditional linkages between forest, livestock, and cropping systems is adversely affecting fertility. This study used triangulated data from surveys of households, discussion groups, and key informants in 16 wards in eastern and western Nepal to determine the existing practices for soil fertility management, the extent of such practices, and the perception of the direction of changes in soil fertility. The two principal practices for maintaining soil fertility were the application of farmyard manure (FYM) and of chemical fertilizer (mainly urea and diammonium phosphate). Green manuring, in-situ manuring, slicing terrace risers, and burning plant residues are rarely practiced. FYM usage was variable with more generally applied to khet land (average 6053 kg fresh weight manure ha–1) than to bari land (average 4185 kg fresh weight manure ha–1) with manure from goats and poultry preferred above that from cows and buffaloes. Almost all households (98%) apply urea to khet land and 87% to bari land, with 45% applying diammonium phosphate to both types of land. Application rates and timings of applications varied considerably both within and between wards suggesting poor knowledge transfer between the research and farming communities. The benefits of chemical fertilizers in terms of ease of application and transportation in comparison with FYM, were perceived to outweigh the widely reported detrimental hardening of soil associated with their continued usage. Among key informants, FYM applied in conjunction with chemical fertilizer was the most popular amendment, with FYM alone preferred more than chemical fertilizer alone – probably because of the latters long-term detrimental effects. Key informant and householder surveys differed in their perception of fertility changes in the last decade probably because of differences in age and site-specific knowledge. All key informants felt that fertility had declined but among households, only about 40% perceived a decline with the remainder about evenly divided between no change and an increase. Householders with small landholdings (< 0.5 ha) were more likely to perceive increasing soil fertility while those with larger landholdings (> 2 ha) were more likely to perceive declining fertility. Perceived changes in soil fertility were not related to food self-sufficiency. The reasons for the slow spread of new technologies within wards and the poor understanding of optimal use of chemical fertilizers in conjunction with improved quality FYM may repay further investigation in terms of sustaining soil fertility in this region.Colin Pilbeam graduated from the University of Oxford with an MA in Agriculture and Forest Sciences. He has worked in Australia, Syria, Nepal, India, and West Africa and is the chair of Global Environmental Change and Food Systems – an international research project on food security.Padma Bahadur Shakya is an Agricultural Economist who has worked for the Department of Agriculture under the Ministry of Agriculture and Cooperatives in Nepal for more than 20 years. He has also been a short-term consultant for various national and international organizations such as FAO, the UNs World Food Programme, Swiss Development Corporation, Asian Development Bank, JICA, HARP, and several local NGOs. To provide good quality of drinking water, a biological system to remove ammonium-nitrogen (NH4-N) from groundwater was studied in this research. The NH4-N removal system consists of two attached growth reactors: one for nitrification and the other for hydrogenotrophic denitrification (H. denitrification). The nitrification reactor, fed by the NH4-N contained water, could remove NH4-N without any need of aeration. The nitrification efficiency was increased by reactor length; the highest efficiency of 92?% was achieved at the longest reactor of 100?cm. A high Fe in groundwater affected the reactor performance by decreasing the efficiency, while a low inorganic carbon (IC) had no effects. Despite of good efficiency in terms of NH4-N removal, the nitrification reactor increased the concentration of NO3-N in its effluent. To treat the NO3-N, a H. denitrification reactor was set up after the nitrification reactor. Efficiency of the H. denitrification reactor was enhanced by increasing H2 flow rates. The efficiencies were 3, 27, and 90?% for 30, 50, and 70?mL/min of H2 flow rates, respectively. It was also found that the NO3-N contained water (water from the nitrification reactor) had to supply IC (i.e., NaHCO3 or CO2) for efficient H. denitrification; however, an on-site reactor showed that it can be achieved even without IC addition. The treated water contained low NH4-N and NO3-N of <1.5 and <11.3?mg/L, respectively, which comply with drinking water standards. The good performance of the reactors in terms of high efficiency, no aeration need, and low H2 supply indicated appropriateness of the system for groundwater treatment.  相似文献

This community forestry case study examines village forest councils (van panchayat) in India’s northern state of Uttarakhand. We describe forest stocking levels of 20 community forests and address the question of whether these forests are in a position to provide commodity resources, such as fuelwood and fodder, on a sustainable basis. In addition, we report on the perceptions of the villagers on whether their respective community forest is meeting their fuelwood and fodder needs. At the biophysical level, none of the community forests were in a position to provide fuelwood and fodder on a sustainable basis. Most forests did not have adequate overstory stocking, and there was no evidence of deliberate management to improve the situation. Interviews with 400 villagers (20 in each village) supported the field findings. Villagers unanimously reported that their respective community forest was inadequate in supplying commodity needs. Furthermore, they wished the Forest Department would provide more technical and financial assistance to guide them with forest management needs. Women are a critical part of community forestry as they are the primary gatherers of fuelwood and fodder. Although the perceptions of women toward forest management issues are important, our interviews with female villagers indicate they play a passive role in decision-making. Decentralization in natural resources management sounds good at the theoretical level as it provides villagers with some authority and autonomy, but it cannot be effectively implemented without professional guidance coupled with resources. The villagers are ready and willing to improve the situation, but they do not have the resources to make change unilaterally—they need professional and financial support from the Forest Department.  相似文献

Secondary metabolites in potato tubers include both phytonutrients and plant defense compounds. The extent these small molecules vary among different potato genotypes is not well characterized. LC-MS analysis of tuber extracts from seven potato genotypes showed that one large source of small molecule variation is the glycoalkaloids. Glycoalkaloids are involved in the resistance of potatoes to pathogens and pests, but they also have implications for human health and nutrition. This study focused on glycoalkaloids with solanidane or solanidane-like aglycones, of which over 50 were tentatively identified, many of which appeared to be novel glycoalkaloids. Results suggested the variety of glycoalkaloids in potatoes is considerably greater than previously thought. Dissecting the role of these many glycoalkaloids in human health or pest and pathogen resistance will be a formidable undertaking.  相似文献
