In the United States, yellow starthistle (Centaurea solstitialis) is an annual invasive weed with Mediterranean origins. Malformed plants displaying witches' broom, fasciations, abortion of buds and flower virescence symptoms were observed in central Italy. Attempts to transmit the causal agent from the natural yellow starthistle host to periwinkle by grafting, resulted in typical symptoms of a phytoplasma, i.e. yellowing and shortening of internodes. The detection of phytoplasmas was obtained from both symptomatic yellow starthistle and periwinkle by the specific amplification of their 16S-23S rRNA genes. PCR amplification of extracted DNA from symptomatic plant samples gave a product of expected size. Asymptomatic plants did not give positive results. An amplicon obtained by direct PCR with universal primers P1/P7 was cloned and sequenced. The homology search using CLUSTALW program showed more than 99% similarity with Illinois elm yellows (ILEY) phytoplasma from Illinois (United States) and 97% with Brinjal little leaf (BLL) phytoplasma from India. Digestion of the nested-PCR products with restriction enzymes led to restriction fragment length polymorphism patterns referable to those described for phytoplasmas belonging to the clover proliferation (16S-VI) group. Since this is a previously undescribed disease, the name Centaurea solstitialis virescence has been tentatively assigned to it. This is a new phytoplasma with closest relationships to ILEY and BLL, but distinguishable from them on the basis of 16S rDNA homology, the different associated plant hosts and their geographical origin.  相似文献

A study was conducted to evaluate a non-destructive method able to predict Magness–Taylor (MT) penetrometric firmness of blood oranges, potentially applicable for on-line selection. The method consisted of multivariate modelling of non-destructive penetrometric measurements performed on intact oranges with a 11.3 mm diameter probe by digital dynamometric equipment. The tests were carried out on 366 blood oranges (Citrus sinensis (L.) Osbeck cv. Tarocco); 60 were used as a control group to verify the non-destructive nature of the tests. Four test factors were used on the fruit: two different levels of non-destructive deformation (1 and 3 mm), at two different deformation speeds (5 and 30 mm s?1). In order to obtain the reference parameter (MT firmness), a destructive penetrometric test followed each non-destructive deformation on the same fruit (deformation of 20 mm at 1 mm s?1). The force–deformation curves of the non-destructive tests were elaborated to calculate different texture parameters (max force, elasticity, work). These parameters with raw curve data (force values for constant deformation steps) were used, after autoscaling pre-processing, as X-block datasets in supervised multivariate modelling (partial least squares) to predict MT firmness values. Among the test factors used, the 30 mm s?1 speed and 3 mm deformation provided the best overall prediction of MT firmness. This comprised an r value of 0.76 and standard error of prevision (SEP) of 2.99 N for a MT firmness mean value of 35.65 N (8.4%). The time needed to apply the selected deformation may lead to a sorting speed of about 10 fruit per second, representing an acceptable value for commercial application.  相似文献

Numerous plants ofSilene nicaeensis having symptoms resembling those associated with the presence of phytoplasmas were observed in an extensive coastal area in the south of Italy. Microscopic observation showed histological abnormalities in the organization of tissues in symptomatic plants, and molecular tests, including PCR/RFLP analyses and nucleic acid sequencing, revealed the presence of phytoplasmas belonging to the aster yellows group (‘Candidatus phytoplasma asteris’). The squid pen (gladius) from the Loligo vulgaris was used for preparation of β-chitin materials characterized by different chemical, micro- and nano-structural properties that preserved, almost completely the macrostructural and the mechanical ones. The β-chitin materials obtained by alkaline treatment showed porosity, wettability and swelling that are a function of the duration of the treatment. Microscopic, spectroscopic and synchrotron X-ray diffraction techniques showed that the chemical environment of the N-acetyl groups of the β-chitin chains changes after the thermal alkaline treatment. As a consequence, the crystalline packing of the β-chitin is modified, due to the intercalation of water molecules between β-chitin sheets. Potential applications of these β-chitin materials range from the nanotechnology to the regenerative medicine. The use of gladii, which are waste products of the fishing industry, has also important environmental implications.  相似文献

The response of potted pear trees grafted on quince (Cydonia oblonga Mill.) Sydo stock to increasing concentrations (0, 50, 100, 200, 400, 600, 800, and 1000 mg kg?1) of copper (Cu) in sandy and clay-loam soils were evaluated and crop toxicity thresholds and symptoms were determined. Reductions of shoot growth and leaf area were observed only for pear grown in sandy soil with more than 400 mg Cu/kg. During the growing season, carbon assimilation and soil microbial biomass were reduced as concentration of soil Cu increased. However, the effect was always stronger in sandy than clay-loam soil. Copper accumulated in roots, with response to soil Cu additions described by a linear and a quadratic function for sandy and clay-loam soil, respectively. A possible antagonism was observed between Cu and zinc (Zn) in the roots. Root Zn concentration decreased as soil Cu concentration increased.  相似文献

A novel application of a hybrid biosensor is here employed as an analytical method for the detection and presumptive identification of beta-lactam residues in milk. The method is based on measurements of carbon dioxide (CO2), the production of which is related to the microbial growth of the test microorganism Bacillus stearothermophilus var. calidolactis. The presence of beta-lactams in milk inhibits microbial growth and, consequently, the CO2 production rate. The analysis is based on the variation of CO2 between a milk sample spiked with beta-lactams and a twin milk sample containing beta-lactams plus a broad spectrum beta-lactamase, using an electrochemical device of biosensor. A blank milk sample is included as control. The result is obtained starting from the first 120 min. Moreover, the ability to recognize all of the beta-lactams speeds the total time of analysis when chemical identification and quantification are required. The analytical method appears to be adequate for milk control for qualitative screening purposes, complying with the requirements stated in Decision 2002/657/EC.  相似文献

The toxicity of organophosphorus pesticides is attributed to their ability to inhibit acetylcholinesterase (AChE), which catalyses the hydrolysis of the neurotransmitting agent acetylcholine (ACh). A simple, reproducible and stable AChE-based amperometric bioelectrode was used, for measuring the enzyme activity before and after the incubation with paraoxon dissolved in organic solvents.The inhibition percentage induced by a paraoxon in organic solvent solutions increases in the following sequence: acetonitrile < water < hexane, suggesting that the paraoxon repartition between the organic solvent and the essential water for enzyme activity plays an important role in establishing the analytical and kinetic parameters of the bioelectrode. Considering the AChE denaturation, induced only by the organic solvent, as a pseudo-inhibition process, the kinetic treatment of the bioelectrode response revealed that it follows a mixed competitive/non-competitive mechanism, while the inhibition due to a non-aqueous solution of paraoxon corresponds to a non-competitive/uncompetitive mechanism. In the last case, the synergetic effect of denaturation and inhibition of enzyme was dominated by the inhibitor presence.  相似文献

Epidemiological studies have demonstrated that many antioxidants and the total antioxidant capacity (TAC) of the diet may protect against cancers and cardiovascular disease. Common fruits and vegetables are good sources of antioxidants, although in some Mediterranean areas traditional wild greens are responsible for a significant percentage of total dietary antioxidant intake. In the European Prospective Investigation into Cancer and Nutrition cohort of Ragusa (Sicily), a high number of subjects were found to frequently eat wild greens, including Sinapis incana and Sinapis nigra, Diplotaxis erucoides, Cichorium intybus, Asparagus acutifolius, and Borrago officinalis. On the basis of these observations, detailed characterization of single antioxidant components (i.e., polyphenols, carotenoids, chlorophylls, and ascorbic acid) and the TAC of these edible wild traditional plants was performed. The wild plants examined were found to be very rich in antioxidants, such as flavonoids and carotenoids, with high TAC values, suggesting that the importance of these vegetables, not only in the traditional but even in the contemporary diet, needs to be emphasized.  相似文献

In this study, 120-l annular columns were used to cultivate Tetraselmis suecica outdoors. The mass transfer at different aeration rates and the influence of the harvest rate on productivity and biochemical composition were investigated. The potential of the system was evaluated by estimating productivity at full-scale. Two different arrangements to simulate a full-scale plant and determine the “overall areal productivity” (OAP) were experimented with. In August 2003, one experimental column (full-scale column) was placed between seven dummy columns. All the reactors were positioned at a distance of 0.8 m wall to wall and centred at the vertices of equilateral triangles. A second experimental column (isolated column) was placed in a separate area under full sunlight. In August 2004, the columns were placed side by side in an east-west oriented row at a distance of 0.24 m wall to wall.In the first experiment, the mean volumetric productivity of the full-scale column was not significantly lower than that achieved by the isolated column (0.46 against 0.49 g l− 1 day− 1) in spite of the shading by the dummy units. The average OAP and efficiency of conversion of visible solar radiation (PE) were 36.3 g m− 2 day− 1 and 9.4%, respectively. In the second experiment, the full-scale column attained a mean volumetric productivity of 0.42 g l− 1 day− 1. The OAP and the PE were 38.2 g m− 2 day− 1 and 9.3%, respectively.  相似文献
