Tricyclazole (5-methyl-1,2,4-triazolo[3,4-b]benzothiazole) inhibits melanin synthesis in Pyricularia oryzae at concentrations less than 0.01 μg/ml. The primary site of inhibition in the biosynthetic pathway occurs between scytalone and vermelone. Accumulation of several metabolites derived from melanin precursors along branch pathways is associated with inhibition of melanin biosynthesis. At low tricyclazole concentrations (0.01–1 μg/ml), predominant accumulation of 2-hydroxyjuglone and 3,4-dihydro-3,4,8-trihydroxy-1-(2H)-naphthalenone (3,4,8-DTN) occurs as a result of the primary block between 1,3,8-trihydroxynaphthalene and vermelone. As the concentration of tricyclazole is increased from 1 to 10 μg/ml, flaviolin accumulation is markedly enhanced, whereas that of 3,4,8-DTN and 3,4-dihydro-4,8-dihydroxy-1-(2H)-naphthalenone is depressed, indicating possible secondary sites of inhibition in the main and branch pathways. Five melanin-deficient mutants of P. oryzae that phenotypically resemble the tricyclazole-treated wild-type strain were nonpathogenic or rarely infected two rice varieties. Three of the mutants studied were genetically defective in the melanin biosynthetic pathway at the site blocked by tricyclazole in the wild type. The wild-type strain converted both scytalone and vermelone to melanin; whereas the three mutants and the tricyclazole-treated wild type converted only vermelone to melanin. The data suggest a relationship between melanin biosynthesis and pathogenicity in P. oryzae. 相似文献
Using a cross-sectional survey, we determined the prevalence and risk factors associated with bovine brucellosis in herds under extensive production system in southwestern Nigeria. Antibodies to Brucella species in serum samples were tested using the Rose Bengal test (RBT) and competitive enzyme-linked immunosorbent assay (cELISA); for milk, the milk ring test (MRT) and indirect-ELISA (i-ELISA) were used. Questionnaire was administered to cattle herdsmen to determine factors predisposing the animals to bovine brucellosis. Data were analyzed using STATA 12. From 513 serum and 635 milk samples tested among 120 herds, overall animal-level prevalence of 10.1% (95% CI 7.5–12.7%) and 20.2% (95% CI 17.1–23.3%) were recorded by RBT and MRT, respectively; while 9.4% (95% CI 6.9–11.9%) and 17.8% (95% CI 14.8–20.8%) were obtained using cELISA and i-ELISA, respectively. In all, from the 120 herds tested, 29.2% and 43.3% were positive by RBT and MRT, respectively. Multivariable logistic regression revealed that herd location (OR?=?8.12, 95% CI 1.68–38.90) and improper disposal of placenta/fetus (OR?=?17.33, 95% CI 4.81–62.33) were predictors for a seropositive herd using RBT; while herd location (OR?=?5.13, 95% CI 1.27–20.28), large herd size (OR?=?2.62, 95% CI 1.15–5.85), and occurrence of abortion for a year or more (OR?=?4.62, 95% CI 1.53–13.71) were predictors of seropositivity to antibodies to Brucella spp. using MRT. We found high prevalence of brucellosis in cattle herds under extensive management system in southwestern Nigeria. Urgent and coordinated control strategies are required to mitigate this problem.
Myxoma virus (MV) is a poxvirus that evolved in Sylvilagus lagomorphs, and is the causative agent of myxomatosis in European rabbits (Oryctolagus cuniculus). This virus is not a natural pathogen of O. cuniculus, yet is able to subvert the host rabbit immune system defenses and cause a highly lethal systemic infection. The interaction of MV proteins and the rabbit immune system has been an ideal model to help elucidate host/poxvirus interactions, and has led to a greater understanding of how other poxvirus pathogens are able to cause disease in their respective hosts. This review will examine how MV causes myxomatosis, by examining a selection of the identified immunomodulatory proteins that this virus expresses to subvert the immune and inflammatory pathways of infected rabbit hosts. 相似文献
Switchgrass (Panicum virgatum L.) is a promising bioenergy crop for temperate regions. Overwintering has been used to improve biomass quality, resulting in a more efficient combustion, partially due to a reduction in minerals concentration. This study examines the effects of soil composition on overwintered switchgrass composition. Samples were collected in the spring from 58 environments in Southern Quebec to determine possible relationships between soil composition and biomass quality. Principal component analysis and stepwise regressions were used to identify relationships between soil and biomass compositions. Soil parameters monitored explained 74% of the variation in biomass silicon (Si) concentration, 45% of the variation in ash, and 32% of the variation in magnesium (Mg). Soil composition had limited effects on the concentration of other elements in switchgrass biomass. Switchgrass biomass quality is influenced by soil composition and appears well suited to biomass combustion when overwintered and harvested in the spring. 相似文献
The ability to detect ecological networks in landscapes is of utmost importance for managing biodiversity and planning corridors.
Objectives
The objective of this study was to evaluate the information provided by a synthetic aperture radar (SAR) image for landscape connectivity modeling compared to aerial photographs (APs).
Methods
We present a novel method that integrates habitat suitability derived from remote sensing imagery into a connectivity model to explain species abundance. More precisely, we compared how two resistance maps constructed using landscape and/or local metrics derived from AP or SAR imagery yield different connectivity values (based on graph theory), considering hedgerow networks and forest carabid beetle species as a model.
Results
We found that resistance maps using landscape and local metrics derived from SAR imagery improve landscape connectivity measures. The SAR model is the most informative, explaining 58% of the variance in forest carabid beetle abundance. This model calculates resistance values associated with homogeneous patches within hedgerows according to their suitability (canopy cover density and landscape grain) for the model species.
Conclusions
Our approach combines two important methods in landscape ecology: the construction of resistance maps and the use of buffers around sampling points to determine the importance of landscape factors. This study was carried out through an interdisciplinary approach involving remote sensing scientists and landscape ecologists. This study is a step forward in developing landscape metrics from satellites to monitor biodiversity.
Biological activity of fenitrothion on stored maize at various moisture contents and at different times after application was measured by biological assay using adults of Tribolium castaneum (Herbst). Inactivation of actual residues over time was then determined after making the necessary allowance for chemical breakdown. At a given moisture content, the inactivation process was substantially completed during the first 6 weeks after application and loss of effectiveness from 6 weeks onwards resulted mainly from chemical breakdown. At a given time after application, residues were less active at higher moisture content (m.c). Differences in activity between moisture contents were apparent within a few hours of application and continued to increase for up to 3 days, with relatively little change thereafter during storage of 24 weeks. Thus after 24 weeks, residues on maize of 18% m.c. had an activity about 20% that of similarly-aged residues at 10% m.c. and 4% that of freshly-applied residues at 10% m.c. These results were in general accord with changes in the proportion of the residue which was collected from the kernels by a surface wash with methanol, this readily-extractable residue presumably representing the insecticide that may be picked up by insects. 相似文献
We describe the abnormal magnetic resonance (MR) imaging findings in the deep digital flexor tendon (DDFT) and distal sesamoid bone in horses with radiographic changes compatible with navicular syndrome. Thirteen postmortem specimens were examined using a 1.5-T magnetic field, with spin echo (SE) T1-weighted, turbo SE (TSE) proton density-weighted (with and without fat saturation), and fat saturation TSE T2-weighted sequences. The limbs were then dissected to compare the MR findings with the gross assessment and histologic examination of the DDFT and distal sesamoid bones. Tendonous abnormalities were detected by MR imaging in 12 DDFTs and confirmed at necropsy. Most tendon lesions were located at the level of the distal sesamoid bone and the proximal recess of the podotrochlear bursa. Tendon lesions were classified based on their MR imaging features as core lesions, dorsal lesions, dorsal abrasions, and parasagittal splits. Areas of increased MR signal in the DDFTs were characterized by tendon fiber disturbance and lack of continuity of the collagen fibers, foci of edema, hemorrhages, and formation of lakes containing eosinophilic plasma-like material or amphophilic material of low density. Bone marrow signal alterations in the distal sesamoid bone were seen in all digits. Two main phenomena were responsible for the abnormal signal, respectively, in T1-weighted (decreased signal) and in T2-weighted fat-suppressed images (increased signal): a decrease in the fat marrow content in the trabecular spaces and an increase in the fluid content. Histologic examination revealed foci of bone marrow edema, hemorrhage, necrosis, and fibrosis. Cyst formation and trabecular abnormalities (disorganization, thinning, remodelling) were also observed in areas of abnormal signal intensity. Pseudomonas syringae pv.pisi, pea bacterial blight agent, is seed-transmitted. Some aspects of its life cycle and its biology were investigated. The colonization of pea plants obtained from naturally infected seeds was studied in natural conditions while high populations of bacteria developed on plants showing no symptoms. Two streptomycin-resistant mutants were used to study the epiphytic life of the pathogen. Populations were monitored in different host-parasite compatibilities. When race 2 or race 6 of the pathogen was surface-inoculated on susceptible cultivars, a decrease of population size was observed during the following one to three days but was followed by an increase to levels 1000 times greater than the initial number detected, without symptoms for most of the plants. When race 2 was surface-inoculated on resistant genotypes or race 6 on non-host plants, bacteria did not multiply but population levels slightly decreased.Pseudomonas syringae pv.pisi shows a resident phase and its development is race-specific. Weeds collected in naturally contaminated pea fields, diseased or not, often harboured the pathogen but with levels smaller than those observed on peas. Pea crop debris and volunteers kept high levels of bacteria for at least eight months after the harvest of a diseased crop. As long as two pea crops are not grown one after the other in the same field, it is unlikely that debris and volunteers will act as an important inoculum source. The development of this pathogen during the growing season is considered as an important parameter to take into account for controlling the disease through seed health testing. 相似文献
