Amaranthus leaf mottle virus (AmLMV) was classified as a member of the genus Potyvirus on the basis of its particle morphology, serology, and biological properties (Casetta et al., 1986). Based on these properties, an Amaranthus viridis-infecting virus isolated in Spain, causing mottle and leaf blistering as well as reduced growth has been identified as AmLMV. The 3′ terminal genomic region of this and a reference isolate from Italy has been sequenced and reveals a 95% nucleotide identity between the two isolates. The sequenced part comprises the coat protein with 281 amino acids and 315 nucleotides of the 3′ untranslated region (UTR) preceding a polyadenylated tail. Pairwise comparisons and phylogenetic analysis of the nucleotide and deduced amino acid sequences of the CP and 3′ UTR of the cloned cDNAs with those of other potyviruses shows that AmLMV is a distinct potyvirus closely related to Potato virus Y.  相似文献

Diverse actinomycetes produce a family of structurally and biosynthetically related non-ribosomal peptide compounds which belong to the chromodepsipeptide family. These compounds act as bisintercalators into the DNA helix. They give rise to antitumor, antiparasitic, antibacterial and antiviral bioactivities. These compounds show a high degree of conserved modularity (chromophores, number and type of amino acids). This modularity and their high sequence similarities at the genetic level imply a common biosynthetic origin for these pathways. Here, we describe insights about rules governing this modular biosynthesis, taking advantage of the fact that nowadays five of these gene clusters have been made public (thiocoraline, triostin, SW-163 and echinomycin/quinomycin). This modularity has potential application for designing and producing novel genetic engineered derivatives, as well as for developing new chemical synthesis strategies. Leaf rust caused by Puccinia triticina is the most common and widely distributed of the three wheat rusts. Losses from leaf rust are usually less damaging than those from stem rust and stripe rust, but leaf rust causes greater annual losses due to its more frequent and widespread occurrence. Yield losses from leaf rust are mostly due to reductions in kernel weight. Many laboratories worldwide conduct leaf rust surveys and virulence analyses. Most currently important races (pathotypes) have either evolved through mutations in existing populations or migrated from other, often unknown, areas. Several leaf rust resistance genes are cataloged, and high levels of slow rusting adult plant resistance are available in high yielding CIMMYT wheats. This paper summarizes the importance of leaf rust in the main wheat production areas as reflected by yield losses, the complexity of virulence variation in pathogen populations, the role cultivars with race-specific resistance play in pathogen evolution, and the control measures currently practiced in various regions of the world.  相似文献

Coffee is globally one of the most important export crops and is a prominent part of the economy in more than 50 countries in Latin America, Africa and Asia. In Colombia, it has been the leading export commodity for more than a century. However, genetic research on coffee has been rather sparse and mainly focused on the two major cultivated species, Coffea arabica L. and C. canephora P., leaving unexplored the genetic potential in other species. In this study, an interspecific mapping population consisting of 101 F1 hybrid plants from a cross between the diploid species C. liberica and C. eugenioides was evaluated for genetic segregation at 618 molecular marker loci. Of these, 168 SSRs and two ESTs exhibited polymorphic patterns that allowed segregation analysis and genetic linkage estimations. A genetic map consisting of 146 co-dominant loci and 11 predicted linkage groups was constructed using the mapping software JoinMap 3.0. The conjoined maternal/paternal map length is 798.68 cM, has an average saturation density of 6.01 cM/interval, and covers an estimated 66–86 % of the diploid coffee genome. Approximately 24 % of loci had null alleles, and 23.5 % exhibited segregation distortion. Knowledge derived from this study has important applications for quantitative trait locus analysis and marker-assisted selection in Colombian coffee breeding programs.  相似文献

Pro-inflammatory cytokines (particularly IL-12) are important for initiating protective T helper 1 (Th1)-type immune responses and hence vital for combating intracellular infections and tumours. In situ hybridization (ISH) provides a powerful diagnostic tool allowing the identification and localization of cells producing these mediators in fixed tissues. The objective of this work was to produce a bovine IL-12p40 probe that allows detection of IL-12p40 mRNA in fixed tissues from different ruminant species. The RNA probe sequence is 447 bp in length and from a region with high cross-species-sequence homology (>97.3% homology) to the ovine, cervine, caprine and bubaline IL-12p40 genes. ISH was carried out on paraformaldehyde fixed tissues collected from cattle, sheep and goats. The probe was efficient in identifying IL-12p40 expressing cells in fixed tissues from all these species. In conclusion, the IL-12p40 probe was efficient in identifying and localizing cells that express IL-12p40, and provides a good immuno-diagnostic technique to characterize immune responses in fixed tissues.  相似文献

We hypothesized that the inclusion of flaxseed in the diets of lactating dairy cows will increase urinary and fecal concentrations of the lignans, secoisolariciresinol diglycoside (SDG), enterolactone and enterodiol, reduce intrafollicular concentrations of IGF-I and estradiol, and subsequently reduce estradiol and oxytocin receptor expression in the endometrium. To test this hypothesis, 27 cycling, lactating Holstein cows were assigned to 1 of 3 diets supplemented with saturated fatty acids (SAT), flax (FLX), or sunflower (SUN) seed. Rations were formulated to provide 750 g supplemental fat/cow/d in all dietary groups. Ovulation (Day 0) was synchronized, and 5 d later, follicles>8 mm were ablated by an ultrasound-guided procedure in all cows. Samples of blood (Days 0 to 14), follicular fluid (Day 5 and 15), endometrium (Day 15), as well as urine and feces were collected in a subset of the animals. The fecal concentrations of SDG and enterodiol were higher (P<0.05) in cows fed FLX than in those fed SAT or SUN. Enterodiol increased (P<0.05) in urine samples of cows fed FLX, compared to those of cows fed SUN. However, follicular estradiol concentrations on Day 5 and 15 and endometrial concentrations of estradiol and oxytocin receptors on Day 15 did not differ among the dietary groups. Mean plasma progesterone concentrations were higher (P<0.05) in cows fed FLX and SUN than in those fed SAT. In summary, a diet supplemented with flaxseed increased the concentrations of SDG and enterodiol in feces, as hypothesized, but did not alter intrafollicular concentrations of IGF-I or estradiol, or endometrial populations of oxytocin or estrogen receptors in lactating dairy cows.  相似文献

Northern scallop Argopecten purpuratus aquaculture relies on an efficient all year-round larval supply. Larvae are generally produced in closed aquaculture systems (CAS) using the batch techniques with periodical water changes. For instance, survival rates are greatly variable and can range from 0 to 80% making production of scallop larvae uncertain. The main goal of this study was to determine the feasibility of rearing scallop larvae in a recirculating aquacultural system (RAS), and secondarily to compare scallop larval growth rate and time length to reach the settling stage when reared with a traditional Chilean CAS technique and in a novel RAS technique in an industrial-like approach.Several batches of larvae were cultured in CAS and RAS. Larvae were fed on Isochrysis galbana cultured in 35-L tubular photobioreactors. Growth rates were significantly different (F11,2840 = 274.66; p < 0.001). All scallop larvae cultured in CAS showed lower growth rates ranging within 4.49 and 7.30 μm day?1 and protracted period of culture until settlement (at least 10 more culture days) than those reared in RAS (growth rates between 9.56 and 13.15 μm day?1). However, final survival (from D-larvae until settlement) of larvae reared in CAS showed higher values than those values recorded for larvae cultured on RAS. Higher growth rates observed in RAS could be attributed to a reduction in daily manipulation of the animals and/or more feed availability as well as higher temperatures and a steady state conditions in water quality. Even though, the reduction in time for rearing larvae until settlement in RAS was high, the comparison between systems is more significant in view of the reduction in make up seawater from 100% of system volume (CAS) to less than 10% of system volume (RAS). Therefore, RAS was independent from daily water quality variation from natural seawater by increasing water retention time, and with that improve water quality steady state conditions. Results of this research show that a more efficient use of water and heating systems than generally used in the Chilean hatchery industry is achievable. This is an important result since it could lead to significant reductions in the cost of operating a scallop hatchery, however further work is required to accurately compare the two systems (CAS and RAS). The main result from this research is that scallop larvae can be cultivated using recirculating aquaculture systems (RAS) as a method to increase production. The information reported in this paper will be useful for the improvement of scallop larvae culture techniques under controlled conditions.  相似文献

An experiment was conducted to evaluate whether supplementation with a probiotic could enhance digestion and reduce mortality in the volcano rabbit in captivity. Two enclosures at Chapultepec Zoo, Mexico(114 individuals) were used in a crossover design(two periods of 60 days) with the following treatments: control group and supplementation with Saccharomyces cerevisiae(2×108 CFU/exhibit/day). Supplementation with the probiotic negatively affected(P0.01) the digestibility of dry matter, organic matter, neutral detergent fiber(NDF) and energy. Mortality increased(P0.04) following supplementation with the probiotic(4.26% vs. 8.89%), primarily in the juvenile rabbits. The results indicate that yeast supplementation in the volcano rabbit negatively affects digestion and mortality in captivity.  相似文献
