In the genetic code, UGA serves as a stop signal and a selenocysteine codon, but no computational methods for identifying its coding function are available. Consequently, most selenoprotein genes are misannotated. We identified selenoprotein genes in sequenced mammalian genomes by methods that rely on identification of selenocysteine insertion RNA structures, the coding potential of UGA codons, and the presence of cysteine-containing homologs. The human selenoproteome consists of 25 selenoproteins. 相似文献

Tumor necrosis factor (TNF) is a key regulator of inflammatory responses and has been implicated in many pathological conditions. We used structure-based design to engineer variant TNF proteins that rapidly form heterotrimers with native TNF to give complexes that neither bind to nor stimulate signaling through TNF receptors. Thus, TNF is inactivated by sequestration. Dominant-negative TNFs represent a possible approach to anti-inflammatory biotherapeutics, and experiments in animal models show that the strategy can attenuate TNF-mediated pathology. Similar rational design could be used to engineer inhibitors of additional TNF superfamily cytokines as well as other multimeric ligands. 相似文献

The objective of this paper is to review the published literature on improving properties of wood composites through thermal pretreatment of wood. Thermal pretreatment has been conducted in moist environments using hot water or steam at temperatures up to 180 and 230 °C, respectively, or in dry environments using inert gases at temperatures up to 240 °C. In these conditions, hemicelluloses are removed, crystallinity index of cellulose is increased, and cellulose degree of polymerization is reduced, while lignin is not considerably affected. Thermally modified wood has been used to manufacture wood–plastic composites, particleboard, oriented strand board, binderless panels, fiberboard, waferboard, and flakeboard. Thermal pretreatment considerably reduced water absorption and thickness swelling of wood composites, which has been attributed mainly to the removal of hemicelluloses. Mechanical properties have been increased or sometimes reduced, depending on the product and the conditions of the pretreatment. Thermal pretreatment has also shown to improve the resistance of composites to decay. 相似文献

Intensified by global warming, heat stress is one of the major concerns in many farmlands that severely disturb plant productivity. Plant genetic engineering by regulating heat shock proteins, RNA-binding proteins, and molecular chaperones is an efficient strategy for maintaining plant productivity under heat stress conditions. Plant performance and tolerance to heat stress are highly dependent on their balance, interactions, structural, and functional diversities. Thus, this review article is undertaken to discuss the effectiveness of these proteins in conferring tolerance to heat stress and to incentivize their coregulation to achieve better results in breeding programs in future. 相似文献

Drought stress is considered one of the major constraints to crop production with devastating effects worldwide. Methylglyoxal (MG) homeostasis plays an essential role in promoting plant growth, development, metabolic adaptation, signal transduction, and thereby responses to drought stress. Manipulation of genes encoding aldose-aldehyde reductases (ALRs), aldo– keto reductases (AKRs), genes that are involved in maintaining high ascorbate:glutathione (AsA:GSH), glutathione:glutathione disulfide (GSH:GSSG) contents and ratios, as well as jasmonic acid (JA) biosynthesis are of critical importance for improving MG detoxification, and thereby tolerance to drought stress. MG detoxification by maintaining optimal abscisic acid (ABA), JA, salicyclic acid (SA), and brassinosteroid (BR) homeostasis and crosstalk is also essential to optimize plant performance under both normal and drought stress conditions. 相似文献

The early and accurately detection of brucellosis incidence change is of great importance for implementing brucellosis prevention strategic health planning. The present study investigated and compared the performance of the three data mining techniques, random forest (RF), support vector machine (SVM) and multivariate adaptive regression splines (MARSs), in time series modelling and predicting of monthly brucellosis data from 2005 (March/April) to 2017 (February/March) extracted from a national public health surveillance system in Hamadan located in west of Iran. The performances were compared based on the root mean square errors, mean absolute errors, determination coefficient (R2) and intraclass correlation coefficient criteria. Results indicated that the RF model outperformed the SVM and MARS models in modeling used data and it can be utilized successfully utilized to diagnose the behaviour of brucellosis over time. Further research with application of such novel time series models in practice provides the most appropriate method in the control and prevention of future outbreaks for the epidemiologist. 相似文献

In response to concerns that novel infectious agents were introduced through the movement of eggs as Atlantic salmon aquaculture developed in British Columbia (BC), Canada, we estimated the prevalence of infectious agents in archived return‐migrating Sockeye salmon, from before and during aquaculture expansion in BC (1985–94). Of 45 infectious agents assessed through molecular assays in 652 samples, 23 (7 bacterial, 2 viral and 14 parasitic) were detected in liver tissue from six regions in BC. Prevalence ranged from 0.005 to 0.83 and varied significantly by region and year. Agent diversity ranged from 0 to 12 per fish (median 4), with the lowest diversity observed in fish from the Trans‐Boundary and Central Coast regions. Agents known to be endemic in Sockeye salmon in BC, including Flavobacterium psychrophilum, Infectious haematopoietic necrosis virus, Ceratonova shasta and Parvicapsula minibicornis, were commonly observed. Others, such as Kudoa thyrsites and Piscirikettsia salmonis, were also detected. Surprisingly, infectious agents described only recently in BC salmon, Ca. Branchiomonas cysticola, Parvicapsula pseudobranchicola and Paranucleospora theridion, were also detected, indicating their potential presence prior to the expansion of the aquaculture industry. In general, our data suggest that agent distributions may not have substantially changed because of the salmon aquaculture industry. 相似文献

As amino acids (AAs) are vital molecules in the metabolism of all living organisms and are the building blocks of enzymes, a 6-week feeding trial was conducted for determining the influence of dietary essential amino acid (EAA) deficiencies on pancreatic, plasma, and hepatic enzyme activities in silvery-black porgy (initial weight 4.7?±?0.01 g) juveniles. Eleven isoproteic (ca. 47%) and isoenergetic (ca. 20.5 MJ kg?1) diets were formulated including a control diet, in which 60% of dietary nitrogen were provided by intact protein (fish meal, gelatin, and wheat meal) and 40% by crystalline AA. The other 10 diets were formulated by 40% reduction in each EAA from the control diet. At the end of the experiment, fish fed with threonine-deficient diet showed the lowest survival rate (P?<?0.05), whereas growth performance decreased in fish fed all EAA-deficient diets, although the reduction in body growth varied depending on the EAA considered. Pancreatic enzymes (trypsin, lipase, α-amylase, and carboxypeptidase A) activities significantly decreased in fish fed the EAA-deficient diets in comparison with fish fed the control diet (P?<?0.05). Fish fed with the arginine-deficient diet had the highest plasma and liver alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels (P?<?0.05). Plasma and liver lactate dehydrogenase and superoxide dismutase showed the highest and lowest values, respectively, in fish fed the arginine and lysine-deficient diets (P?<?0.05). Plasma metabolites were significantly affected by dietary EAA deficiencies (P?<?0.05). The results of this study suggesting dietary EAA deficiencies led to reduction in growth performance as well as pancreatic and liver malfunction. Furthermore, arginine and lysine are the most limited EAA for digestive enzyme activities and liver health in silvery-black porgy. 相似文献
