Anatomical and physiological characters of the leaf surface and its physico‐chemical environment substantially influence the density and diversity of phyllosphere‐inhabiting microorganisms, which may include natural antagonists of important pathogens. The objective of this investigation was to quantify the phyllosphere (i.e. leaf surface) microbial population in a range of rice varieties grown in Sri Lanka and to identify the leaf anatomical and physiological characters that determine the density and diversity of phyllosphere microbes. Fifteen rice varieties including both traditional and new high‐yielding varieties were used in a planthouse experiment and a field experiment in two consecutive seasons to quantify the phyllosphere microbial population and measure leaf characters that may influence it. There were highly significant intervarietal variations in the density and diversity of epiphytic bacterial, fungal and total microbial populations under both planthouse and field conditions. However, there was no difference between traditional and new, high‐yielding varieties in their capacity to harbour epiphytic microbes in the phyllosphere. Total microbial density (TMD) under both conditions showed positive correlations with leaf hair density, stomatal density and transpiration rate. Under planthouse conditions, TMD was also positively correlated with leaf hair length and negatively correlated with leaf temperature. These correlations can be explained in terms of providing favourable microsites on the phylloplane for epiphytic microbial growth.  相似文献

Very few studies deal with the rare earth element (REE) content within paddy soil systems. This paper presents, for the first time, the distribution of REEs in paddy soils from three different regions in Sri Lanka. Seventy samples from the Kalutara and Kandy regions located in the Wet Zone and Anuradhapura region in the Dry Zone were selected for this study. ICP-MS measurements were performed on the soil samples to determine their respective REE concentrations. The sum content of all the REEs (REE) in these three regions were 182, 186 and 132 g/g, respectively. The mean concentrations for the 70 soil samples in µg/g were 35(La), 77 (Ce), 7.6 (Pr), 27.6 (Nd), 4.8 (Sm), 0.87 (Eu), 4.1 (Gd), 0.54 (Tb), 2.91 (Dy), 0.56 (Ho), 1.57 (Er), 0.21(Tm), 1.44 (Tb), and 0.23 (Lu). The chondrite-normalized REE patterns of the paddy soils from the three study areas were similar, which may be a reflection of comparable parent materials. Kalutara soils, which are richer in organic matter and kaolinite, showed a higher lighter REE content and lower heavy REE content with a more pronounced negative Eu anomaly with respect to the other regions. Whilst this may be a result of subtle differences in soil parent materials, it could also reflect secondary processes related to clay mineral and organic matter adsorption of REEs in paddy soils  相似文献

Guayule (Parthenium argentatum Gray) is a potential source of commercial natural rubber. Its commercialisation depends mainly on economical plant production. The objective of this study was to evaluate the performance of improved lines in Australia. Seeds from five improved lines (AZ-1, AZ-2, AZ-3, AZ-5 and AZ-6) and two previously developed guayule lines (N 565 and 11591) were obtained from the Agricultural Research Service (ARS) of the United States Department of Agriculture (USDA). Seedlings from these lines were grown in a glasshouse for 3 months and later transplanted in a field experiment in early September 2001. Plant height and width were monitored from transplanting to 62 weeks at regular intervals. After 62 weeks, plant dry matter production, rubber and resin content, and yields were analysed. Plant height and width of the improved lines were higher than N 565 and 11591. Plant dry matter, rubber and resin yields were significantly different among lines. Of the five lines, AZ-1 and AZ-2 produced rubber yields of 620 and 550 kg/ha, respectively and these yields were significantly greater than for N 565 (371 kg/ha) and 11591 (391 kg/ha). AZ-1 and AZ-2 also produced significantly higher resin yields, 727 and 668 kg/ha, respectively, than those for N 565 (436 kg/ha) and 11591 (325 kg/ha). Rubber and resin yield increase of lines, AZ-1 and AZ-2, were in the range of 41–68% and 53–123%, respectively over N 565 and 11591. AZ-1 tended to produce higher rubber and resin yields than AZ-2 but exhibited highly variable plant height (CV=25%) and width (CV=41%) indicating potential for further genetic improvement. AZ-2 offers the best combination of desirable characters including early vigour, uniformity and comparatively higher rubber and resin yields.  相似文献

Pectin is a biodegradable polysaccharide, and it has been recently applied as a gene delivery, drug delivery, wound healing and tissue engineering agent. In this study, pectin was extracted from pomelo (Citrus maxima) peel and characterized. The extraction recovery of pectin form pomelo peel was 14.5%, and it had 72.56% degree of esterification, 1,245.56 equivalent weight, 7.82% methoxyl and 68.27% anhydrouronic acid contents. Use of pomelo pectin as a hatching enhancing agent for fish embryos and its effect on hatching enzyme 1 (ZHE1) was investigated. The pectin‐exposed zebrafish embryos (100 µg/ml) showed significantly (p < .05) higher hatching rate (96.6%) compared with untreated (control) embryos (66.6%) at 60 hpf. The mRNA expression of ZHE1 was also significantly (p < .05) elevated up to 55.6‐fold in pectin‐exposed embryos at 24 hpf. In situ hybridization results revealed remarkably strong expression of ZHE1 in pectin‐exposed embryos compared with the control group. In addition, considerably larger size of the hatching gland was observed in pectin‐exposed larvae than that of the unexposed larvae group. These results clearly indicate that pectin isolated from pomelo peel has an ability to enhance the hatching process of zebrafish embryos via upregulation of ZHE1.  相似文献

Five distinct lipopolysaccharide (LPS) core types, namely R1-R4 and K12 have been identified in Escherichia coli. The aims of this study were to determine, primarily by means of PCR, the distribution of those oligosaccharide core types among avian pathogenic E. coli and their relationship to phylogenetic groups. To identify putative avian pathogenic E. coli, serum resistance and the presence of three virulence genes encoding temperature sensitive haemagglutinin (tsh), increased serum survival (iss) and colicin V (cvaC) were determined. Of the 143 clinical isolates examined 62% possessed the R1 core, 22% were R3, 13% were R4 and 3% were R2. Fifty commensal isolates consisted of 58% with R1 core, 38% with R3 core, 4% with R4 core, and none with R2. None of the isolates were of K12 core type. The distribution of core oligosaccharide types in clinical and commensal isolates were not statistically significant (P=0.51). Three genes, tsh, iss and cvaC were found in E. coli of all four core types. The genes tsh (P<0.001) and iss (P=0.03412) were significantly associated with the R4 core oligosaccharide type. The isolates containing R4 core type LPS were mainly confined to phylogenetic group D. The widespread R1 core type showed less ability to possess virulence genes and 83% were in the phylogenetic group A. Results of this study indicated that E. coli with R1, R2, R3 and R4 were important in causing infections in chickens and further, the E. coli with R4 core type were less common among commensals, possessed more virulence genes and were related to phylogenetic groups pathogenic for poultry.  相似文献

Avian pathogenic Escherichia coli (APEC) causes economically significant infections in poultry. The genetic diversity of APEC and phylogenetic relationships within and between APEC and other pathogenic E. coli are not yet well understood. We used multilocus sequence typing (MLST), PCR-based phylogrouping and virulence genotyping to analyse 75 avian E. coli strains, including 55 isolated from outbreaks of colisepticaemia and 20 from healthy chickens. Isolates were collected from 42 commercial layer and broiler chicken farms in Sri Lanka. MLST identified 61 sequence types (ST) with 44 being novel. The most frequent ST, ST48, was represented by only six isolates followed by ST117 with four isolates. Phylogenetic clusters based on MLST sequences were mostly comparable to phylogrouping by PCR and MLST further differentiated phylogroups B1 and D into two subgroups. Genotyping of 16 APEC associated virulence genes found that 27 of the clinical isolates and one isolate from a healthy chicken belonged to highly virulent genotype according to previously established classification schemes. We found that a combination of four genes, ompT, hlyF, iroN and papC, gave a comparable prediction to that of using five and nine genes by other studies. Four STs (ST10, ST48, ST117 and ST2016) contained APEC isolates from this study and human UPEC isolates reported by others, suggesting that these STs are potentially zoonotic. Our results enhanced the understanding of APEC population structure and virulence association.  相似文献

Shrimp farming industries are subjected to severe economic loss due to a disease called white spot syndrome, a viral disease caused by white spot syndrome virus (WSSV) in penaeid shrimp. Numerous active compounds in the market possess anti-viral activity against the white spot syndrome virus, yet the issue remains unsolved. The present study was carried out to determine the anti-viral activity of methyl 1-chloro-7-methyl-2-propyl-1h-benzo[d] imidazole-5-carboxylate (C13H15N2O2Cl) against WSSV. The anti-viral activity of the synthetic compound was determined in freshwater crabs. Crabs were divided into three different experimental groups: healthy control groups (N.C.) received NTE buffer, positive control group (P.C.) crabs received WSSV, and treatment group crabs received WSSV along with synthetic weight compound. Experimental groups were observed for 30 days post-infection. Three different organs (gills, muscles, and head soft tissue (HST)) were dissected from all three groups and analyzed using molecular-based techniques, including polymerase chain reaction (PCR), Western blot, and histopathology. Clinical signs of WSSV were observed in the positive and N.C. groups; however, the treatment group showed a 100% survival rate. Confirmation was done using PCR, Western blot, and histopathology. These results demonstrated that the given synthetic compound has significant anti-viral activity against WSSV.

Summary

Chlorophyll (Chl) degradation and the formation of Chl derivatives in stored leaves of Japanese bunching onion (JBO; Allium fistulosum L., genome FF) were determined to elucidate the mechanism of Chl degradation by using alien monosomic addition lines (AMALs; FF+1A to FF+8A) of JBO from shallot (A. cepa L. Aggregatum group, genome AA). Year-round changes in Chl content in JBO and in the AMALs were also detected over 2 years. The Chl content in FF was significantly influenced by the alien chromosomes from shallot. In particular, the Chl contents of FF+4A or FF+5A, were higher in Winter than in FF, which suggests that chromosomes 4A and 5A from shallot had a larger influence on the formation of Chl than the other chromosomes. Chl contents in FF+3A and FF+5A decreased greatly in the leaves of AMALs and JBO during storage at 25°C, whereas in FF+4A the Chl content was the lowest compared to the control, FF. Chlorophyllide (Chlide) a, C132-hydroxychlorophyll (OHChl) a, pheophorbide a, and pheophytin (Phy) a, as derivatives of Chl a, were present mainly during storage of JBO and the AMALs. Phy alevels increased with Chl degradation, especially in FF+3A, while OHChl alevels in FF and FF+3A showed a decline during storage. In addition, Chlide alevels in all lines diminished during storage.Thus, Chl amay be degraded, in part, through Phy a, as well as Chlide aand OHChl a, in JBO and the AMALs during storage.  相似文献

Thirty isolates of Fusarium oxysporum from wilted Welsh onion plants were examined for their diversity in nucleotide sequences of the ribosomal DNA (rDNA) intergenic spacer (IGS) region and for pathogenicity with regard to five Welsh onion cultivars. Phylogenetic analysis based on the IGS sequences revealed polyphyletic origins of the isolates and a relationship between phylogeny and pathogenicity; low virulence isolates differed genetically from those with high and moderate virulence. Mating type analysis revealed that all F. oxysporum isolates were MAT1-1 idiomorphs, suggesting that the pathogens may be clonal in the fields examined.  相似文献
