The degree of crop sensitivity to water deficit, during the different developmental stages of a plant, is an important aspect to consider in tropical semiarid regions. The effect of water deficit on flowering dynamics and fruit production of sweet pepper (Capsicum chinense) was evaluated under several water regimes, during three consecutive years. A complete randomized block design with three replicates was employed. Flowering dynamics and fruit production were measured weekly, and water potentials were obtained twice during the experiments. For all years, flowering began at approximately 70 days after transplanting. In the most favourable year (trial 1996), there were no differences in total flower and fruit production in terms of irrigation frequency. For the other less favourable years, flowering and fruit production were significantly different between irrigation frequencies. Our results show that low water availability, prior to flowering, reduces the number of flowers produced and retards the occurrence of maximum flowering. On the other hand, a water deficit during the period between flowering and fruit development reduces final fruit production. This suggests that a water deficit during this last stage affects final yield to a greater extent.  相似文献

Terpenic acids are under development as therapeutic agents in numerous treatments. In support of pharmacokinetic and toxicological evaluations, a robust assay based on high-performance liquid chromatography (HPLC) was developed for the analysis of the terpenoids in human serum. For a clear understanding of the differences in biological activity of these compounds, the interactions between oleanolic or betulinic acids and human serum protein have been studied by ultraviolet-visible (UV-vis) absorption under physiological conditions. A combination of liquid/liquid extraction, centrifugation, and consecutive HPLC resulted in simultaneous separation, identification, and quantification of the oleanolic, betulinic, and ursolic acids. The validity of the developed method was established by determining linearity, recovery, precision, accuracy, limit of detection, and quantification. Detection limits were in the range of 3.3-4.3 ng/mL, and linearity values ranged up to 1 μg/mL. The repeatability of the method was good. All compounds can be well-distinguished by order of elution during liquid chromatography. The pentacyclic triterpenoids have been identified by retention time comparison to pure standards and quantified by an internal standard. The results by UV-vis absorption spectra experiments (240-340 nm) indicate that protein structures have been perturbed in the presence of oleanolic and betulinic acids.  相似文献

A study was carried out to determine the changes in phenolic composition induced by tuberculosis infection in olive trees. Four ethanolic extracts were compared: olive leaf from shoots affected by Pseudomonas savastanoi pv. Savastanoi, nodules induced by this bacteria, leaf from healthy (asymptomatic) shoots, and shoots. Among the differences found, the presence of a phenolic compound in nodules was significant in much larger quantities than in leaf or shoots. Mass spectrometric analysis showed this compound to be verbascoside. The enhancement of its biosynthesis could be related to the defense mechanisms of the tree in the nodules induced by P. savastanoi and suggests the possibility of exploration of natural and biotechnological sources of this compound.  相似文献

Fifty bifidobacterial strains isolated from different parts of gastrointestinal tract of calves during the milk-feeding period were tested for susceptibility to 28 antibiotics using disc diffusion method on TPY agar. Spectrum of antibiotics included commonly used medicaments in veterinary medicine. Seven strains from culture collection DSMZ were taken as a control. Most of the strains tested were susceptible to penicillins, cephalosporins, macrolids, chloramphenicols, bacitracin, linkomycin and resistant to aminoglycosides and aminocyclitols, quinolones, sulphonamides and colistin sulphate. All tetracyclines and enrofloxacin showed a variable effect against bifidobacterial strains. Our results suggest that strains resistant to antibiotics tested seem to be suitable for administration to young ruminants during antibiotic treatment.  相似文献

Chemical indicators of heat treatment in fortified and special milks   总被引:6,自引:0,他引:6  
Carbohydrate and furosine contents in 12 commercial fortified and special milk samples (pasteurized goat's and ewe's milks; ultrahigh-temperature (UHT) goat's milk, UHT milks fortified with calcium, magnesium, fiber, or royal jelly and honey; and lactose-hydrolyzed milks) were analyzed. Except for lactose-hydrolyzed milks, furosine, lactose, lactulose, galactose, glucose, N-acetylgalactosamine, N-acetylglucosamine, and myo-inositol contents were similar to the previously reported values for UHT or pasteurized milk samples. In lactose-hydrolyzed milks, lactulose was not detectable and lactose was present in low amount; high levels of glucose, galactose, fructose, tagatose, and furosine were also detected in this type of milk. Results found in commercial milks were compared to those obtained in laboratory-prepared UHT milks with lactose hydrolyzed prior to heating. Hydrolysis of lactose before thermal treatments promoted elevated accumulation of reducing sugars (galactose and glucose) that could be partially converted to the corresponding isomers (tagatose and fructose) during heating. In addition, the reducing sugars could also react with the amino groups of proteins, giving rise to the corresponding Amadori compound. According to the obtained results, heating prior to hydrolysis of lactose is suggested to avoid a considerable loss of available lysine.  相似文献

We studied the distribution of Hg in sediments, fish, and crayfish in a 60 km reach of the Upper Wisconsin River that formerly received Hg in discharges from pulp and paper mills. The most heavily contaminated strata of sediments were deposited during the 1950s and early 1960s and buried under subsequent deposits; however, surficial sediments remained substantially enriched at certain sites in 1981. Median concentrations of Hg in surficial sediments, adjusted for grain size, were at least 10-fold greater at the main study area than at an upstream reference site. Total concentrations exceeded 1.0 μg g?1 wet weight in axial muscle tissue in only 2 of 173 fish analyzed from the study area; however, historical comparisons revealed that Hg contamination of fish (common carp Cyprinus carpio and walleye Stizostedion vitreum vitreum) and crayfish (Orconectes) in the river had not decreased since the early 1970s. The availability of Hg to biota in this system may be enhanced by rapid methylation of the metal in surficial sediments, despite burial of the most heavily contaminated sediments. Management practices for this river should be designed to prevent conditions favoring mobilization and methylation of Hg in sediments.  相似文献

In order to evaluate the effects of hyperthermia on adriamycin cardiomyopathy and its relationship with heat shock protein induction and myosin accumulation, female Sprague-Dawley rats (21-24 days) were randomized into four groups: the control, adriamycin, temperature and temperature-adriamycin groups. Adriamycin was injected i.v. at a dose of 27 mg/Kg (0.1 ml). The rats were exposed to a temperature of 45oC for 35 min, followed by a recovery (1 h) at room temperature prior to adriamycin treatment. Body weight was recorded weekly. The thickness of the ventricular wall and percentage of cellular damage were biometrically and ultrastructurally evaluated, respectively. Heat shock protein 25 and myosin accumulation were determined through Western blot analysis. The determinations were carried out monthly until the third month after treatment. At eight and twelve weeks after treatment, the thickness of the ventricular wall seemed to decrease in the adriamycin-treated rats in relation to the other groups. An electron microscopic analysis of the adriamycin group's left ventricular wall samples, showed more sarcomeric changes and loss of myofibrils than the control, temperature and temperature-adriamycin groups. At 24 hours after treatment with adriamycin, higher levels of heat shock protein 25 and myosin were observed (week 0) in the temperature-adriamycin group than in the control and adriamycin groups (4, 8 and 12 weeks). Hyperthermia was confirmed by a multivariate approach to induce heat shock protein 25 and myosin, which would strengthen cardiac-sarcomeric myosin arrangement.  相似文献

Five media were evaluated to determine their selectivity for Bifidobacterium sp. in hen and rabbit caecal samples. The colonies arising on the plates inoculated with the caecal samples were Gram stained and screened for the presence of fructose-6-phosphate phosphoketolase activity. Rogosa agar modified by the addition of cysteine-hydrochloride (0.05% w/v), Beeren's agar (with 5 ml/l of propionic acid as a selective agent), BS 2 agar (containing per one litre sodium propionate 15 g, lithium chloride 3 g, paromomycin sulphate 50 mg, neomycin sulphate 200 mg), and Wilkins-Chalgren agar (MW) modified by the addition of acetic acid (1 ml/l) and mupirocin (100 mg/l) were selective for Bifidobacterium sp. from rabbit caecal samples. In contrast, only MW medium was suitable for the isolation and enumeration of bifidobacteria in hen caecal samples. In conclusion, the results suggest that MW agar showed the greatest selectivity. A further advantage of this medium is its case of preparation. Therefore this agar could contribute to the study of the effects of the ingestion both probiotics and prebiotics. Finally, it could be noted that the bifidobacteria selective media should be chosen in respect of the animal species origin of the sample tested.  相似文献

The infusion of dandelion root (Taraxacum officinale) stimulated in vitro the growth of 14 strains of bifidobacteria. The utilization of oligofructans, glucose, fructose and total saccharides was determined by enzymatic and phenol-sulfuric methods. Dandelion oligofructans were important source of carbon and energy for bifidobacteria tested.  相似文献
