In vegetable brassicas, cytoplasmic male sterility (CMS) system is the most preferred mechanism for hybrid seed production and Ogura cybrid cytoplasm is the only source used widely. Effects of the alien cybrid cytoplasm on important quality traits need to be understood for their effective use in breeding programme. In analysing 38 Ogura cybrid cytoplasm‐based cauliflower CMS lines, it was revealed that introgression of Ogura cytoplasm resulted in significant alteration in important quality traits. Cupric reducing antioxidant activity and ferric reducing ability of plasma values were increased up to 5–10 times in different genotypes. Among 38 analysed CMS lines, concentration of plant pigments such as anthocyanin, total chlorophylls and ascorbic acid was reduced in 21, 17 and 32 genotypes, respectively. However, the concentration of total carotenoids and β‐carotene was elevated in most of the CMS lines and total carotenoid was increased up to 20 times in the CMS line, Ogu13‐85‐2A. The results indicate the role of nuclear–cytoplasmic interaction and mitochondrial genome in determining concentration of different quality traits.  相似文献

Cold-adapted bioinoculants are considered as harbingers of sustainable hill agriculture. Therefore, two previously characterized psychrotolerant diazotrophs, Pseudomonas jesenii MP1 and Rhodococcus qingshengii S10107, were evaluated for their plant growthpromoting potential for chickpea (Cicer arietinum L.) grown under natural field conditions. Comparative analysis of agronomical and biochemical crop parameters revealed the irrelevance of chemical fertilizers for chickpea production; the diazotrophs alone were sufficient to fulfil the crop''s nutritional requirement. However, the integrated use of bacterial strains in combination with urea at 20 kg N ha-1 as urea was being recommended for higher crop yield and better soil nitrogen status. Quantitative polymerase chain reaction (qPCR) and denaturing gradient gel electrophoresis (DGGE)-based soil bacterial dynamics unveiled the persistence of both diazotrophs until the end of the crop maturation period without affecting the native micro-flora. Therefore, these bioinoculants can be explored as natural nitrogen resource, and an additional incentive in their bio-formulation will be a step towards agricultural sustainability.  相似文献

Western Indian Himalaya is very rich in biodiversity. Being a cold climatic region, it possesses various psychrotolerant and psychrophilic microorganisms. Psychrotolerant bacterium Dyadobacter sp. was isolated from this region and studied for its plant growth promoting potential against four legumes and finger millet. This bacterium was able to grow at nitrogen (N) deficient medium at both 10°C and 28°C and gave positive nifH amplification that confirms the psychrotolerant and diazotrophic nature of this bacterium. Pot trial-based study showed that this bacterium was able to promote plant growth by fixing atmospheric nitrogen (N2) and making it available to plants. Agronomical parameters, leaf nitrate reductase activity, and total chlorophyll content were recorded at 30, 45, 60, and 90 days after sowing and found to be increased over their respective controls. The 16S rDNA and nifH genes were quantified by q-PCR to study the dynamics of total bacterial and diazotrophic abundance due to inoculation of Dyadobacter sp. in soil. Soil chemical properties related to soil fertility were also studied at different time intervals after sowing. We found positive correlation among soil pH, soil nifH gene abundance, soil nitrate concentration, and plant leaf nitrate reductase activity. PCR-DGGE was performed to study persistence of Dyadobacter sp. in soil after inoculation, which showed good persistence of plant growth promoting rhizobacteria (PGPR). Hence, it is concluded that Dyadobacter sp. has potential to promote plant growth by fixing atmospheric N2 and making it available to plant. Further, psychrotolerant nature of this bacterium can be exploited to enhance plant growth in cold climate agriculture due to its ability to fix atmospheric N2 at low temperature.  相似文献

Background

Tail biting is a common welfare problem in pig production and in addition to being a sign of underlying welfare problems, tail biting reduces welfare in itself. The aim of this study was to evaluate the effects of tail biting on different pre and post mortem indicators of stress in slaughter pigs and on carcass and meat characteristics. A total of 12 tail bitten (TB) and 13 control (C) pigs from a farm with a long-term tail biting problem were selected for salivary cortisol analyses before and after transport to the slaughterhouse. After stunning, samples were taken for the analysis of serum cortisol, blood lactate, intestinal heat shock protein 70 (HSP70), and meat quality characteristics. In addition, body temperature immediately after and muscle temperature 35 min after stunning were measured, as well as lean meat percentage and carcass weight.

Results

TB pigs showed a lower cortisol response to the transport-induced stress than C pigs and also had a lower serum cortisol concentration after stunning. HSP70 content in the small intestine was higher in the TB pigs than in C pigs. TB pigs had a considerably lower carcass weight therefore produced a lower total amount of lean meat per carcass than C pigs.

Conclusions

This study suggests that prolonged or repeated stress in the form of tail biting causes a blunted stress response, possibly a sign of hypocortisolism. In addition, it underlines the importance of reducing tail biting, both from an animal welfare and an economic point-of-view.  相似文献

OBJECTIVE: To detect monocarboxylate transporters (MCTs) in canine RBC membranes and to determine the distribution of lactate between plasma and RBCs. SAMPLE POPULATION: Blood samples obtained from 6 purpose-bred Beagles. PROCEDURES: Monocarboxylate transporter isoforms 1, 2, 4, 6, 7, and 8 and CD147 were evaluated in canine RBCs by use of western blot analysis. Lactate influx into RBCs was measured as incorporation of radioactive lactate. RESULTS: 2 MCT isoforms, MCT1 and MCT7, were detected in canine RBC membranes on western blot analysis, whereas anti-MCT2, anti-MCT4, anti-MCT6, and anti-MCT8 antibodies resulted in no signal. No correlation was found between the amount of MCT1 or MCT7 and lactate transport activity, but the ancillary protein CD147 that is needed for the activity of MCT1 had a positive linear correlation with the rate of lactate influx. The apparent Michael is constant for the lactate influx in canine RBCs was 8.8 +/- 0.9mM. Results of in vitro incubation studies revealed that at lactate concentrations of 5 to 15mM, equilibrium of lactate was rapidly obtained between plasma and RBCs. CONCLUSIONS AND CLINICAL RELEVANCE: These results indicated that at least half of the lactate transport in canine RBCs occurs via MCT1, whereas MCT7 may be responsible for the rest, although an additional transporter was not ruled out. For practical purposes, the rapid equilibration of lactate between plasma and RBCs indicated that blood lactate concentrations may be estimated from plasma lactate concentrations.  相似文献

The paucity of research on development of antioxidant-rich hybrids in cabbage by utilizing combining ability estimates, gene action, and heterosis motivated us to undertake this study. We made 60 crosses between 5 cytoplasmic male-sterile lines and 12 male-fertile testers during the summer of 2015, as per the line × tester design. The seedlings of all the parents and 60 F1 crosses, along with three checks, were transplanted during the Rabi (winter) season of 2015–2016 and evaluated using a randomized complete-block design. Combining ability, gene action, and heterosis were determined for different antioxidant compounds. Experimental results revealed that the range of cupric ion reducing antioxidant capacity (CUPRAC) [parents = 1.26–7.33 and hybrids = 0.04–6.54 μmol trolox/g], ferric reducing ability of plasma (FRAP) [parents = 1.65–4.76 and hybrids = 0.16–4.67 μmol trolox/g], β-carotene (parents = 0.44–2.29 and hybrids = 0.04–1.89 μg/100 g), and chlorophyll-a (parents = 0.71–4.08 and hybrids = 0.19–3.08 mg/g f.w.) for hybrids was lower than that of the parents because of outbreeding depression. The parental lines 6A, 208A, 83-5-8, and Sel-5-83-6 were found to be good general combiners for most of the antioxidant compounds studied. Based on the mean performance, specific combining ability effects and heterosis, five hybrid combinations viz., 9A × KIRC-8 for CUPRAC and FRAP; 208A × C-122 for ascorbic acid; 6A × Chhaki-2 for total carotenoids and β-carotene; 831A × Chhaki-2 for chlorophyll-a; and 6A × 83-5-8 for chlorophyll-b and total chlorophyll content, were most promising. The ratio of general combining ability (GCA) and specific combining ability (SCA) variances, i.e., [2 σ2g/(2σ2g + σ2s)], which reflects the relative importance of GCA versus SCA, was less than unity for different antioxidant compounds, which implied that for these traits, nonadditive gene effects were more important than additive effects. The numerical values of range for contribution of lines × testers interaction for different traits (41.47–70.18%) were found to be higher than the individual contribution of lines (11.24–47.22%) and testers (8.31–21.76%). Hence, heterosis could be exploited for developing antioxidant-rich hybrid cabbage.  相似文献

The capabilities of soil microorganisms to solubilize phosphate have been known for many years, but their isolation and use as crop inoculants have met with little success. Thirty-five bacterial isolates were screened for their phosphate-solubilizing ability, and two of them, PSR10 and RGR13, identified through 16S rDNA sequencing as Chryseobacterium sp. PSR10 and Escherichia coli RGR13, respectively, screened for plant growth promotion in the greenhouse. Seed inoculation of Macrotyloma uniflorum (horsegram) by Chryseobacterium sp. PSR10 showed better plant growth promotion in sterilized and unsterilized soil under greenhouse conditions and was selected for a field experiment with 100, 50 and 30% of recommended doses of nitrogen and phosphorus fertilizer. Seed inoculation with 50% of the recommended dose of nitrogen and phosphorus increased plant growth (agronomical parameters, chlorophyll content, nitrate reductase activity, phosphorus content and crop yield). We conclude that effective plant growth-promoting bacterium Chryseobacterium sp. PSR10 broadens the spectrum of phosphate solubilizers available for field applications and might be used together with 50% dose of nitrogen and phosphate.  相似文献

The Brassicas are affected by several diseases, of which black rot, Xanthomonas campestris pv. campestris (Pam.) Dowson (Xcc), is one of the most widespread and devastating worldwide. The black rot bacteria causes systemic infection in the susceptible plants and penetrate the plants through the hydathodes or wounds. Typical disease symptoms are ‘V’ shaped necrotic lesions appearing from the leaf margins with blackened veins. Periodic outbreaks of the black rot pathogen have occurred worldwide, especially in the continental regions, where high temperatures and humidity favor the incidence of disease occurrence causing huge yield loss. The challenge to control the losses in vegetable brassicas production is made more difficult by the adverse climatic changes and evolution of new pathogenic races. The development of black rot resistant hybrids/varieties is the most reliable long term practical solution for effective disease control. Identification of new resistant genetic resources, tightly linked markers with resistance loci and QTL mapping would facilitate the breeding programme for black rot resistance. Information regarding genetics of resistance and mapping of resistance genes/QTLs will accelerate the marker assisted resistance breeding in brassica crops against Xcc. In future we need to identify the race specific candidate genes for and their validation through transgenics and gene expression. Moreover, it is imperative to identify functional markers for resistance genes through identification of R gene families and their relationship with resistance expression. This comprehensive review will help the researchers working in this area to understand the dynamics of black resistance breeding and to formulate future breeding strategies.  相似文献

The Brassica vegetable crops are rich source of important antioxidant compounds having anticancer and health promoting properties. Development of F1 hybrids with better nutritional traits is one of the main breeding objectives in different vegetable crops. Our study is the first report of determining heterotic combinations utilizing cytoplasmic male sterile (CMS) and doubled haploid (DH) inbred lines for antioxidant compounds in snowball cauliflower. Twenty genetically diverse Ogura CMS lines of cauliflower and six DH male fertile inbred lines were crossed to develop 120 F1 hybrids in line?×?tester mating design. The resulting 120 test cross progenies along with 26 parents and 4 standard checks were evaluated in 10?×?15 alpha lattice design with three replications during next cropping season. The CMS lines Ogu33-1A, Ogu122-5A and Ogu119-1A were good general combiner and CMS line Ogu118-6A was poor general combiner for majority of traits. Most of the heterotic hybrids were associated with high positive SCA effects. The proportions of σ2A/D and \(\upsigma^{2}_{\text{gca}} /\upsigma^{2}_{\text{sca}}\) ratios were less than unity in all the cases indicating preponderance of non-additive gene action in the genetic control of all the traits. Highest number of heterotic hybrids with SCA effects in desired positive direction was recorded for ascorbic acid content and phenolic content followed by total carotenoid content. The F1 hybrids with better combining ability and better per se performance could be useful in accumulation of favourable allele for higher concentration of important anti-oxidant compounds.  相似文献
