The present study was conducted to demonstrate the spindle formation and behavior of chromosomes and microtubules during first division in reconstructed rat embryos produced by somatic cell nuclear transfer (SCNT) with cumulus cell nuclei. To demonstrate the effect of oocyte aging after ovulation on the cleavage of SCNT embryos, micromanipulation was carried out 11, 15 and 18 h after injection of hCG. SCNT oocytes were activated by incubation in culture medium supplemented with 5 microM ionomycin for 5 min followed by treatment with 2 mM 6-dimethylaminopurine (6-DMAP) in mR1ECM for 2-3 h. For immunocytochemical observation, the SCNT embryos were incubated with monoclonal anti-alpha-tubulin antibody and then fluorescein isothiocyanate (FITC)-conjugated goat anti-mouse IgG. Cleavage rates were significantly higher for oocytes collected after 15 and 18 h rather than for those collected 11 h after injection of hCG (56 and 53%, respectively vs. 28%; P<0.05). Premature chromosome condensation occurred before activation of the SCNT oocytes, but adequate spindle formation was only rarely observed. The distribution of microtubules in SCNT embryos after activation was different from those of fertilized and parthenogenic oocytes, i.e., a dense microtubule organization shaped like a ring was observed. Eighteen to 20 h post-activation, most SCNT embryos were in the 2-cell stage, but no nucleoli were clearly visible, which was quite different from the fertilized oocytes. In addition, first division with and without small cellular bodies containing DNA was observed in the rat SCNT embryos in some cases. The present study suggests that reorganization of transferred nuclei in rat SCNT embryos may be inadequate in terms of formation of the mitotic assembly and nucleolar reorganization.  相似文献

Feces were collected from two female and one male Siberian tigers, Panthera tigris altaica. Steroid hormones were extracted from lyophilized feces and quantified by enzyme immunoassay. The fecal contents of estradiol-17beta (E(2)) and testosterone in the females and male, respectively, changed markedly throughout the year. The fecal E(2) contents of females Nos. 179 and 238 increased at 26.4 +/- 8.0 and 28.0 +/- 14.2 day intervals, respectively. However, the fecal contents of progesterone (P(4)) in the female kept alone did not change. In contrast, the other female, which was kept with a male, had increased fecal P(4) contents after copulation. The fecal progesterone levels of the pregnant female remained high during her 106-day pregnancy.  相似文献

Xanthomonas oryzae pv. oryzae (X. o. pv. oryzae) T7174 is virulent on rice cultivar IR24 and avirulent on IR-BB2. From recent reports, some virulence and avirulence factors of plant pathogenic bacteria are transferred to plant cells through the hrp-dependent type III secretion system. In this study, we investigated the involvement of hrp genes in the compatible and the incompatible interactions between rice and X. o. pv. oryzae after co-inoculation with hrpXo mutants derived from T7174 and virulent strains. Growth of the mutants, named 74ΔHrpXo and 76ΔHrpXo, was repressed in IR24 when the mutants were applied alone. However, growth of the mutants was complemented by co-inoculation with virulent strains. Growth of bioluminescent hrpXo mutant 76ΔHrpXo in IR24 and its growth in IR-BB2 after co-inoculation with T7133, which is virulent on both cultivars, was equally complemented, as detected by bioluminescence from the mutant. On the other hand, only partial complementation of growth of T7174L76, which is a bioluminescent and pathogenic derivative of T7174, by T7133 was observed in IR-BB2. Thus, growth of the hrpXo mutant of X. o. pv. oryzae was complemented by virulent strains in both susceptible and resistant rice leaves with the parental strain. Neospora caninum was isolated from the brain of a 2-year-old dairy cow that had aborted confirmed N. caninum-infected fetuses on two occasions. The cow had an indirect fluorescent antibody titer of 1:1600 to N. caninum. The cow was killed 24 days after its second abortion and the brain was bioassayed for N. caninum in nude mice. Multifocal areas of perivascular cuffing and glial nodules were observed in the cerebrum and mesencephalon of the cow, but N. caninum was not identified in histological sections of the brain. All three nude mice inoculated with brain homogenate of the cow, developed emaciation and paralysis. Microscopical examination of the nude mice revealed systemic N. caninum infection with demonstrable tachyzoites in various organs. The parasites isolated from fresh mouse brain were transferred successfully into Vero cell cultures. PCR procedure on the purified tachyzoites obtained from the Vero cell cultures amplified the specific DNA sequence for N. caninum.  相似文献

We reported that meat chicks have either a greater capability to acclimatize to novel environments, or a blunted hypothalamic‐pituitary‐adrenal response to novel environments compared with layer chicks in a commercial base. The present study compared the differences in behavior and plasma corticosterone concentrations under isolation‐induced stress between neonatal meat and layer Nagoya chicks which had been separated from the same population. Both types of neonatal chicks reared in groups were individually separated and their spontaneous activity and distress‐induced vocalizations were monitored for 10 min. The responses of the two types were remarkably different, with the meat chicks being less active than the layer chicks. Distress‐induced vocalizations were fewer in the meat than in the layer chicks. The meat chicks spent more time in a sleeping posture during isolation‐induced stress. Plasma corticosterone concentrations measured at the end of the test tended to be higher in the layer chicks than in meat ones, but not significantly. In conclusion, the selection of Nagoya chickens for meat and layer may have trends similar to those observed in commercial chickens in relation to stress susceptibility.  相似文献

Arisaema sikokianum (Araceae) native to Japan is classified as a vulnerable species in the Red Data Book of Japan. Control of dormancy is essential for efficient corm production and forcing culture. Sprouting of both vegetative and reproductive corms was enhanced by exposure to low temperature. Vegetative corms exposed to low temperatures at 5 °C longer sprouted faster when grown at 20 °C. Effective temperatures for breaking dormancy was 5 °C. Reproductive corms treated at 5 °C longer showed shorter days to flower. Successful forcing culture was achieved; corms treated at 5 °C from November for 30 days flowered on 5th February.  相似文献

Summary Using sulfur dioxide (SO2) as catalyst, wood specimens were treated with nonformaldehyde cross-linking reagents such as glyoxal, glutaraldehyde and dimethylol dihydroxy ethyleneurea (DMDHEU). The results of dimensional stability, acoustic properties, and mechanical strength tests were compared with those obtained from formaldehyde treatment. With glyoxal and glutaraldehyde treatments, antiswelling efficiency (ASE) reached around 70%, which is comparable to the values attained by formaldehyde treatment, although the accompanying weight gain was much larger than for the latter, whereas DMDHEU did not give sufficiently high ASE. None of the treatments, significantly increased specific dynamic Young's modulus (E/) with the exception of the DMDHEU treatment, loss tangent (tan ) decreased substantially, about 50 and 60% in longitudinal and radial directions respectively, which were somewhat exceeding the formaldehyde treatment. The results suggested that the improvement of the dimensional stability and acoustic properties is partly attributed to the formation of cross-links.  相似文献

Fractions of methanol and ethanol extracts from the heartwood of white cypress pine (Callitris glaucophylla Thompson et Johnson) were investigated for their repellent activity against subterranean termite Coptotermes formosanus Shiraki worker using a two-choice semicircular filter paper test at 0.5% (w/w) concentration. Fraction CY-E2 composed of (−)-citronellic acid, guaiol, α-, β-, and γ-eudesmol isomers as well as an unknown compound, showed the highest statistically significant repellency (97.8% ± 2.2 SEM) of all fractions tested. Bioactivity-guided fractionations using high-performance liquid chromatography led to the isolation of two, oxygenated eudesmane-type sesquiterpenes with α-methylene moieties, both termite-repellent compounds. These compounds were subsequently identified as ilicic acid methyl ester (IAME) and costic acid by means of spectroscopic analyses, electron impact mass spectrometry, and nuclear magnetic resonance spectroscopy. We report the isolation of both IAME and costic acid from C. glaucophylla heartwood for the first time. This article discusses the effect of aluminum on root growth of Querucus serrata Thunb. In a 9-week hydroponics experiment, the effects of various concentrations (0 to 5.0 mM) of Al on root growth were examined. Results revealed that root biomass increases with the increase in Al concentration up to 2.5 mM, and then it tended to decrease. In the next experiment, the effects of H+ and Al3+ on roots were compared in a 4-week hydroponics experiment using three treatment solutions: control (pH 6.0), −Al (without Al, pH 3.5), and +Al (with 2.5 mM Al, pH 3.5). No clear difference in the biomass and root length between the control and −Al treated roots was observed, and root and shoot biomass were increased by Al treatment. These results confirmed that the H+ concentration level, at a pH of 3.5, is not toxic for Q. serrata and the Al-induced increase in root biomass is not caused by the amelioration of H+ toxicity by Al. In the third experiment, roots were exposed to an Al solution (pH 3.6) intermittently. This treatment clarified that Al stimulated rooting and root elongation. In the fourth experiment, the effect of 1 mM Al on root growth during a 15-month period in a sand culture were examined. This experiment confirmed that Al stimulated good growth and development of root systems at appropriate concentrations. Therefore, it is considered that Al-induced root growth enhancement occurs as a long-term and short-term phenomenon.  相似文献
