African trypanosomosis is a potentially fatal disease that is caused by extracellular parasitic protists known as African trypanosomes. These parasites inhabit the blood stream of their mammalian hosts and produce a number of pathological features, amongst which is anemia. Etiology of the anemia has been partly attributed to an autoimmunity-like mediated erythrophagocytosis of de-sialylated red blood cells (dsRBCs) by macrophages. Lactose infusion to infected animals has proven effective at delaying progression of the anemia. However, the mechanism of this anemia prevention is yet to be well characterized. Here, the hypothesis of a likely induced further modification of the dsRBCs was investigated. RBC membrane galactose (RBC m-GAL) and packed cell volume (PCV) were measured during the course of experimental trypanosomosis in mice infected with Trypanosoma congolense (stb 212). Intriguingly, while the membrane galactose on the RBCs of infected and lactose-treated mice (group D) decreased as a function of parasitemia, that of the lactose-untreated infected group (group C) remained relatively constant, as was recorded for the uninfected lactose-treated control (group B) animals. At the peak of infection, the respective cumulative percent decrease in PCV and membrane galactose were 30 and 185 for group D, and 84 and 13 for group C. From this observed inverse relationship between RBCs membrane galactose and PCV, it is logical to rationalize that the delay of anemia progression during trypanosomosis produced by lactose might have resulted from an induction of galactose depletion from dsRBCs, thereby preventing their recognition by the macrophages.  相似文献

The recent discovery of mammalian bitter, sweet, and umami taste receptors indicates how the different taste qualities are encoded at the periphery. However, taste representations in the brain remain elusive. We used a genetic approach to visualize the neuronal circuitries of bitter and sweet tastes in mice to gain insight into how taste recognition is accomplished in the brain. By selectively expressing a transsynaptic tracer in either bitter- or sweet and/or umami-responsive taste receptor cells, and by comparing the locations of the tracer-labeled neurons in the brain, our data revealed the potential neuronal bases that underlie discrimination of bitter versus sweet.  相似文献

In order to investigate the acid rain formation under the coexistence of SO2(g), H2O2(g), and HNO3(g) in the air, a mathematical model has been built and some numerical simulations have been carried out with use of the model. The simulation reveals that SO2(g) absorbed into a raindrop is released and then re-absorbed as the fall distance increases. The desorption and re-absorption processes of SO2(g) are caused by: (1) the fact that the equilibrium concentration of H2O2(aq) and HNO3(aq) in raindrops are much higher than that of SO2(aq), and (2) the fact that the oxidation reaction rate of HSO3 ? with H2O2(aq) increases with H+ concentration in raindrops. The degree of acidification of the rainwater has been estimated by introducing a raindrop size distribution. The acidification is mainly caused by the adsorption of SO2(g) in the usual case where the atmospheric concentration of SO2(g) is much higher than that of HNO3(g). With the increase in the atmospheric concentration of HNO3(g), the concentration of H+ generated from SO2(g) decreases and the contribution of HNO3(g) to the generation of H+ becomes dominant.

1. Growth changes of the collagen content and architecture in the pectoralis (PT) and iliotibialis lateralis (ITL) muscles were examined using cockerels from 1 to 14 weeks of age. 2. Total collagen content in PT muscle showed little change, but in ITL muscle reached a maximum at 5 weeks and thereafter decreased slightly until 14 weeks. The collagen content was markedly larger in ITL muscle after 5 weeks. Pyridinoline content of collagen increased abruptly from 5 to 14 weeks in both muscles, but no difference between muscle types was detected. 3. The cell size of the endomysial honeycombs increased with the development of myofibres, and the mesh size of the perimysium around the honeycombs enlarged. 4. In both muscles endomysia were an incomplete network of collagen fibrils with many foramina at one week, became a very thin membrane of felt-like fabric in 2 to 5 weeks and thereafter increased in thickness until 11 to 14 weeks. 5. Perimysial width around the secondary fasciculus differed between the muscle types after 5 weeks. In the wider perimysium of ITL muscle, the collagen fibres increased in number and size to make a stack of collagen bands around the fasciculus. In the narrower perimysium of PT muscle, a few platelets of collagen fibres also developed. 6. The perimysial collagen fibre at 1 to 2 weeks had a smooth surface and appeared to be composed of fine collagen fibrils. The fibre at 11 to 14 weeks showed a rugged surface and was composed of coarser collagen bundles that combined with each other into a net-like configuration with very slim meshes. 7. Our results showed that the collagenous components of chicken intramuscular connective tissue changed markedly during the early period of muscle growth in distribution, architecture and quality but with little difference in quantity.  相似文献

Studies were made on the number of sperms required for fertilization of ova ovulated from the bilateral ovaries in bitches. The experiment was carried out with 44 female beagles, which were divided into four groups of seven to eleven bitches each. The four groups were inseminated with 40 X 10(6) sperms/0.2 ml, 20 X 10(6) sperms/0.1 ml, 10 X 10(6) sperms/0.1 ml, and 3-5 X 10(6) sperms/0.1 ml, respectively. All the bitches were laparotomized at an optimum time for mating and inseminated at the tip of the horn of an unilateral uterus. In the four groups conception took place at a rate of 7/7, 8/8, 10/11, and 2/7, respectively. The number of puppies exceeded the number of ova ovulated on the inseminated side at a rate of 5/7, 6/8, 0/10, and 0/2 in the four groups, respectively. Therefore, it was clarified that when more than 20 X 10(6) sperms/0.1 ml were inseminated in the ipsilateral uterine horn, ova ovulated on the contralateral side were fertilized also. Then a unilateral ovary was ovariectomized in 11 bitches, which were inseminated with 10 X 10(6) sperms/0.1 ml in the ipsilateral uterine horn. As a result, only one bitch became conceptive and gave birth to only one puppy. From these results, it was considered that 10 X 10(6) sperms/0.1 ml may be most adequate for the ova on the side of insemination alone to be fertilized.  相似文献

The objective of the present study was to evaluate the effect of plant‐ingredient‐based feeds on fillet fatty acid profiles, proximate composition, carcass traits, and sensory characteristics of three Indian major carps (IMCs), Catla catla, Labeo rohita, and Cirrhinus mrigala, when fed at different levels of their body weight. An experimental feed was prepared with only plant ingredients. The IMCs were fed twice daily at 1.0, 1.5, 2.0, and 2.5% of their body weight for 150 d in 0.06‐ha earthen pond polyculture systems. The maximum growth and best feed utilization was achieved with feeding at 2% of fish biomass in the ponds. Fillet yield and lipid recovery increased significantly (P < 0.05), with increasing feeding level up to 2% of body weight and plateaued thereafter. In fillets, eicosapentaenoic acid (20:5n‐3) and docosahexaenoic acid (22:6n‐3) concentration increased with increasing feeding levels. Carcass traits, sensory characteristics, and consumer acceptance of IMC fillets were not influenced by feeding plant‐ingredient‐based feeds at different feeding rates.  相似文献

Granzyme B plays an important role in granule-mediated apoptosis by CTL. It is a well characterized component of the cytolytic machinery in mammals and a candidate for the evaluation of cytotoxic activity of CTL as an alternative to conventional cytotoxicity assay. In this study, we examined the effects of granzyme inhibitors to assess the characteristics of fish granzymes in terms of substrate specificity and the involvement of granzyme B-like in the cytotoxic response. 3,4-dichloroisocoumarin (DCI), which inhibit the activity of serine protease including all members of the granzyme family, markedly suppressed the cytotoxic activity of CTL. However, CTL-mediated cytotoxicity was significantly but not completely suppressed by the addition of carbobenzyloxy-Ile-Glu-Thr-Asp-fluoromethyl ketone (Z-IETD-FMK) that specifically blocks granzyme B activity. These results suggest that additional serine proteases as well as granzyme B-like are involved in cytotoxicity of CTL in fish. We further compared cytotoxicity with the granzyme B-like hydrolytic activity against fluorogenic substrate acetyl-Ile-Glu-Thr-Asp-4-methylcoumaryl-7-amide (Ac-IETD-MCA) and found that granzyme B-like activity correlated well with the cytotoxicity of CTL in ginbuna crucian carp. Present results suggest that the granzyme activity assays is useful to assess cytotoxic activity of CTL in fish in which genetic information on granzymes and specific tools for cytotoxicity assay are not available because of well conserved catalytic triad residues and substrate binding sites in granzyme B throughout vertebrates.  相似文献

1. The effects of nutritional level on muscle development, histochemical properties of myofibre and collagen architecture in the pectoralis muscle were evaluated using male broilers of Red Cornish x New Hampshire stock, reared on diets of high nutritional value for up to 80 d (H80d) and low nutritional value for up to 80 d (L80d, same age as H80d) or 95 d (L95d, same body weight as H80d). 2. The total live weight and the weight of pectoralis muscle were lower in L80d than in both H80d and L95d. The muscle weight as a percentage of live weight was 8.7% in L80d, 10.7% in H80d and 11.5% in L95d. 3. Pectoralis muscle was composed only of type IIB myofibres and showed no differences in myofibre type composition among the chicken groups. The largest diameter of type IIB myofibres was observed in L95d, followed by H80d and the smallest in L80d. 4. The total amount of intramuscular collagen did not differ among the chicken groups (1.92 to 1.99 mg/g). Types I and III collagens were immunohistochemically detected in both the perimysia and endomysia. The thin perimysia around the primary myofibre fascicles showed larger width in H80d than L80d and L95d, and also the thick perimysia around the secondary fascicles in H80d than L80d. 5. The collagen structure of the perimysium was most developed in H80d, followed by L95d and on the least in L80d. The development of perimysial collagen fibres could be enhanced by a rapid growth rate of the muscle induced by high nutritional level and depressed by a slow growth rate with low nutritional foods. 6. The endomysial collagen architecture was observed as a felt-like tissue of the fibril bundles with many slits. The thinnest endomysial wall was observed in L80d, followed by H80d and the thickest in L95d. 7. From these results, it was indicated that foods of high nutritional value could enhance growth of the pectoralis muscle of broilers, and this is accompanied by hypertrophy of the type IIB myofibres and development of the perimysial collagen architecture.  相似文献

Growing cloud droplets absorb such atmospheric gaseous pollutant as SO2(g), condensing atmospheric water vapor into themselves. Then, the cloud droplets are acidified by absorption of SO2(g) during condensational growth on cloud condensation nuclei (CCN). Characteristics of this process, which is a part of rainout, have not been made clear yet. In order to estimate the contribution of rainout to acid rain formation, the acidification of growing cloud droplets is investigated numerically, using a mathematical model. The numerical simulations show that: (1) the time to attain the equilibrium state for mass transfer (acidity and growth) and heat transfer (temperature) is much longer than the time for disappearance of CCN; (2) time variation of acidity and temperature of cloud droplets are greatly dependent on the existence of undissolved CCN; and (3) there seems to be a close correlation between the time variation of the acidity and that of the temperature.  相似文献

Exploring extent and pattern of non-timber forest product (NTFP) income contribution to livelihoods of forest-adjacent people and the income related socio-economic factors is crucial in designing any development and conservation initiative. This paper examines NTFPs' role in contributing to the household economy and how different socio-economic factors may influence this contribution. Field methods involved quantitative and qualitative data collection of forest-adjacent households of Chittagong Hill Tracts in Bangladesh. Results show that the contribution of different types of NTFPs to household economy varies in terms of subsistence and cash income. However, overall NTFP income is much higher than income from timber or firewood which indicates a larger dependence of the households on NTFPs. Income data analysis also finds that the poor households are relatively more dependent on NTFPs for their subsistence and cash income than the better-off households. Moreover, the study reveals that there are many other socio-economic factors at the household level such as number of members in household and total value of household implements and furniture that are significantly correlated with the NTFP income. More site-specific applied research on NTFP income and influential factors is necessary to generalize the findings for other parts of the world with similar socio-economic and environmental settings.  相似文献
