The plasmalemma of sporidia of Ustilago avenae has been investigated by means of the freezeetching technique after treatment with the fungicide, triadimenol (5 μg ml?1, 10 and 17 hr). The control samples are characterized in the exponential and stationary growth phases by homogeneous plasmatic fracture (PF) and extraplasmatic fracture (EF) faces with a random intramembrane particle (IMP) distribution. Treatment with triadimenol induces clusters of similar size IMPs in hexagonal arrangement. In between the flat clusters the plasmalemma is significantly deformed, showing hemispherical pits and protrusions on both membrane fracture halves. In the stationary growth phase no hexagonal clustering of the IMPs is observed under the described conditions.  相似文献

This study involved the comparison of suitability of different methods for routine diagnostics of Cryptosporidium spp. Two staining methods, one concentration-sedimentation method, seven concentration-floatation methods and one combined floatation-sedimentation method were compared. The methods were tested with two concentrations (1 x 10(5) and 1 x 10(6)/g) of C. parvum and C. andersoni. The methods were evaluated using light microscope, magnification 400x for concentration methods and 1000x for stained samples respectively. Specificity of both staining methods was 95-100%. Ziehl-Neelsen with P < 0.01 is more suitable for identification of C. andersoni and modified Milácek-Vítovec with P < 0.01 for identification of C. parvum. Concerning specificity and sensitivity, the floatation-concentration method by Sheather was found to provide the best results of all selected methods. The merthiolate iodine formaldehyde concentration (MIFC) method was the least specific one. The least suitable method concerning sensitivity and costs was the floatation method with caesium chloride (CsCl) with a specificity of 29%.  相似文献

Ultrastructural changes were observed in sporidia of Ustilago avenue (Pers.) Rostr. after treatment with the fungicides nuarimol and imazalil nitrate. Electron microscopic studies of ultrathin sections, as well as of freeze-fracture replicas, showed that nuarimol caused the following marked changes in the ultrastructure of the cells: increased cell wall thickness and invaginated plasmalemma, with concentrations of extracytoplasmic vesicles in some places between plasmalemma and the cell wall; pores were visible in the septa. Nuarimol-treated sporidia were characterised by vacuolisation and accumulation of lipid bodies. Compared with nuarimol, imazalil nitrate caused small cytological changes; elongation of the sporidia and enlarged cell diameters, as well as an increased thickness of the septa, were observed, and an enhanced number of mitochondria with reduced diameters were found.  相似文献

The effects of the systemic fungicides triadimefon, nuarimol, and imazalil nitrate (5 μg ml-1 for 8 and 16 h) on the ultrastructure of sporidia of Ustilago avenae were studied using transmission and scanning electron microscopes. Thin-section and freeze-fracture techniques demonstrated fine structural alterations to the cytology of the fungus. Compared with untreated controls, in which the cells were generally unicellular and uniform in shape and ultrastructure, fungicide-treated cells exhibited considerable thickening of peripheral cell walls and septa, an increased number of mitochondria that were enlarged and irregular in shape, greater amounts of endoplasmic reticulum, and extensive vacuolisation and accumulation of lipid bodies. The vacuoles were filled with membrane-like aggregates and vesicles, which could be extruded by exocytosis. The plasmalemma ultrastructure of fungicide-treated sporidia, observed by scanning electron microscopy after freeze-fracture was significantly different from that of control cells. The antifungal agents caused hemispherical pits, protrusions, invaginations, and aggregation of intramembrane particles on the membrane fracture faces. Specific structures, on the plasmalemma of plasmatic and extraplasmatic fracture faces, support the assumption of exocytosis. The ultrastructural changes were more pronounced after triadimefon and nuarimol treatment than after imazalil nitrate treatment. An extended incubation time (16 h) with triadimefon and nuarimol resulted in complete destruction of the cellular system of most sporidia.  相似文献

The uterine acetylcholinesterase and total cholinesterase (acetylcholinesterase plus butyrylcholinesterase) activities in normal and fenchlorphos treated blue fox vixens were determined during various reproductive states.AChE and Total-ChE of non-medicated vixens in oestrus were about one half of those in anoestrus. In pregnant uteri (luteal phase) the activities were 25 % and 30% compared to anoestrus.In vixens given 100 mg/kg fenchlorphos for 3 weeks during anoestrus, the remaining activity of AChE in uterus were in average 37%. Pregnant and non-pregnant vixens in the luteal phase medicated prior to mating and during time of implantation, displayed AChE activities which were only moderabely reduced (remaining activities 83% and 72% compared to medicated animals in anoestrus: remaining activity 37%).Plasma ChE-activity increased during pregnancy in the controls while enzyme activity was strongly reduced in animals given 100 mg/kg fenchlorphos daily through the whole pregnancy.It was concluded that the previous reported embryotoxic effect of fenchlorphos in the blue fox did not seem to be directed towards the moderate inhibition of the uterine cholinesterases.  相似文献

A novel insertion element belonging to the IS110 family was identified in Mycobacterium avium subsp. paratuberculosis. The IS element, ISMpa1, is 1500 bp and has one ORF encoding a putative transposase. Three copies of ISMpa1 were identified in the M. avium subsp. paratuberculosis genome. The element had inserted into the 3' end of the highly conserved mycobacterial genes prrB and a homologue of M. tuberculosis Rv1593c, and between a putative cytochrome p450 oxygenase and a putative hydrolase. The IS element was present in all (n = 11) M. avium subsp. paratuberculosis strains but not detected in most other mycobacterial species examined, including 10 M. avium subsp. avium isolates of human, avian and porcine origin. However two porcine isolates of M. avium subsp. avium and the reference strain IWGMT49 did harbour ISMpa1. These three strains belong to a previously described subgroup of M. avium subsp. avium based on IS1245 restriction fragment length polymorphism (RFLP) pattern and serovars. All of the M. avium subsp. paratuberculosis strains examined had an identical RFLP pattern when probed with sequences corresponding to the 5' end of ISMpa1, whereas a different pattern was seen in the positive M. avium subsp. avium strains. This novel IS element might be a useful tool in strain classification of M. avium subsp. avium and also for the identification of M. avium subsp. paratuberculosis when used in combination with IS900.  相似文献

Detection of Mycobacterium avium subsp. paratuberculosis (M. paratuberculosis) by polymerase chain reaction (PCR) is often hampered by the lack of efficient methods for sample treatment. We report a protocol for analysis of faecal samples based on buoyant density centrifugation in Percoll and IS900 sequence capture PCR combined with a dot blot assay for detection of low-grade infection of M. paratuberculosis. Serial dilutions of M. paratuberculosis genomic DNA and M. paratuberculosis bacteria were used to assess the sensitivity of the method. The final evaluation was performed with spiked faecal samples, which also were analysed by culture. The presence of PCR inhibitory substances in processed faecal samples was evaluated by including a PCR internal control. By using buoyant density centrifugation, sequence capture PCR, and dot blot hybridisation, we achieved a sensitivity of 10(3)CFU (colony forming units)/g of faeces. The detection limit by culture was assessed to 10(2)CFU/g of faeces. We conclude that the described protocol is a fast and sensitive alternative to bacterial culture of faecal samples.  相似文献

We describe the morphological, biological, and molecular characteristics of Cryptosporidium pig genotype II and propose the species name Cryptosporidium scrofarum n. sp. to reflect its prevalence in adult pigs worldwide. Oocysts of C. scrofarum are morphologically indistinguishable from C. parvum, measuring 4.81–5.96 μm (mean = 5.16) × 4.23–5.29 μm (mean = 4.83) with a length to width ratio of 1.07 ± 0.06 (n = 400). Oocysts of C. scrofarum obtained from a naturally infected pig were infectious for 8-week-old pigs but not 4-week-old pigs. The prepatent period in 8-week-old Cryptosporidium-naive pigs was 4–6 days and the patent period was longer than 30 days. The infection intensity of C. scrofarum in pigs was generally low, in the range 250–4000 oocysts per gram of feces. Infected pigs showed no clinical signs of cryptosporidiosis and no pathology was detected. Cryptosporidium scrofarum was not infectious for adult SCID mice, adult BALB/c mice, Mongolian gerbils (Meriones unguiculatus), southern multimammate mice (Mastomys coucha), yellow-necked mice (Apodemus flavicollis), or guinea pigs (Cavia porcellus). Phylogenetic analyses based on small subunit rRNA, actin, and heat shock protein 70 gene sequences revealed that C. scrofarum is genetically distinct from all known Cryptosporidium species.  相似文献

A study of ewe performance was undertaken in 795 sheep flocks based on data from the Norwegian Sheep Recording System. Herd level data included in the annual report for 1998 were used as the basis for the study. Average flock size was 75.3 ewes. The material was considered as representative of the better and more motivated sheep farmers in the mountain valley regions in south-eastern Norway. The average number of lambs born per ewe, inclusive (and exclusive) of barren ewes, was 1.89 (2.02), and of autumn lambs (those surviving from birth in the spring to autumn) 1.60 (1.66). The average autumn weight of the lambs corrected to 145 days of age was 42.4 kg, giving an overall performance of 67.9 kg lambs live weight per ewe. When including artificially reared lambs, overall performance was 70.4 kg per ewe. Artificial rearing of lambs, mostly triplets which had been removed from the ewe, was practised on 395 of the farms. In the 74 flocks with a rate of artificial rearing of more than 10% of all lambs, overall performance per ewe was 13.3 kg higher than in flocks with no artificial rearing. Total lamb mortality from birth to autumn recording, including stillborn lambs, was 12.44%. Mortality had a skewed distribution, some flocks having very high mortality figures. The arithmetic means (and medians) for mortality were as follows: 2.75 (2.29)% stillbirths, 2.93 (2.30)% died during the indoor feeding period, 0.88 (0.00)% at spring pasture, and 6.16 (4.40)% at summer pasture. In regression models using Proc Genmod in SAS®, it was found that average number of autumn lambs per ewe was negatively associated with the rate of barren ewes, late lambing date (flock average day in year when lambing took place), and total lamb mortality. The corrected autumn weight of the lambs was positively associated with the average number of lambs born per ewe and non-fertility rate, and negatively associated with flock size and total lamb mortality. The Dala breed had a positive effect on number of autumn lambs per ewe, and Spæl breed a negative effect on corrected autumn live weight of the lambs.  相似文献

Leguminous leys are important sources of nitrogen (N), especially in forage-based animal production and organic cropping. Models for estimating total N2 fixation of leys—including below-ground plant-derived N (BGN)—are based on grazed or harvested leys. However, green manure leys can have different proportions of above-ground plant-derived N (AGN) and BGN when subjected to different cutting regimes. To investigate the effects of cutting on N distribution in white clover, a pot experiment was carried out using 15N techniques to determine N2 fixation, N rhizodeposition and root C and N content of cut and uncut white clover (Trifolium repens L. cv. Ramona) plants. Percentage N derived from air (%Ndfa) was lower in uncut (63%) than in cut (72%) plants, but total Ndfa was not significantly affected by cutting. The higher reliance on N2 fixation in cut plants was thus counterbalanced by lower biomass and total N content. With BGN taken into account, total plant-derived N increased by approximately 50% compared with AGN only. Cutting did not affect the proportion of BGN to standing shoot biomass N after regrowth, but decreased the proportion of BGN to total shoot biomass production during the entire growth period. Thus, estimates of N fixation in green manure leys should consider management practices such as cutting regime, as this can result in differences in above- and below-ground proportions of plant-derived N.  相似文献
