Kernel vitreousness is an important grading characteristic for segregation of subclasses of hard red spring (HRS) wheat in the United States. This research investigated the protein molecular weight distribution (MWD) and the flour and baking quality characteristics of different HRS wheat market subclasses. The U.S. regional crop quality survey samples obtained from six regions for three consecutive growing years were used for subclass segregation based on the dark, hard, and vitreous (DHV) kernel percentage. Flour milled from HRS wheat with greater percentages of DHV kernel showed higher water absorption capacity for breadmaking. Protein MWD parameters could be related to the association between DHV kernel level and water absorption. Specifically, flour protein fractions rich in gliadins and high‐molecular‐weight polymeric proteins in the SDS‐unextractable fraction were identified to have significant and positive correlations with both DHV kernels and flour water absorption levels. An example further showed the importance of flour water absorption on potential economic incentives that can be gained with having a greater percentage of vitreous kernels. This information could help the flour milling and baking industry to segregate the different subclasses of HRS wheat with varying DHV content for their intended end‐use applications.  相似文献

This research aims to investigate the relationship between the solvent retention capacity (SRC) test and quality assessment of hard red spring (HRS) wheat flour samples obtained from 10 HRS cultivars grown at six locations in North Dakota. The SRC values were significantly (P < 0.05) correlated with flour chemical components (protein, gluten, starch, and damaged starch contents, except arabinoxylan); with farinograph parameters (stability [FST], water absorption, peak time [FPT], and quality number); and with breadmaking parameters (baking water absorption [BWA], bread loaf volume [BLV], and symmetry). Differences in locations and cultivars contributed significantly to variation in quality parameters and SRC values. Suitability of SRC parameters for discriminatory analysis of HRS wheat flour is greatly influenced by molecular weight distribution (MWD) of SDS‐unextractable proteins. SRC parameters, except for sucrose SRC, showed significant (P < 0.01) and positive correlations with high‐molecular‐weight (HMW) polymeric proteins in SDS‐unextractable fractions, whereas only lactic acid SRC exhibited significant (P < 0.01) correlations with low‐molecular‐weight polymeric proteins. HMW polymeric proteins also exhibited positive associations with FPT, FST, BWA, and BLV. The discrepant variation in association of SRC parameters with respect to MWD of SDS‐unextractable proteins could improve segregation of HRS wheat flour samples for quality.  相似文献

The aim of this study was to investigate the effects of feed withdrawal and darkening on the antioxidative status of laying hens in high ambient temperatures between 30-40 degrees C. We determined vitamins A and E, lipid peroxidation as thiobarbituric-acid reactive substances (TBARS), glutathione peroxidase (GSH-Px) and reduced glutathione (GSH) in the serum, liver and muscle. Sixteen week old hens (Ross Brown) were divided into 3 groups of 30 hens each. The first group was used as a control. Hens in the second (feed withdrawal) group were subjected to feed removal between 14-18 h during the summer. Hens in the third (darkening) group were subjected to darkening of the hens house with black curtains between 14-18 h. At the end of 16 weeks, blood serum, liver and breast muscle samples were taken from all animals and analysed. Vitamins A and E levels in serum and liver were significantly (P < 0.05, P < 0.01) higher in feed withdrawal and darkened groups than in control group whereas the serum levels of the darkening group were highest. GSH-Px activity and GSH level in liver and muscle were also significantly (P < 0.05, P < 0.01) higher in feed withdrawal and the darkening group than in control. However, TBARS levels in liver and muscle were significantly (P < 0.05, P < 0.01) lower in feed withdrawal and darkening groups compared with control group, whereas the level of the darkening group was the lowest. In conclusion, we observed that feed withdrawal and darkening of hens in high temperatures during summertime resulted in an increase of antioxidant enzymes and vitamins levels and in a decrease of lipid peroxidation.  相似文献

The splenic lobe (Lobus splenicus) of the pancreas of young meat-type quails (Coturnix c. japonica) was examined by immunohistochemical and light microscopic methods. The endocrine cells are mainly grouped as alpha, beta and mixed islets. A large region consisting of alpha cells is located in the central region of the splenic lobe whereas numerous beta islets are detected in the periphery of the splenic lobe. Alpha islets are in the majority composed of toluidine blue positive A cells and a few toluidine blue negative D and / or avian pancreatic polypeptide (APP) endocrine cells. Beta islets contain only a few toluidine blue negative B and a few D cells. Immunohistochemical staining of the splenic lobe reveal in the centre of beta islets numerous insulin immunoreactive cells and scarcely in alpha islets, exocrine tissue and / or among acinar cells. Somatostatin immune-reactive cells form a circular layer in the periphery of beta islets whereas these cells are uniformly distributed throughout the alpha islet parenchyma and exocrine tissue. In conclusion, the morphology but also the endo- and exocrine functions of the splenic lobe of quails are similar to observations in other avian species such as chicken, duck, goose and pigeon.  相似文献

Effects of genotype and environment on (1→3), (1→4)‐β‐d ‐glucan (β‐glucan) extractability, flour slurry viscosity, and β‐glucan polymer fine structure in oats were tested. One environment had a severe negative effect on slurry viscosity as evaluated with a rotational viscometer. Environment also had a strong effect on β‐glucan extractability, whereas genotype had no significant effect. Fine structure of β‐glucan was evaluated from the frequencies of oligosaccharides from lichenase hydrolysis of the β‐glucan polymer. Significant differences in degree of polymerization (DP) fragment frequencies were found associated with both genotype and growth environment. The high‐β‐glucan cultivar HiFi had lower DP3 fragment frequency and higher frequencies of DP4 and DP6 fragments than other cultivars with moderate β‐glucan concentration. Drier environments tended to yield lower DP3 fragment frequencies as well. Drier environments and genotypes with more β‐glucan synthetic potential may have provided cellular environments with more competition for substrate for β‐glucan synthesis, which appeared associated with lower DP3 fragment frequency. In a separate experiment, we found that extractable β‐glucan had higher frequencies of DP3 fragments and lower frequency of DP4 fragments. The observed variations deserve consideration for influence on functional properties, such as viscosity or health benefit potential.  相似文献

A 2‐year‐old, female Persian cat was presented with a history of distocia. In her first pregnancy, she had whelped four kittens and had eaten all of them right after parturition. She had mated again with the same tomcat. Well‐developed foetuses with weak foetal heart beats were observed in the ultrasonographic examination. En block ovariohysterectomy was performed. Three live and mature foetuses were obtained from the uterus; two of them were female foetuses and had no anatomical problem but the third one exhibiting multiple malformations was a male and diagnosed as ‘schistosoma reflexum’ (SR). The vertebral column deviated markedly to the right (scoliosis) at thoracolumbar region, and the middle lumbar and the sacral vertebrae were directed dorsocranially (lordosis). The entire small intestine, a part of large intestine, stomach, spleen and the right kidney were displayed out of the body, and it seemed that the listed internal organs were protruded from an abdominal cleft associated with the allantoic membrane. Liver, lungs and heart were hypoplastic. The large intestine was seen to have blind end (atresia recti), but anus was normal. Cerebrum and cerebellum were noticed as normal in sizes. Chromosome preparations from lymphocyte cultures of the foetus showed chromosomal aberrations including chromatid and chromosome breaks, exchange figures, non‐homologous pairing, whereas no abnormalities were detected in the chromosome preparations from mother’s cultures. This is probably the first case of SR in a cat, which was examined in detail from clinical, pathological, radiological and chromosomal angles.  相似文献

The prevalence of Dirofilaria immitis infection was evaluated in stray dogs of Erzurum, Turkey. A total of 123 whole-blood and 93 sera samples were collected from stray dogs older than 6 months were lived in animal shelter. The PCR and direct microscopic examinations were used for the detection of microfilaria and indirect-ELISA was performed for the detection of anti-D. immitis antibodies. The prevalence of D. immitis in the canine population was 8.1% by PCR, 2.1% by ELISA. In addition, microfilaria burdens of Dirofilaria sp. was 4.8% by direct blood smear examination. There was a statistical difference (P=0.05) in the prevalence between males (10.5%) and females (2.3%) by direct blood smear examination. Similarly there was a statistical difference (P<0.05) in the prevalence between males (15.8%) and females (4.7%) by PCR. Dogs belonging to the 0.5-1 years old group showed the highest prevalence than 2-4 ages group with three tests. Among the 93 samples screened by the ELISA, two samples were positive for the D. immitis antibodies. Both positive dogs with ELISA were females.  相似文献

Arabinoxylans (AX), xylanase, and xylanase inhibitors have an important role in many cereal food processing applications. The effects of genotype, growing location, and their interaction (G × L) on AX, apparent xylanase activity, and apparent xylanase inhibition activity of Triticum aestivum xylanase inhibitor (TAXI) and xylanase inhibiting protein (XIP) were investigated for six hard red and six hard white spring wheat genotypes grown at three locations. Difference in total AX level among genotypes was not determined to a significant level by genotype. Instead, variability in total AX content was largely dependent on G × L. However, total AX content was significantly different between the two wheat classes. For bran xylanase activity, 25% of the variability could be attributed to G × L interaction. Moreover, there was significant difference between the bran xylanase activities in the two wheat classes. Bran TAXI activity and XIP activity were significantly different among genotypes. Genotype contributed 72% to the variability in TAXI activity and 39% in XIP. However, no significant difference was observed among the two wheat classes for TAXI or XIP activity. These results indicate that TAXI might be a stable parameter in segregating wheat genotypes with varying xylanase activity.  相似文献
