Adrenomedullin (AM) has been characterized as an endogenous tissue survival factor and modulator of many inflammatory processes. Because of the increased susceptibility of the mammary gland to infection during the time surrounding parturition in the cow, we investigated how milk and tissue content of AM and its binding protein (AM-BP) might be affected by the stage of lactation and the udder health status. Milk and mammary biopsy samples were obtained from Holstein cows 21 days prior to and at various times after calving to represent the dry period and early and mid-stages of lactation. Additional cows received an intramammary challenge with Escherichia coli for immunohistochemical characterization of AM and AM-BP. Milk AM concentrations were relatively constant across the stages of lactation while AM-BP increased two-fold (P<0.04) between early and mid-lactation. Milk AM (P<0.04) and AM-BP (P<0.03) increased as somatic cell counts (SCCs) increased within a given stage of lactation. Tissue content of both (AM and AM-BP) were significantly affected by stage of lactation, lowest in the dry period and progressively increasing to peak at mid-lactation as well as increasing in association with higher levels of SCCs. Following E. coli challenge, AM increased in epithelial cells surrounding mammary alveoli presenting high levels of SCCs. The data suggest that AM and AM-BP are cooperatively regulated in the mammary gland during lactation; changes in localized tissue AM and AM-BP content reflect a dynamic regulation of these tissue factors in the bovine mammary gland consistent with their protective effects within inflamed tissue.  相似文献

The sequential extraction procedure proposed by the European Commission Measurement and Testing Programme, combined with Scanning Electron Microscopy and Energy Dispersive X-ray Analysis(SEM/EDS), was applied to identify and quantify the chemical andmineralogical forms of Cu, Ni, Fe, Mn, Zn, Pb, Cr and Cd presentin the topsoil from a mining and smelting area near Sudbury (Ontario, Canada). The possible mobility of the chemical forms was also assessed. The metal fractions: (1) soluble and exchangeable, (2) occluded in manganese oxides and in easily reducible iron oxides, (3) organically bound and in form of sulphides, (4) residual mainly present in the mineral lattice structures were separated. Cu and Ni were the major metallic contaminants, occurring in soils in broad ranges of concentrations: Cu 11–1890 and Ni 23–2150 mg kg-1. Cu was uniformly distributed among allthe extracted fractions. Ni was found associated mainly withthe residual forms, accounting for 17–92%, with an averageof 64%, of the total Ni present in the soils. Fe, Mn, Zn,Pb, Cr and Cd, while occurring in most analysed samples innormal soil concentrations, were primarily held in theresidual mineral fraction (on average >50%). The solubleand exchangeable forms made a small contribution (≤8.1%)to the total content of metals extracted. At least 14% ofthe total Cd, Mn and Pb was mobilised from the reducibleforms. The oxidizable fraction assumed mean values higher than10% only for Pb and Zn. Statistical treatment of the experimental data showed significant correlations between totalmetal content of the soils, some soil properties such as pH value, clay and organic matter content, and metal concentrationsin the various fractions. SEM/EDS analysis showed Fe in form ofoxides and sulphides in soils and Cu, Ni, Mn, Zn and Cr in association with iron oxides. Numerous black carbonaceous particles and precipitates of aluminium fluoride salts, observedin the solid residue left after `total’ digestion, were found tocontain Fe, Ni and Cr.  相似文献

The effect of increasing concentrations of Cd and Zn in a sandy soil on spring wheat (Triticum vulgare L.) yields and the metal contents of the plants was examined in a pot experiment to establish critical levels of these metals in soil. The metals were added (individually and jointly) to the soil as sulfates in the following doses (in μg g?1, dry wt.): Cd — 2, 3, 5,10, 15, 25, and 50; Zn ?200, 300, 500, 1000, 1500, 2500, and 5000. Cadmium added to soil did not affect yields of wheat. The Zn dose of 1000 μg g?1 strongly reduced crop yields; at 1500 μg g? Zn dose wheat did not produce grain. The metal contents of wheat increased with increasing concentrations of Cd and Zn in soil up to 10.3 and 1587 μ g? of Cd and Zn in straw, respectively. The concentrations of both metals were higher in straw than in grain by factors of 3–7 and 1.5–2 for Zn and Cd, respectively. The relationships between Cd and Zn contents of the plants and soils were best expressed by exponential equations. High concentrations of Zn in soils (1042 and 1542 μg g?1) enhanced uptake of Cd by plants. The tested threshold concentrations of the metals in soils (3 μg g?1 for Cd and 200–300 μg g?1 for Zn) are safe for Zn but are too high for Cd in terms of protecting plants from excessive metal uptake. The critical Cd content of sandy soil should not exceed 1.5 μg g?.  相似文献

Weight, heartgirth, backfat and body condition of sows was monitored in a commercial, farrow-to-finish unit during 1 yr. Measurements were obtained during the third, ninth and fifteenth week of gestation and the day after weaning. Litter performance and rebreeding rate also were recorded. Body weight and heartgirth increased (P less than .01) over parities because gestational gains were larger than lactational losses. Backfat tended to be lower in later parities. Body weight, heartgirth and backfat, but not condition score, declined from weaning to the third week of the subsequent gestation in both first and second litter sows. Correlations among measures of body condition were low (less than .45), except the overall correlation between body weight and heartgirth. Number of pigs born alive increased and interval to estrus decreased in later parities. There were no significant relationships between changes in body condition and rebreeding performance of sows. These results suggest that changes in body condition typically observed in sows housed in commercial production units may be too subtle to have an effect on reproductive performance.  相似文献

The causes of lameness in dogs were analyzed on the basis of radiologic examinations performed in the years 1980–1985. From 1982, there was an increase in the frequency of secondary nutritional hyperparathyroidism and eosinophilic panosteitis and a decrease in frequency of osteochondrosis. The pathogenesis of these diseases is discussed in relation to alternations in diet during this time.  相似文献

In pigs, plasma prolactin concentration markedly changes during the oestrous cycle and the regulation of its secretion is very complex. The contribution of neurokinins in this process has not been sufficiently delineated. The aim of the study was to examine the effects of neurokinin A (NKA) on prolactin synthesis and secretion in cyclic gilts. The expression of NKA precursor (Ppta) and receptor (Tacr2) genes as well as NKA and TACR proteins content in the porcine pituitaries (days 2–3, 9–10, 12–13, 15–16 and 19–20 of the cycle) was determined. Furthermore, the in vitro influence of NKA on the expression of prolactin (Prl), dopamine receptor (D2r), TRH receptor (Trhr) genes and prolactin secretion by the porcine pituitary cells (days 9–10, 15–16 and 19–20 of the cycle) was assessed. The expression of Ppta and Tacr2 as well as NKA and TACR proteins in the pituitary tissue has been changing throughout the oestrous cycle. NKA affected in vitro the expression of studied genes and prolactin secretion depending on the stage of the cycle, dose of NKA and/or duration of the cell incubation. Altogether, the study indicates that NKA is engaged in the modulation of prolactin secretion in the pig during the oestrous cycle.  相似文献

ABSTRACT

The potential of keratin wastes originating from poultry farms in practical application as a valuable organic fertilizer, gives rise to the need for intensive study on their effect on plants. In this study, for the first time there has been examined the influence of hen feather keratin bio-hydrolysate (FKH) and hen feather keratin compost (FKC) on plant growth, and the following features that indicate the plant condition: the leaf chlorophyll content, the activity of phenylalanine ammonia lyase (PAL) and guaiacol peroxidase (GPX) enzymes. The results of pot experiments showed a potential plant growth promoting effect of FKH, applied as a leaf treatment and of FKC manure. Fertilization of tested plants with FKC resulted in significant increase of their fresh weight. There was no effect on plant biomass after FKH treatment. The stimulating effect on plant physiology was expressed by decreased PAL activity after FKC treatment, and enhanced GPX activity, after FKH and FKC treatment. The application of FKC as a manure gave better effects for the plant condition expressed by the activity of PAL and GPX in comparison with FKH spraying. The chlorophyll content did not prove to be an efficient parameter to evaluate the impact of FKH or FKC on white cabbage, tomato, and maize plant. However, a significant increase in the leaf chlorophyll a, b, and a+b concentration was observed in cucumber plant after FKC treatment. Among the tested plants, the cucumber has shown the most profitable effect of feather compost on plant growth.  相似文献

The length of dormancy and sprouting characteristics of ten cultivars (Bison, Kennebec, Norchip, Norgold Russet, Norland, Red Pontiac, Russet Burbank, Viking, Dakchip, and ND8891-3), were compared. Russet Burbank had the longest dormancy period and the slowest rate of sprouting after dormancy ended. Viking and Norgold Russet had the most sprouts and Kennebec the fewest. Dakchip had the shortest dormancy period, the greatest weight of sprouts and the greatest weight loss. Dormancy was shortest in the warmest (20°C) storage. Storage at 2°C for six to nine weeks increased sprouting after dormancy ended compared with continuous 10 or 20°C storage.  相似文献

BACKGROUND: Cycloate inhibits the biosynthesis of very‐long‐chain fatty acids, the essential constituents of plant waxes and suberin. Fatty acids also serve as precursors of aliphatic carbon chains in resorcinolic lipids, which play a fundamental role in the plant defence system against fungal pathogens. In this study, the effect of cycloate on the biosynthesis of 5‐n‐alkylresorcinols in rye seedlings (Secale cereale L.) grown under various light and thermal conditions was examined. RESULTS: The content of alkylresorcinols biosynthesised in rye was generally increased by the herbicide in both green and etiolated plants. The presence of cycloate also affected patterns of alkylresorcinol homologues in plants grown at 15 and 22 °C; very‐long‐side‐chain compounds were less abundant, whereas both short‐chain saturated and unsaturated homologues were generally accumulated. No cycloate‐related effects caused by homologue pattern modifications were observed at elevated temperature. CONCLUSION: This study extends present understanding of the mode of action of thiocarbamate herbicides. Cycloate markedly affected the biosynthesis of very‐long‐side‐chain resorcinolic lipids in rye seedlings, confirming the existence of parallels in both fatty acid and alkylresorcinol biosynthetic pathways. The observed cycloate‐driven accumulation of 5‐n‐alkylresorcinols may improve the resistance of cereals to infections caused by microbial pathogens.
