For the selection of donors with valuable characteristics for breeding, 39 thyme accessions were evaluated in three years
according to a staggered schedule. The criteria investigated were: winter hardiness, beginning of flowering, growth height,
yield of the dry herb, content of essential oil, composition of the essential oil, DNA content of cell nuclei and number of
chromosomes. The most strongly varying traits between the populations were the yield of dry herb, the content of essential
oil and the content of volatile phenols with coefficients of variation (CV) between CV 40% and 50%. The largest variation
within a population was detected for the yield of dry herb (CV 25–46%) and the content of essential oil (CV 17–48%). The homogeneity
of the populations was different. The minimal average coefficient of variation of all traits (CV 19%) was determined in the
population of the cultivar ‘Varico II’ and in a population from Lithuania. The ploidy level of T. vulgaris was diploid (2n = 30). 相似文献

Immunoblotting assays using commercial antibodies were established to investigate the unexpected persistence of the immunoactive Cry1Ab protein in the bovine gastrointestinal tract (GIT) previously suggested by enzyme-linked immunosorbent assay (ELISA). Samples of two different feeding experiments in cattle were analyzed with both ELISA and immunoblotting methods. Whereas results obtained by ELISA suggested that the concentration of the Cry1Ab protein increased during the GIT passage, the immunoblotting assays revealed a significant degradation of the protein in the bovine GIT. Samples showing a positive signal in the ELISA consisted of fragmented Cry1Ab protein of approximately 17 and 34 kDa size. Two independent sets of gastrointestinal samples revealed the apparent discrepancy between the results obtained by ELISA and immunoblotting, suggesting that the antibody used in the ELISA reacts with fragmented yet immunoactive epitopes of the Cry1Ab protein. It was concluded that Cry1Ab protein is degraded during digestion in cattle. To avoid misinterpretation, samples tested positive for Cry1Ab protein by ELISA should be reassessed by another technique. 相似文献

The present study was conducted to study the impact of the virulence factors invC and sseD of the two type III secretion systems of Salmonella enterica serovar Typhimurium (S. Typhimurium) on the pathogenesis of the porcine S. Typhimurium DT104 infection. For this purpose, two S. Typhimurium mutant strains with a disrupted invC gene of the Salmonella pathogenicity island 1 or with a disrupted sseD gene of the Salmonella pathogenicity island 2 have been studied in experimental infection of pigs. Twenty-two 7-week-old male hybrid pigs were either infected with one of the mutants or the wild-type S. Typhimurium DT104 strain. Each group was examined for clinical signs, Salmonella shedding rate and the specific antibody response. Survival and replication were evaluated by qualitative and quantitative determination of the colonization rate. The humoral and cellular immune responses were examined using isotype-specific ELISA and quantitative real-time PCR of IL-2, IL-4, IL-10, IL-12 and IFN-gamma. The results proved that both mutants had a lower virulence (with marked differences between both mutants) than the wild-type and that both virulence factors have importance in porcine salmonellosis. Only pigs infected with the wild-type S. Typhimurium DT104 exhibited typical clinical symptoms of salmonellosis like anorexia, vomiting, disturbed demeanour, fever and diarrhoea. Deletion of the invC gene resulted in a significantly reduced colonization rate. Interestingly, the mRNA expression of both type-1 and type-2 cytokines were significantly decreased in pigs infected with either the invC-mutant and the sseD-mutant strain. 相似文献

The estimation model PhytoCalc allows a non-destructive quantification of dry weight and nutrient pools of understorey plants
in forests by using the relationship between species biomass, cover and mean shoot length. The model has been validated with
independent samples in several German forest types and can be a useful tool in forest monitoring. However, in open areas within
forests (e.g. clearcuts), the current model version underestimates biomass and produces unreliable nutrient pool estimations.
Thus, tissue density, as approximated by leaf dry matter content (LDMC), is systematically higher under high light compared
to low light conditions. We demonstrate that the ratio of LDMC under clearcut conditions to LDMC under forest conditions can
be used to adjust the PhytoCalc model to clearcut conditions. We investigated the LDMC ratio of five exemplary species commonly
occurring on clearcuts. Integrating the square of the ratio as a correction factor improved estimates of biomass to more than
70% fit between observations and predictions. Results also suggest this ratio can be used to correct nutrient concentrations
modelled in PhytoCalc, which tend to be overestimated in clearcuts. As morphological groups of plant species exhibit significantly
different ratios, we advise using group-specific correction factors for clearcut adjustments in the future. 相似文献

Coatings of biomaterials or implants that facilitate biomineralization possess a great potential for applications focused to the replacement, augmentation, and regeneration of bone tissue. Biomimetic approaches utilize biomolecules for either templating or supporting the crystallization process. One of these promising biomolecules is phosvitin (PV), an egg yolk protein known to transport and store inorganic phosphates and calcium ions. The incorporation of PV into polyelectrolyte multilayers is favorable due to PVs high degree of phosphorylation and thus a high acidity. Utilizing the reflectometric interference spectroscopy, the adsorption kinetics of this novel polyelectrolyte system composed of poly-L-lysine and the heavily phosphorylated phosvitin were monitored. The results demonstrate an unexpected nonregular growth regime called overshoot. Effective measures of shifting this irregular polyelectrolyte adsorption process back to a regular multilayer growth regime are reported in this paper. 相似文献

The conversion of even-aged Norway spruce (Picea abies (L.) H. Karst.) stands into more site adapted mixed stands is the main silvicultural goal in Germany and many other European countries. The conversion will primarily be achieved with the help of selection cutting, removing single target diameter trees and creating small gaps. At sites highly susceptible to windthrow, however, small scale clear cutting (up to 1 ha) might be an appropriate alternative. Both selection and clear cutting will affect the understorey vegetation, an important component of forest biodiversity and ecosystem functioning. Thus, with the help of a BACI-experiment (before–after/control–impact), the short-term effects of selection and small scale clear cutting on the herb and shrub layer vegetation were studied at two sites in the Solling Hills (Lower Saxony, Germany). Both treatments led to an increase in herb and shrub layer species numbers, due to persistent and invading (mainly pioneer tree species and indicators of disturbance) species and to a shift in plant traits. Most responsible for the changes in vegetation was a higher light availability, and to a lesser degree soil disturbance. The number of annuals and ruderals, however, increased only immediately after clear cutting, whereas on selection cutting plots the numbers remained high. Gaining in number and coverage were competitive perennials (Epilobium angustifolium, Juncus effusus, Rubus idaeus) especially on the clear cuttings, fulfilling a nutrient retention function. The competitives did not deter tree species from regenerating in the clear cut plots, whereas on selection cutting plots the large expansion of R. idaeus appeared to be hindering tree colonisation. Suffering under clear cut conditions were predominantly shade-tolerant, closed-forest species, but no loss of these species could be detected. A stilbene synthase gene along with the selectable marker gene bar for herbicide resistance was transferred via Agrobacterium tumefaciens mediated transformation into apple (Malus domesticaBorkh.) cvs. 'Elstar' and 'Holsteiner Cox'. The stilbene synthase catalyzes the conversion of 1 molecule of p-coumaroyl-CoA and 3 molecules of malonyl-CoA into 3,4',5-trihydroxystilbene, commonly known as resveratrol. This phytoalexin has implications in both phytopathology and human health. Greenhouse-grown transgenic and nontransformed control plants were grafted onto dwarfing rootstock M27. Flowering and fruiting occurred within the following years, offering the opportunity to analyze transgenic apple fruit and fertility of transgenic plants as well as inheritance of the transgenes into the seedling progeny. Molecular analysis revealed that the stilbene synthase is expressed in transgenic plants and in the skin and flesh of transgenic apple fruit. After formation, resveratrol is modified by the addition of a hexose sugar. The resulting component was characterized as piceid. With the aim of characterizing the influence of the novel biosynthetic pathway on the accumulation of other phenolic compounds naturally present in apple fruit, the amounts of flavanols, flavonols, phloretin derivatives and hydroxycinnamic acids in wild type and transgenic fruit were determined by HPLC. In all investigated transformed lines that accumulated piceid, no negative correlation between levels of piceid and the above-mentioned compounds was observed, except for the flavonol contents, which slightly decreased. Inheritance of the transgenes was confirmed in the seedling progeny, which were obtained after pollination of transgenic plants with nontransgenic pollen and vice versa after pollination of nontransgenic plants with pollen obtained from transgenic plants. The fertility of stilbene synthase transgenic plants was demonstrated. To the authors' knowledge this is the first time that data are available on piceid synthesis in transgenic apple fruit and the effects of its accumulation on levels of other phenolic compounds present in the fruit. 相似文献

Milk production in Malawi is still unsatisfactory despite efforts by different stakeholders to boost the dairy sector. To
investigate the roots of the problem, a survey on the current production practices and constraints on smallholder dairy farming
was conducted in the Northern Region of the country. A total of 210 farmers were interviewed. The results revealed that farmers
had small herd sizes in the region with an average of 2.2 ± 0.6 cattle per farmer. Average herd size was larger in male-managed
farms than in female-managed farms (2.6 ± 2.8 vs. 1.8 ± 1.3), farmers with more than 5 years of dairy farming experience had
larger herds than those with less experience (2.6 ± 2.8 vs. 1.9 ± 1. 2) and farmers who grazed their animals tended to have
larger herds than those that stall-fed their animals (4.4 ± 5.1 vs. 1.9 ± 1.3). Average milk production was 8.2 ± 6.5 l per
cow per day. Higher average daily milk production was observed in farmers with above primary school education (10.3 ± 8.3
vs. 7.7 ± 5.6), those with dairy farming as main activity (9.3 ± 6.6 vs. 6. 5 ± 6.1) and farmers with more than 2 years of
experience in dairy farming (9.3 ± 6.3 vs. 6.1 ± 6.4). Unreliable supply of improved animal genetics, poor animal health,
feed shortage and poor prices for milk were considered to be the most important constraints to smallholder dairy farming in

Authenticity assessment of trans-anethole is deduced from (2)H/(1)H and (13)C/(12)C isotope ratios, determined by gas chromatography-isotope ratio mass spectrometry (GC-IRMS). For that purpose, self-prepared anise and fennel oils, and synthetic and "natural" samples of trans-anethole, as well as commercially available anise and fennel oils have been investigated. Authenticity ranges of (2)H/(1)H and (13)C/(12)C isotope ratios of trans-anethole were defined. Scope and limitations of the applied online GC-IRMS techniques are discussed. 相似文献

Consistent evidence suggests that the probable human carcinogen acrylamide is formed in starch-rich foodstuffs through heat-induced interaction of asparagine and reducing sugars during Maillard browning. However, information regarding the influence of processing parameters on acrylamide formation is scarce. We investigated the impact of temperature, heating time, browning level, and surface-to-volume ratio (SVR) on acrylamide generation in fried potatoes. Acrylamide content was determined by liquid chromatography (LC) and electrospray ionization tandem mass spectrometry (ESI-MS/MS). In potato shapes with low SVR, acrylamide content consistently increased with increasing temperature and processing times. By contrast, in shapes with intermediate to high SVR, maximal acrylamide formation occurred at 160-180 degrees C, while higher temperatures or prolonged processing times caused a decrease of acrylamide levels. Moreover, browning levels were not a reliable measure of acrylamide content in large-surface products. 相似文献
