The efficacy of Leishmania donovani-derived lipophosphoglycan (LPG) plus Mycobacterium bovis Bacille Calmette-Guerin (BCG) as a vaccine candidate against visceral leishmaniosis in susceptible BALB/c mouse and Syrian golden hamster (Mesocricetus auratus) models was investigated. Following a triple vaccination with a total dose of 150 microl BCG plus 60 microg or 30 microg of LPG for hamsters and BALB/c mice respectively, there were no noticeable side effects both locally and systemically; implying that the molecule was safe at this dosage level. Vaccinated animals demonstrated an activation of both the humoral as well as cell-mediated responses to LPG, which correlated with resistance against the disease. Protection by LPG plus BCG, was however, poor as the remaining immunized animals showed disease progression leading to severity of the disease as illustrated by emaciation, mass loss and heavy splenic parasitaemia in hamsters. These data nevertheless suggest that it may be rewarding to further evaluate the potential of LPG as a vaccine candidate in leishmaniosis using other adjuvants, which may enhance its immunogenicity.  相似文献

The solubilization site of two betalain pigments, namely, betanin and indicaxantin, into l-alpha-dipalmitoyl-phosphatidylcholine (DPPC) and dimyristoylphosphatidylcholine (DMPC) vesicles was investigated by a spectrophotometric study. Pigment absorbance was monitored by varying phospholipid concentration, at a constant temperature that was varied in a range including the main phase transition temperature (Tm) of the relevant phospholipid bilayer. Maximum betanin absorption increased with the increase of DPPC concentration within the entire temperature range, reaching a plateau. The binding constant (Kb) of the pigment, calculated according to a pseudo-two-phase model, varied with the temperature, indicating that betanin is located at the hydrophobic interior of the bilayer. Other measurements of binding of betanin to DMPC and of indicaxanthin to either DPPC or DMPC vesicles ruled out that these compounds were solubilized in the hydrophobic interior of these bilayers.  相似文献

Understanding the dissipation rates of chemicals in unsaturated and saturated zones of subsurface soils will help determine if reductions of concentrations to acceptable levels will occur. Chemical properties and microbial biomass and activity were determined for the surface (0-15 cm), lower root (50-105 cm), and vadose (175-220 cm) zones in a Huntington silty clay loam (Fluventic Hapludoll) collected from an agricultural field near Piketon, OH. The rates of sorption, mineralization, and transformation (formation of bound residues and metabolites) of atrazine were determined. Microbial activity was estimated from the mineralization of (14)C-benzoate. We observed decreased levels of nutrients (total organic carbon, N, and P) and microbial biomass with depth, while activity as measured with benzoate metabolism was higher in the vadose zone than in either the surface or the root zones. Sorption coefficients (K(f)) declined from 8.17 in the surface to 3.31 in the vadose zone. Sorption was positively correlated with organic C content. Rates of atrazine mineralization and bound residues formation were, respectively, 12-2.3-fold lower in the vadose than in the surface soil. Estimated half-lives of atrazine ranged from 77 to 101 days in the surface soil, but increased to over 900 days in the subsurface soils. The decreased dissipation of atrazine with increasing depth in the profile is the result of decreased microbial activity toward atrazine, measured either as total biomass or as populations of atrazine-degrading microorganisms. The combination of reduced dissipation and low sorption indicates that there is potential for atrazine movement in the subsurface soils.  相似文献

A critical step in the infectious cycle of Leishmania is the differentiation of parasites within the sand fly vector to the highly infective metacyclic promastigote stage. Here, we establish tetrahydrobiopterin (H4B) levels as an important factor controlling the extent of metacyclogenesis. H4B levels decline substantially during normal development, and genetic or nutritional manipulations showed that low H4B caused elevated metacyclogenesis. Mutants lacking pteridine reductase 1 (PTR1) had low levels of H4B, remained infectious to mice, and induced larger cutaneous lesions (hypervirulence). Thus, the control of pteridine metabolism has relevance to the mechanism of Leishmania differentiation and the limitation of virulence during evolution.  相似文献

The aim of this work was to determine the selenium content in nut samples by cathodic stripping potentiometry. Dry-powdered nuts were digested by HNO(3) and dissolved with concentrated hydrochloric acid. To avoid the interference of natural oxygen, the potentiometric determination of selenium was carried out in an electrolyte solution consisting of 2 M CaCl(2) and 4 M HCl. The analysis was executed applying an electrolysis potential of -150 mV for 60 s and a constant current of -30 microA. Under these conditions, detection limits lower than 1.0 ng g(-)(1) were obtained for selenium analysis in nuts. The relative standard deviation of these measurements (expressed as rsd %) ranged from 0.44 to 0.88% while recoveries ranged from 90.2 to 95.3%. The results obtained with the proposed method were compared with those obtained via hydride vapor generation atomic absorption spectroscopy, a common method for determining selenium. The results of the two methods agreed within 5% for almond, hazelnut, and pistachio samples. The mean concentrations of selenium determined in Sicilian samples of almond, hazelnut, and pistachio were 531 +/- 1, 865 +/- 1, and 893 +/- 4 microg/kg, respectively.  相似文献

Microbial degradation of fungicides on leaf surfaces after repeated applications to turfgrass was investigated. Prior andcurrent work in our laboratory has identified two characteristicsof the turfgrass leaf system that may contribute to the enhanceddegradation of fungicides after repeated application to turfgrass:(1) The leaf surface is rich in microorganisms (~108 g-1 dr wt leaf), and (2) Leaf surface microorganisms may respond to repeated fungicide applications in a manner consistentwith the phenomena of enhanced biodegradation. Field studies wereconducted on ‘Penncross' creeping bentgrass with four fungicidesrepresenting three chemical families applied either two or eight times in one growing season. Biodegradation was estimated using data from both a field study and a parallel laboratory study thatfollowed the fate of 14C-labelled fungicides. For the laboratory incubations, the locations of the residual 14C fungicides were estimated using a sequential extraction protocolthat fractionated the materials into three pools: available,retained and bound. Data from both the field and laboratory studyrefuted our hypothesis that enhanced biodegradation would developfollowing repeated applications of the fungicides onto the leafsurface. Our studies support a conclusion that a two-stagephysical sorption process leads to plant incorporation and thiscontrols most of the fungicide's fate. Thus, our data suggestthat microbial activity plays a less important part in theprocess than would be indicated by considering the size of themicrobial population on the leaves.  相似文献

An improved understanding of factors that influence the survival and/or growth of Escherichia coli (E. coli) in soil is essential to allow the formation of land management practices to control the spread of the pathogenic strains of the bacteria, whose transmission to fresh produce is a threat to food safety. Persistence of E. coli in soils held at different water potentials and with carbon additions then subjected to post-freezing incubation temperatures and in the presence of Klebsiella terrigena (K. terrigena) were investigated. Soil samples adjusted to different water potentials (?0.03, ?0.1 and ?1.5 MPa) were inoculated with a multi-antibiotic resistant strain of E. coli (E. coli 2+), which allowed recovery of the organism from soil samples. In addition to manipulation of water content, different C levels were added and samples were frozen for varying lengths of time, thawed and incubated. In freezing studies, initial soil moisture content significantly affected E. coil 2+ survival in soils following thawing, resulting in lower survival rate (k) at water potential of ?0.03 than at ?0.1 and ?1.5 MPa. The effect of length of freezing time was significant only at ?0.03 MPa. Glucose addition at 1.25 mg C g?1 improved survival rate versus glucose at 0.125. The low level glucose increased die-off rate versus no addition, suggesting that unless amendments provide C above a certain threshold level, they might facilitate the death of the bacteria. E. coli 2+ survival improved in the presence of K. terrigena at 6°C but not at 23°C. Persistence of E. coli under the interactive influence of various environmental factors highlights the urgency and importance of understanding its potential for transmission to fresh produce and water bodies.  相似文献

Changes in physiological parameters that are induced by acute exercise on a treadmill in healthy military dogs have not been thoroughly investigated, especially with regard to age. This study investigated the effects of acute exercise on a treadmill on cardiovascular function, biochemical parameters and gastric antral motility in military dogs. Thermography was used to assess variations in superficial hindlimb muscle temperature. Nine healthy dogs were distributed into three groups according to their age (Group I: 25 ± 7 months; Group II: 51 ± 12 months; Group III: 95 ± 10 months) and sequentially subjected to running exercise on a treadmill for 12 min (3.2 km/h at 0° incline for 4 min, 6.4 km/h at 0° incline for 4 min and 6.4 km/h at 10° incline for 4 min). Heart rate, systolic and diastolic arterial pressure (DAP), gastric motility, haematocrit and biochemical analyses were performed at rest and after each session of treadmill exercise. Infrared thermographic images of muscles in the pelvic member were taken. Exercise decreased DAP in Group I, increased systolic arterial pressure in Groups II and III and increased mean arterial pressure in Group III (all p < 0.05). After the exercise protocol, plasma creatine kinase and aspartate aminotransferase levels increased only in Group I (p < 0.05). Exercise increased heart rate and decreased the gastric motility of a solid meal at 180 min in all groups (all p < 0.05). Exercise also elevated temperature in the femoral biceps muscles in Group I compared with the older dogs. The results indicate that acute exercise decreased gastric motility in dogs, regardless of age, and caused more pronounced cardiovascular changes in older dogs than in younger dogs. Acute exercise also altered biochemical parameters and superficial hindlimb muscle temperature in younger military dogs.  相似文献
