Field performance and molecular diversification of lemon selections   总被引:1,自引:1,他引:0  
Lemon (Citrus limon (L.) Burm. f.) is one of the most important Citrus fruit for Turkey because of its great amount of production and export. It has been cultivated for a long time in Turkey, and therefore variations for agronomical traits are likely among cultivated lemons due to bud mutations and, hybridizations. The objectives of this study were to determine variations for some selected agronomical traits and genetic markers among 12 new lemons derived from selections. Tree growth, yield, fruit quality, and molecular diversification of these clones were determined. After four years of evaluation, ‘Kutdiken’ M-51 indicated the highest canopy volume. For yield per tree, the best clone was ‘Kutdiken’ M-51. After five years of evaluation, ‘Kibris’ M-54 had the highest fruit weight and acidity. ‘Italian Memeli’ M-56 contained the lowest seed number and the highest total soluble solids. Molecular analysis, as assessed with 22 random amplification of polymorphic DNA (RAPD) and 11 inter simple sequence repeats (ISSR) primers, indicated that seven of twelve clones were separated with RAPD markers, whereas four were distinguished with ISSR markers. Combined analysis of RAPD and ISSR data detected that similarity values among the lemons clones were between 0.97 and 1.00. It can be concluded that variations in orchards are abundant and mainly due to mutations.  相似文献

Many stress-related interactions such as pathogen infection, insect tolerance, salt tolerance, auxin degradation, cell wall lignification, tissue suberization, and plant senescence involve various isoforms of peroxidases. Peroxidase gene polymorphism (POGP) markers have been used to estimate diversity, relationships and population structure among 80 Citrus and their relatives in Aurantioideae by using unweighted pair group method arithmetic average (UPGMA) and bayesian substructuring analysis. Fourteen primers produced a total of 148 fragments and 147 of them were polymorphic. The UPGMA analysis demonstrated that the accessions had a similarity range from 0.27 to 0.98 and were distinguished. The results of this study were mostly consistent with previous reports of different marker systems, but few different findings were also detected. The subtribe Clauseninae (tribe Clauseneae) did not clearly separate from the subtribes of the tribe Citreae. Substructuring analysis indicated that there were six subpopulations among the accessions studied. This study revealed that the POGP markers can be utilized to estimate genetic diversity, relationships and population structure in Citrus and related species in the Aurantioideae subfamily.  相似文献

‘Mal secco’ is a serious disease of lemon and other citrus species, reduces lemon production, and limits the use of susceptible cultivars especially in the Mediterranean countries, against which there is no effective management system to control the disease. The objective of this study was to determine yield efficiency, tree and fruit characters, and molecular profiles of the selected promising genotypes in lemon breeding program to develop ‘mal secco’ tolerant lemon cultivars. After 4 years of evaluation, ‘Tuzcu 894’, a progeny of ‘Finike Yerli Yuvarlak’ and ‘Kutdiken’, was designated as promising genotype with high yield, fruit weight, juice content and low seed number contrary to ‘Kutdiken’ as control. ‘Tuzcu 8911’ was suitable for dense planting having the highest yield in CV. In addition, these two promising genotypes survived after many years being subjected to artificial and natural P. tracheiphila infections and harsh winter temperatures, which were attractive to the citrus industry. Molecular analysis, as assessed with 11 inter-simple sequence repeats (ISSR) primers, indicated that ‘Tuzcu 09 Aklimon’ and its selections, ‘Tuzcu 896’, ‘Tuzcu 897’ and ‘Tuzcu 898’, were distinct from the other genotypes with similarity value of 0.82. Genetic variation among the other lemon genotypes was low and some of the genotypes were identical. It was concluded that variations in the agronomical characters are mainly due to mutations.  相似文献

This study was conducted to assess the genetic basis and develop a regression model for a QTL trait, fruit setting of a full sib population of 164 hybrids obtained between ‘Clementine’ mandarin (Citrus reticulata Blanco ‘Clementine) and ‘Orlando’ tangelo’ (Citrus paradisi Macf. ‘Duncan’ × C. reticulata Blanco ‘Dancy’). Fruit setting of a 164 full sib population field-planted in 2007 was evaluated by visual counting of fruits in 2008, 2009, and 2010. We estimated linkage groups and effects of QTLs by using MapQTL5. Based on interval mapping, seven linkage groups of the maternal and eight linkage groups of the paternal maps harboured QTLs of the fruits setting, covering a total 300 of 1744 cM Citrus map. Only three segments were associated with all three-year data (one in ‘Clementine’ and two in ‘Orlando’ map) spanning 60 cM of 1744 cM of Citrus linkage map. Twenty-seven (17%) and 13 (8%) hybrids consistently produced less than 5 fruits and the highest number of fruits (≥50) in their 2nd, 3rd, and 4th ages. Based on BACKWARD elimination procedure of PROC REG option nested in SAS program, regression models constructed for 2008, 2009, and 2010s fruit setting explained 35, 81, and 38% of the total phenotypic variation (R2) observed in the 164 full-sib hybrids, respectively (P ≤ 0.05). This study indicated that early fruit setting was a complex trait affected by many QTLs and the regression model developed in this study might be used to predict performances of hybrids with similar genetic background.  相似文献

Sequence-related amplified polymorphism (SRAP), simple sequence repeats (SSR), inter-simple sequence repeat (ISSR), peroxidase gene polymorphism (POGP), resistant gene analog (RGA), randomly amplified polymorphic DNA (RAPD), and a morphological marker, Alternaria brown spot resistance gene of citrus named as Cabsr caused by (Alternaria alternata f. sp. Citri) were used to establish genetic linkage map of citrus using a population of 164 F1 individuals derived between ‘Clementine’ mandarin (Citrus reticulata Blanco ‘Clementine) and ‘Orlando’ tangelo’ (C. paradisi Macf. ‘Duncan’ × C. reticulata Blanco ‘Dancy’). A total of 609 markers, including 385 SRAP, 97 RAPD, 95 SSR, 18 ISSR, 12 POGP, and 2 RGA markers were used in linkage analysis. The ‘Clementine’ linkage map has 215 markers, comprising 144 testcross and 71 intercross markers placed in nine linkage groups. The ‘Clementine’ linkage map covered 858 cM with and average map distance of 3.5 cM between adjacent markers. The ‘Orlando’ linkage map has 189 markers, comprising 126 testcross and 61 intercross markers placed in nine linkage groups. The ‘Orlando’ linkage map covered 886 cM with an average map distance of 3.9 cM between adjacent markers. Segregation ratios for Cabsr were not significantly different from 1:1, suggesting that this trait is controlled by a single locus. This locus was placed in ‘Orlando’ linkage group 1. The new map has an improved distribution of markers along the linkage groups with fewer gaps. Combining different marker systems in linkage mapping studies may give better genome coverage due to their chromosomal target site differences, therefore fewer gaps in linkage groups.  相似文献
