Genetics of piglet growth in association with sow's early growth and body composition were estimated in the Tai Zumu line. Piglet and sow's litter growth traits were calculated from individual weights collected at birth and at 3 weeks of age. Sow's litter traits included the number of piglets born alive (NBA), the mean piglet weight (MW) and the standard deviation of weights within the litter (SDW). Sow's early growth was measured by the age at 100 kg (A100), and body composition included backfat thickness (BF100). A main objective of this study was to estimate separately the direct genetic effect (d) and the maternal genetic effect (m) on piglet weight and daily weight gain during lactation. Variance components were estimated using the restricted maximum likelihood methodology based on animal models. The heritability estimates were 0.19 for NBA, 0.15 and 0.26 for SDW and MW at 3 weeks and 0.42 and 0.70 for A100 and BF100. The NBA was almost independent from SDW. Conversely, the A100 and BF100 were correlated unfavourably with SDW (rg<−0.24, SE<0.12). A stronger selection for litter size should have little effect on litter homogeneity in weights. Selection for lean growth rate tends to favour heterogeneity in weights. The direct effect on piglet weight at birth and daily weight gain accounted for 12% (h² (d) = 0.02) and 50% (h² (d) = 0.11) of the genetic variance, respectively. The association between d and m for piglet weight was not different from zero at birth (rg = 0.19, SE = 0.27), but a strong antagonism between d and m for daily weight gain from birth to 3 weeks was found (rg = −0.41, SE = 0.17). Substantial direct and maternal genetic effects influenced piglet growth until weaning in opposite way.  相似文献

The effect of stocking density on growth and size variability in gilthead sea-bream, Sparus aurata (L.), was tested by growing juveniles at densities of 0.35 kgm?3, 1.3 kg m?3 and 3.2 kg m?3. Fish in the highest density group grew 25% slower than fish in the lowest density group. The coefficient of variation of weight (standard deviation/mean) did not change during the experiment. The effect of size variability on growth rate was tested by creating groups with coefficients of variation of weights ranging from 0.11 to 0.32, and growing them at similar density (1.5 kg m?3). No differences in growth rates between groups were found. Only the more heterogeneous group showed a large decrease in the coefficient of variation over time. It was concluded that growth in gilthead sea-bream, although negatively correlated to stocking density, did not seem to be related to intraspecific competition as assessed by changes in size variability.  相似文献

The optimum culture system for in vitro matured and fertilised oocytes still remains to be clarified. Culture media (CM) for mammalian embryos are routinely prepared fresh for use and preserved under refrigeration during one or two weeks. The purposes of this work were (1) to compare the efficiency of a synthetic oviduct fluid (SOF) with two different bovine serum albumin (BSA) concentrations (3 and 8 g/L) for the in vitro production of bovine blastocysts, (2) to test the effect of timing on adding fetal calf serum (FCS) to the SOF, and (3) to evaluate the effects on bovine embryo development of freezing and lyophilisation as procedures for preserving the SOF. Supplementation of SOF with 3 g/L BSA increased Day-7 blastocyst expansion rates (18.3 ± 1.6 vs. 14.4 ± 0.7; P < 0.05), although no differences in hatching rates were found. Addition of FCS to SOFaa (SOF with amino acids) medium supplemented with sodium citrate (SOFaaci) at 48 and at 72 h post-insemination (PI) allowed obtaining higher Day-6 embryo development rates than when FCS was added at 18 or 96 h PI (Day-6 morulae + blastocyst rate: 30.0 ± 1.1, 40.8 ± 1.1, 43.9 ± 2.3 and 39.3 ± 0.5 for FCS addition at 18, 48, 72 and 96 h, respectively). Hatching rates were significantly improved when serum was added at 72 h PI. Finally, both refrigeration and lyophilisation appeared as useful cryopreservation procedures for SOFaaci, although a significant loss of its ability to support embryo development, compared to the control fresh culture medium, was observed.  相似文献

The analysis of marine lipophilic toxins in shellfish products still represents a challenging task due to the complexity and diversity of the sample matrix. Liquid chromatography coupled with mass spectrometry (LC-MS) is the technique of choice for accurate quantitative measurements in complex samples. By combining unambiguous identification with the high selectivity of tandem MS, it provides the required high sensitivity and specificity. However, LC-MS is prone to matrix effects (ME) that need to be evaluated during the development and validation of methods. Furthermore, the large sample-to-sample variability, even between samples of the same species and geographic origin, needs a procedure to evaluate and control ME continuously. Here, we analyzed the toxins okadaic acid (OA), dinophysistoxins (DTX-1 and DTX-2), pectenotoxin (PTX-2), yessotoxin (YTX) and azaspiracid-1 (AZA-1). Samples were mussels (Mytilus galloprovincialis), both fresh and processed, and a toxin-free mussel reference material. We developed an accurate mass-extracted ion chromatogram (AM-XIC) based quantitation method using an Orbitrap instrument, evaluated the ME for different types and extracts of mussel samples, characterized the main compounds co-eluting with the targeted molecules and quantified toxins in samples by following a standard addition method (SAM). An AM-XIC based quantitation of lipophilic toxins in mussel samples using high resolution and accuracy full scan profiles (LC-HR-MS) is a good alternative to multi reaction monitoring (MRM) for instruments with HR capabilities. ME depend on the starting sample matrix and the sample preparation. ME are particularly strong for OA and related toxins, showing values below 50% for fresh mussel samples. Results for other toxins (AZA-1, YTX and PTX-2) are between 75% and 110%. ME in unknown matrices can be evaluated by comparing their full scan LC-HR-MS profiles with those of known samples with known ME. ME can be corrected by following SAM with AM-XIC quantitation if necessary.  相似文献

生长抑素(Somatostatin,SST)是一类神经激素多肽,对脊椎动物的生长和代谢起着关键的调控作用。在斑马鱼(Danio rerio)和其它鱼类中,生长抑素由6个基因编码(sst1-sst6),sst1存在于所有脊椎动物中,并且在进化上最为保守。本研究利用CRISPR/Cas9基因编辑技术在斑马鱼中构建了可稳定遗传的生长抑素基因sst1突变体鱼系。通过比较6 dpf(days post fertilization,dpf)的sst1-/-突变体和野生型仔鱼转录组,发现sst1-/-突变体相关生化过程和信号通路发生了显著变化。相比野生型,sst1-/-突变体仔鱼中有354个基因显著上调,504个基因显著下调。GO(Gene ontology)富集分析表明,大部分差异基因与氨基酸和蛋白质生物合成相关。KEGG(Kyoto encyclopedia of genes and genomes)通路富集显示代谢通路、氨基酸合成、氨基酰基- tRNA合成、核糖体合成和rRNA加工相关通路表达增强。结果表明,sst1-/-突变体蛋白质生物合成和代谢过程更加活跃。RT-qPCR验证结果显示,随机选取的10个基因mRNA表达变化趋势与转录组测序结果一致,说明转录组测序结果真实可靠。6月龄突变体体重体长和野生型相比并无显著差异。本研究结果为进一步挖掘生长抑素基因家族的潜在功能奠定基础。  相似文献

Genetic parameters of litter traits and their relationships with farrowing kinetics traits were estimated in a Large White population to examine the impact of selection for litter size on perinatal mortality and one of its main determinants, farrowing kinetics. Data were collected on 2,947 farrowings from 1,267 sows between 1996 and 2004. Litter traits included the number born in total (NBT), number born alive (NBA), and the number (NSB) and proportion (PSB) of stillborn piglets. Four farrowing kinetics traits were considered: farrowing duration (FD), birth interval (BI = FD/NBT), heterogeneity of birth intervals (SDNB = SD of the number of piglets born each one-half hour), and birth assistance (BA) during the farrowing process. Genetic parameters were estimated using restricted maximum likelihood methodology. All traits were analyzed using a mixed linear animal model including year x month and parity as fixed effects; the additive genetic value of each animal and the sow permanent environment were treated as random effects. To normalize their distribution, kinetics traits were Box-Cox-transformed. Low heritability estimates were obtained for litter size and mortality traits, which was in agreement with literature results (i.e., 0.10 +/- 0.02, 0.08 +/- 0.02, 0.19 +/- 0.02, and 0.14 +/- 0.02 for NBT, NBA, NSB, and PSB, respectively). Heritability values were also low for kinetics traits: 0.10 +/- 0.02, 0.08 +/- 0.02, 0.01 +/- 0.01, and 0.05 +/- 0.03 for FD, BI, SDNB, and BA, respectively. The genetic correlation between NBT and NBA was strongly positive (ra = 0.90). On both phenotypic and genetic scales, NBT was positively associated with stillbirth (ra = 0.45 +/- 0.11, rp = 0.38 for NSB; ra = 0.46 +/- 0.13, rp = 0.17 for PSB). Conversely, NBA had low correlations with SB and PSB. Number born in total was moderately correlated to FD (ra = 0.34 +/- 0.15) and BI (ra = -0.37 +/- 0.15). A stronger relationship was found between NBA and BI (ra = -0.49 +/- 0.13), whereas the relationship with FD was lower (ra = 0.16 +/- 0.17). Moreover, FD was strongly correlated with stillbirth (ra = 0.42 +/- 0.12 with NSB), whereas BI was nearly independent of stillbirth. Contrary to selection on NBT, selection on NBA appears to be a good way to limit the negative side effects on stillbirth. Moreover, selection on NBA would lead to a small increase in FD and a faster and more regular birth process than would be obtained by selecting on NBT.  相似文献

OBJECTIVE: To determine M-mode and Doppler echocardiographic reference values in healthy New Zealand white rabbits. ANIMALS: 52 healthy male rabbits. PROCEDURES: The rabbits were anesthetized and M-mode measurements of the left ventricle, left atrium, and aorta and Doppler measurements of pulmonary and aortic outflow and mitral inflow were recorded. RESULTS: Mean +/- SD heart rate during echocardiographic examination was 155 +/- 29 beats/min. Mean +/- SD measurements in diastole and systole for the interventricular septum thickness, left ventricular internal diameter, and left ventricular free wall thickness were 2.03 +/- 0.37 mm and 3.05 +/- 0.45 mm; 14.37 +/- 1.49 mm and 10.25 +/- 1.22 mm; and 2.16 +/- 0.25 and 3.48 +/- 0.55 mm, respectively. Mean +/- SD left atrial-to-aortic diameter ratio was 1.17 +/- 0.14, and mean +/- SD mitral valve E-point-to-septal separation interval was 1.71 +/- 0.29 mm. Mean +/- SD for fractional shortening and ejection fraction were 30.13 +/- 2.98% and 61.29 +/- 4.66%, respectively. Mean +/- SD maximal aortic and pulmonary artery outflow velocities were 0.85 +/- 0.11 m/s and 0.59 +/- 0.10 m/s, respectively, and the peak E-to-peak A wave velocity ratio of the mitral valve was 2.19 +/- 0.46. CONCLUSIONS AND CLINICAL RELEVANCE: Results provide echocardiographic reference values for young adult male New Zealand white rabbits anesthetized with ketamine and medetomidine. Values obtained from unanesthetized rabbits, rabbits sedated with other agents, or rabbits of different size may differ from those reported here.  相似文献

Limited data are available on the use of more recent echocardiographic parameters in the rabbit. Echocardiographic examination, including conventional echocardiography and tissue Doppler imaging (TDI), was performed on 26 male New Zealand white rabbits under ketamine–midazolam sedation. Particular emphasis was placed on the more recent systolic and diastolic parameters, such as myocardial performance index (Tei index) and mitral annular motion (from septal and lateral sides of the left ventricle) obtained using pulsed TDI.Parameters that assessed systolic and diastolic function (fractional shortening, Tei index, and maximal mitral E- and A-wave velocities) were comparable to those reported in the literature for rabbits in the awake state. The less cardiodepressive anaesthetic protocol could offer a good alternative in performing echocardiographic evaluation whenever such caution is necessary. TDI is feasible in healthy rabbits and potentially suitable for the investigation of left ventricle systolic and diastolic function.  相似文献
