During the last decade, Piscine orthoreovirus was identified as the main causative agent of heart and skeletal muscle inflammation (HSMI) in Atlantic Salmon, Norway. A recent study showed that PRV-1 sequences from salmonid collected in North Atlantic Pacific Coast (NAPC) grouped separately from the Norwegian sequences found in Atlantic Salmon diagnosed with HSMI. Currently, the routine assay used to screen for PRV-1 in NAPC water and worldwide cannot differentiate between the two groups of PRV-1. Therefore, this study aimed at developing a real-time polymerase chain reaction (RT-qPCR) assay to target the PRV-1 genome segments specific for variants associated with HSMI. The assay was optimized and tested against 71 tissue samples collected from different regions including Norway, Chile and both coast of Canada and different hosts farmed Atlantic Salmon, wild Coho Salmon and escaped Atlantic Salmon collected in British Columbia, West Coast of Canada. This assay has the potential to be used for screening salmonids and non-salmonids that may carry PRV-1 potentially causing HSMI.  相似文献

Soil electrical conductivity (ECa) measured by electromagnetic induction (EM) using the EM-38 has shown promise as a soil survey tool. Soil temperature influences ECa readings, and temperature can fluctuate considerably in the upper 10cm of the soil during a day. ECa readings were taken in the horizontal and vertical dipole orientations once an hour from 8a.m. to 8p.m. at four sites on three separate days to determine if ECa values were influenced by diurnal temperature variations. Soil temperature readings were taken at the same times at four depths. EM-38 readings remained steady at all four sites all 3days. Linear regression analysis when temperature in the upper 10cm was plotted against ECa yielded low r 2 values and slopes, indicating no correlation between soil temperature in the upper 10cm and ECa values. Diurnal changes in soil temperature do not significantly influence soil ECa readings obtained with the EM-38 under the conditions encountered during the study.  相似文献

Soil apparent electrical conductivity (EC) as measured by electromagnetic induction (EM) using the Geonics® EM-38 has shown promise as a soil survey tool in Iowa and other parts of the American Midwest. Surveys made with the EM-38 often take place in the fall after harvest or in the spring prior to planting. Because of this, and because use of conservation tillage and other techniques that leave crop residues on the soil surface is prevalent in many parts of the United States, it is common for fields to have crop residues covering the soil surface when EM surveys are conducted. These residues might alter the soil EC values obtained with the EM-38. To test this possibility, soil EC was determined at two sites with three different types of residue cover (corn (Zea mays L.), soybeans (Glycine max L.), and durum wheat (Triticum turgidum L.)). The residue cover at each site was then brushed aside, and the soil EC was determined with bare ground exposed to the EM-38. On average, soil EC readings were 0.2mS/m higher when the EM-38 was exposed to bare ground, and 68% of the bare ground readings were higher than the corresponding readings through crop residues. However, the 0.2mS/m average difference is not significant when compared to the natural variation found in soil EC readings within any given soil series. Therefore, the results of this study indicate that soil EC values obtained with the EM-38 through crop residues are valid representations of the actual soil EC value.  相似文献

Levels of mercury, methylmercuri and selenium were determined in liver samples from wild mink (Mustela vision) caught in the Norwegian countries of Rogaland (38 samples), Sogn og Fjordane (15 samples) and Hedmark (18 samples). The average mercury levels from these counties were 2.6, 3.1 and 2.1 micrograms Hg/g wet weight, respectively. No significant differences in mercury levels were found. The methyl mercury levels (MeHg) were determined in 30 samples. A very strong positive correlation between total mercury (Hg) and methyl mercury (r = 0.91, P less than 0.001) was found. The average methyl mercury level was 35 per cent of total mercury. This indicates that wild mink has the ability to demethylate mercury. The selenium levels were determined in 35 samples. A strong positive correlation between the levels of total mercury and selenium (r = 0.87, P less than 0.001) was found. There was no correlation between age or nutritional condition and mercury level. In the present study mink was examined to see of it could be recommended as an indicator species for monitoring the local environment for mercury contamination. No definite answer to this question could be found. The study did reveal, however, that in all the counties studied there are individuals that are considerably contaminated with mercury.  相似文献

Soils knowledge dates to the earliest known practice of agriculture about 11,000 BP. Civilizations all around the world showed various levels of soil knowledge by the 4th century AD, including irrigation, the use of terraces to control erosion, various ways of improving soil fertility, and ways to create productive artificial soils. Early soils knowledge was largely based on observations of nature; experiments to test theories were not conducted. Many famous scientists, for example, Francis Bacon, Robert Boyle, Charles Darwin, and Leonardo da Vinci worked on soils issues. Soil science did not become a true science, however, until the 19th century with the development of genetic soil science, led by Vasilii V. Dokuchaev. In the 20th century, soil science moved beyond its agricultural roots and soil information is now used in residential development, the planning of highways, building foundations, septic systems, wildlife management, environmental management, and many other applications in addition to agriculture.  相似文献

Mouthrot infections (bacterial stomatitis) have a significant impact on the Atlantic salmon aquaculture industry in Western Canada due to economic losses and fish welfare. Bacteria isolated from lesions in the field have been identified as Tenacibaculum maritimum. Mouthrot is different to classical tenacibaculosis, which is most commonly associated with ulcerative lesions, frayed fins and tail rot. The marine fish pathogen T. maritimum is found worldwide; however, in Western Canada, the knowledge of the genetic profile of T. maritimum is limited. This study looked at increasing this knowledge by genotyping T. maritimum isolates collected from Atlantic salmon from farms in Western Canada. These genotypes were compared to other species of the genus Tenacibaculum, as well as other known sequence types within the species. The Western Canadian isolates belong to two new sequence types within the T. maritimum species. Phylogenetic analysis shows that the isolates form a distinct branch together with T. maritimum NCIMB 2154T separate from other Tenacibaculum type strains, and they are most closely related to strains from Norway and Chile.  相似文献
