The mechanism of apoptosis induced by homoharringtonine in HL-60 cells

AIM: To study the signal transduction pathway of apoptosis initiation induced by homoharringtonine in HL-60 cells. METHODS: After establishing the model of apoptosis initiation induced by homoharringtonine in HL-60 cells, at the point of apoptosis initiation, molecular caspase-3, Bcl-2, Bax and Fas/FasL were measured with flow cytometry and transmission electron microscope. ERK2 and P38 expression in HL-60 cells were detected by using immunohistochemistry. RESULTS: The model of apoptosis initiation induced by homoharringtonine was established in HL-60 cells. At the point of apoptosis initiation, upregulation of caspase-3 and decrease in Bcl-2/Bax were observed. However, the expression of Fas/FasL did not significantly change. ERK2 expression decreased and P38 expression increased. CONCLUSIONS: Caspase-3, Bcl-2, Bax and mitogen activated protein kinase pathways were involved in signal transduction of apoptosis initiation induced by homoharringtonine in HL-60 cells.  相似文献

通过田间试验,研究了新型硝基双效肥对水稻产量、效益和节肥情况的影响。结果表明,在等养分条件下,硝基双效肥相比常规施肥处理增产0.38%,但与农民习惯施肥处理相比减产2.06%。方差分析结果表明,各处理产量差异均未达到显著水平。施用硝基双效肥处理与农民习惯施肥处理和常规肥料处理效益基本相当,但减肥效果比较明显,小区试验减少率为17.2%,示范对比减少率为24.3%。  相似文献   

三种基因型羽衣甘蓝筛选适合叶绿体转化材料

以3种不同基因型的羽衣甘蓝品种"皱叶红心"、"白鸥"和"红鸥"为试材,以MS培养基为基础培养基,分别添加不同质量浓度的6-BA、NAA、IBA激素,筛选不同品种真叶叶片不定芽的诱导频率,同时确定不同材料对除草剂双丙氨膦和抗生素壮观霉素的致死临界质量浓度,为羽衣甘蓝的叶绿体转化研究筛选受体材料。结果表明:影响羽衣甘蓝叶片诱导不定芽因素中,激素质量浓度的影响比品种的影响更大;当诱导培养基中的6-BA质量浓度为1.0mg/L、NAA质量浓度为0.1mg/L时,诱导效果最佳,是3个品种的最适培养基;按照由高到低的顺序排列,3个品种的不定芽诱导率依次为"红鸥"、"白鸥"和"皱叶红心",因此择优利用品种"红鸥"作为后续转基因研究的受体材料;分别利用双丙氨膦和壮观霉素的不同质量浓度梯度进行筛选,研究二者对"红鸥"品种叶片分化的影响,发现双丙氨膦和壮观霉素的最低抑制质量浓度分别为4mg/L和50mg/L。该研究结果对今后利用叶绿体遗传转化技术改良羽衣甘蓝品种提供了技术支持。  相似文献

Basing eliminating on the idea that the generator who has the most influence on the power flow of line is mostly in favor of transmission congestion, the model of congestion management with the object of minimizing adjustments of bufions varied with generation outputs is proposed. It can eliminate the congestion effectively by judging the generators who should be adjusted and the amounts of adjustment. Power Frequency Electric Field Caused by EHV Lines and Its Effect

Mathematical model of power frequency electric field is established based on Charge Simulation Method(CSM) to analyze power frequency electric field caused by EHV lines with overhead ground wires and its effect, which are entering the megalopolis. Calculative method on the electric field under the transmission lines is analyzed and validated. According to the criterion of environmental evaluation, the distributing characteristic of electric field is summarized, in case of various arrangement manners of lines. 以设施红提葡萄为试材,研究不同的整形方式对葡萄叶片光合特性和光合关键酶活性的影响。结果表明:与扇形整形方式相比,L形和FI形整形方式可明显提高叶面积;在果实迅速发育时期,L形整形方式显著提高了叶绿素含量,Rubisco及Rubisco活化酶活性显著升高;同时,叶片质膜ATPase和叶绿体膜ATPase活性也显著升高,从而提高了叶片的光合速率。L形、FI形和V形整形方式均不同程度提高了平均单果质量、可溶性固形物含量,降低了可滴定酸含量,而L形则显著地提高了果实的品质。因此,适当的整形方式可通过调节提高设施葡萄的叶绿含量、光合关键酶和ATPase活性来提高光合速率,最终提高果实的品质。  相似文献
