Field studies were undertaken to explore the weed management strategies for transplanted and direct wet-seeded rice in the Dera Ismail Khan district of North-west Frontier Province, Pakistan. The experimental design was a randomized complete block with a split plot arrangement. The main plots consisted of two planting techniques (transplantation and direct seeding), while weed control practises assigned to the subplots included the use of the granular herbicide Sunstar 15WG (ethoxy sulfuron), Machete 60EC (butachlor), conventional hand weeding, and the weedy check (untreated control). Data were recorded on weed dynamics and the agronomic parameters of the rice crop. Economic analyses on the data were also run. The weed density and biomass were lower in the transplantation plots than the direct-seeding plots. The herbicides ethoxy sulfuron and butachlor reduced the density over the weedy check, with a density comparable to hand weeding in 2002, but slightly higher in 2003. As a result of weed management by hand weeding and herbicides, the paddy yield and its components were significantly higher for the transplanted method compared to the direct-seeded method. The comparable yield of herbicides with hand weeding offers an option for the use of herbicides as an alternative management tool. In light of our findings, it is concluded that for good economic returns, rice crops may be transplanted rather than direct-seeded. The herbicides ethoxy sulfuron and butachlor offer a weed control cover comparable with hand weeding under the transplantation method, while butachlor is good for controlling weeds even under the direct-seeded conditions in the agroclimatic conditions of the area. 相似文献

Recent studies have demonstrated a strong relationship between the intestinal microbiota and the host health. As such, consumers are increasingly becoming more concerned about the potential effect of certain foods/feeds, particularly of transgenic origin on the gut microbiota. Although the European Food Safety Authority has recommended in their guidelines, to study the effect of transgenic food/feed on host-microbiota, yet, few studies have focused on the evaluation of such effects mainly due to culturing difficulties. Therefore, this study was intended to evaluate the potential adverse effects of transgenic diet consumption on some specific gut microflora (Lactobacillus group, Bifidobacterium genus, Escherichia coli subgroup and Enterococcus genus) of rabbits. A total of forty-eight rabbits were randomly assigned into four groups and fed a diet containing a variable proportion of transgenic cottonseeds at 0, 20, 30 and 40% inclusion level, respectively. Changes in the specific or total faecal bacterial population were monitored at five different experimental stages (i.e. 0, 45, 90, 135 and 180 days) using both the traditional plate count method (TM) and quantitative real-time PCR (qPCR). No significant differences (p > .05) were observed concerning numbers of specific bacteria or total bacteria between the control and experimental groups, though qPCR showed numerically higher values in terms of 16S rRNA gene copies as compared to the values obtained from TM. However, such numerical differences were biologically insignificant (p > .05). Similarly, no significant variations were noticed in the calculated B/E (log10 copies of Bifidobacterium per g faces/log10 copies of E. coli genome per g faeces) ratios in all the groups. All the ratios were in the range of 1.24 to 1.30 throughout the experiment, indicating a good balance of intestinal microflora and greater resistance to intestinal disorders. Food security is a high priority issue on the Chinese political agenda. China’s food security is challenged by several anthropogenic, sociopolitical and policy factors, including: population growth; urbanization and industrialization; land use changes and water scarcity; income growth and nutritional transition; and turbulence in global energy and food markets. Sustained growth in agricultural productivity and stable relations with global food suppliers are the twin anchors of food security. Shortfalls in domestic food production can take their toll on international food markets. Turbulence in global energy markets can affect food prices and supply costs, affecting food security and poverty. Policy safeguards are needed to shield food supply against such forces. China must make unremitting policy responses to address the loss of its fertile land for true progress towards the goal of national food security, by investing in infrastructure such as irrigation, drainage, storage, transport, and agricultural research and institutional reforms such as tenure security and land market liberalization. The links between water and other development-related sectors such as population, energy, food, and environment, and the interactions among them require reckoning, as they together will determine future food security and poverty reduction in China. Background: Phenylalanine dehydrogenase (PheDH; EC 1.4.1.20) is a NAD+-dependent enzyme that performs the reversible oxidative deamination of L-phenylalanine to phenylpyruvate. It plays an important role in detection and screening of phenylketonuria (PKU) diseases and production of chiral intermediates as well. The main goal of this study was to find a simple and rapid alternative method for purifying PheDH. Methods: The purification of recombinant Bacillus sphaericus PheDH was investigated in polyethylene glycol (PEG) and ammonium sulfate aqueous two-phase systems (ATPS). The influences of system parameters including PEG molecular weight and concentration, pH and (NH4)2SO4 concentration on enzyme partitioning were also studied. The purity of enzyme was analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Results: A single extraction process was developed for separation and purification of recombinant PheDH from E. coli BL21 (DE3). The optimized conditions for partitioning and purification of PheDH were 9% (w/w) PEG-6,000 and 16% (w/w) (NH4)2SO4 at pH 8.0. The partition coefficient, recovery, yield, purification factor and specific activity values were achieved 58.7, 135%, 94.42%, 491.93 and 9828.88 U/mg, respectively. Also, the Km values for L-phenylalanine and NAD+ in oxidative deamination were 0.21 and 0.13 mM, respectively. Conclusion: The data presented in this paper demonstrated the potential of ATPS as a versatile and scaleable process for downstream processing of recombinant PheDH. 相似文献

Salinity is a major abiotic stress that affects crop production throughout the world. Biochar is an activated carbon soil conditioner that can alleviate the negative impacts of salinity. The research was conducted to evaluate the ameliorative effect of 1% and 2% of biochar application on wheat seed germination and growth attributes under salinity. Both levels of biochar improved the germination and growth conditions under salinity; however, 2% biochar level was more effective compared to 1% level. Root and shoot length increased up to 23% and11% with 2% biochar, respectively. The maximum increase of 16% and 10% in leaf water potential and osmotic potential was noted with 2% biochar at 150 mM salt. The decrease in proline content and soluble sugar at 2% biochar was 51% and 27%, respectively. Decrease in superoxide dismutase activity was 15.3% at 2% level of biochar under stress biochar mitigates the negative effects of salinity and improved wheat productivity. 相似文献

Zinc (Zn) is a plant nutrient; however, at elevated levels it rapidly becomes phytotoxic. In order to obtain insight into the physiological background of its toxicity, the impact of elevated Zn2+ concentrations (1 to 10 μM) in the root environment on physiological functioning of Chinese cabbage was studied. Exposure of Chinese cabbage (Brassica pekinensis) to elevated Zn2+ concentrations (≥ 5 μM) in the root environment resulted in leaf chlorosis and decreased biomass production. The Zn concentrations of the root and shoot increased with the Zn2+ concentration up to 68‐fold and 14‐fold, respectively, at 10 μM compared to the control. The concentrations of the other mineral nutrients of the shoot were hardly affected by elevated Zn2+ exposure, although in the root both the Cu and Fe concentrations were increased at ≥ 5 µM, whereas the Mn concentration was decreased and the Ca concentration strongly decreased at 10 µM Zn2+. The uptake and metabolism of sulfur and nitrogen were differentially affected at ≥ 5 µM Zn2+. Zn2+ exposure resulted in an increase of sulfate uptake and the activity of the sulfate transporters in the root, and in enhanced total sulfur concentration of the shoot, which could be ascribed partially to an accumulation of sulfate. Moreover, Zn2+ exposure resulted in an up to 6.5‐fold increase in water‐soluble non‐protein thiol (and cysteine) concentration of the root. However, nitrate uptake by the root and the nitrate and total nitrogen concentrations of the shoot were decreased upon Zn2+ exposure, demonstrating the absence of a mutual regulation of the uptake and metabolism of sulfur and nitrogen at toxic Zn levels. Evidently, elevated Zn2+ concentrations in the root environment did not only disturb the uptake, distribution and assimilation of sulfate, it also affected the uptake and metabolism of nitrate in Chinese cabbage. 相似文献

The present work was conducted to find the physicochemical properties, chemical components and antioxidant capacity of kiwifruit hydroalcoholic extracts (B-31, HO-1-14, J-242, Bruno, Greenlight, Hayward, Monty, Topstar) grown Yalova, Turkey. Phytochemical tests were performed to determine total phenolic, flavonoid and ascorbic acid. Moreover, the kiwifruit extracts were subjected to evaluate their antioxidant potential using different in vitro assays such as total antioxidant, inhibition of lipid peroxidation, reducing power, metal chelating, free radical, H2O2 and superoxide anion scavenging activities at 50–500 μg mL?1, and also found EC50 and IC50 values. The soluble solid content (SSC), titratable acidity (TA) and pH were analyzed in all the genotypes and cultivars. The highest total phenolic, flavonoid and ascorbic acid content were found in J-242 (89.53 mg GAE/100 g), J-242 (109.13 mg QE/100 g) and HO-1-14 (64.15 mg/100 g), respectively. The linoleic, palmitic, stearic and oleic acid methyl esters were detected in all kiwifruit extracts by GC/MS. Significant differences were found between genotypes and cultivars in the antioxidant tests. The extracts showed high inhibition of lipid peroxidation and compared to standards, significantly (p < 0.05). The GC/MS analyses confirmed quantitative variability in the antioxidant profiles. The SSC, TA and pH of kiwifruit genotypes and cultivars showed a moderate level. The key parameters for the most active kiwifruit included the following: total antioxidant activity for B-31, H2O2 scavenging activity for J-242 and metal chelating activity for Topstar. In conclusion, this study exhibited the important role played by the genotypes and cultivars background on the chemical content and antioxidant profiles. 相似文献

Seed presoaking improves wheat germination under marginal moisture conditions. The duration of seed presoaking was studied at The University of Arid Agriculture, Rawalpindi, using 10 wheat varieties.
