Most temperate terrestrial orchids are endangered species. Attempts to produce plantlets from plants of the genus Epipactis by asexual methods have totally failed. This study was conducted using somatic embryogenesis as a rapid vegetative propagation technique for conservation of E. veratrifolia. For these purposes, effects of different types of plant growth regulators (PGRs), different types of explants, light and dark conditions, and the effect of gibberellic acid (GA3) and Paclobutrazol (PBZ) were investigated on somatic embryogenesis induction. Abscisic acid (ABA) pre-treatment effectiveness on inducting somatic embryogenesis and increasing the number of embryos per explant were investigated. Subsequently, 16 media were tested to find the best medium for plant regeneration and shoot and root proliferation. BA (3 mg L?1) resulted in a better response than the other PGRs by supporting the development of 17 embryos per protocorm. PBZ, which resulted in 11 embryos per explant, was better than GA3. FAST medium supplemented with organic substances was recognized as the best medium for plant regeneration and shoot and root proliferation. ABA pre-treatment had positive effect on somatic embryogenesis initiation. This study, for the first time, succeeded in finding a rapid and suitable protocol for propagation and genetic resource conservation of E. veratrifolia.  相似文献

Background: The protein of Niemann-pick type C1 (NPC1) gene promotes the egress of cholesterol from late endosomes and lysosomes to other cellular compartments and contributes to a process known as reverse cholesterol transport. This study aimed to examine whether promoter methylation of NPC1 is associated with risk of cardiovascular disease (CVD). Methods: Fifty CVD patients and 50 healthy subjects as the control group were recruited in this study. Promoter methylation of NPC1 gene was defined using a nested-methylation specific polymerase chain reaction method. Statistical analyses were done using the chi-square, t-test or ANOVA tests. Results: Our study showed that the frequency of semi-methylated promoter (methylated/unmethylated status) was significantly higher in CVD patients than that in controls (OR = 6.521, 95% CI = 2.211-19.215, P = 0.008). However, a completely methylated promoter (methylated/methylated status) was not detected in any subjects in either of the two groups tested. Additionally, the analysis of clinical data according to the methylation status of NPC1 gene demonstrated that serum levels of total cholesterol, total triglycerides, high low-density lipoprotein cholesterol (LDL-C) and low high-density lipoprotein cholesterol (HDL-C) are influenced by NPC1 methylation, so that subjects with a completely unmethylated promoter (Unmethylated/unmethylated status) held lower levels of total triglycerides, total cholesterol, LDL-C and higher levels of HDL-C. Conclusion: Our findings propose that the NPC1 promoter methylation is a probable mechanism that can result in reduced/impaired NPC1 expression/activity and may thus contribute to progression of CVD. Anaesthetic efficacy of eugenol was investigated on Flowerhorn (Amphilophus labiatus × Amphilophus trimaculatus). A total of 104 fish with average weights of 12 ± 2.5, 28 ± 5 and 53 ±5.1 g were subjected to 25–200 mg L?1 eugenol and behavioural responses as well as induction and recovery times were recorded. Induction and recovery times were significantly affected by eugenol concentration as well as fish weight (P < 0.05). Generally, 49.9–127.3 s after exposure to 50–200 mg L?1 eugenol, fish reached stage 3 anaesthesia (suitable for general handling). Fish entered stage 4 anaesthesia (suitable for surgery and blood sampling) over 57.3–140.4 s post exposure to such concentrations. Recovery time was 91.7–312 s in all weight classes for all eugenol concentrations. Mortality (23%) was only observed in 12‐g fish when were subjected to 200 mg L?1 eugenol. This study showed the behavioural response of Flowerhorn to anaesthesia and eugenol efficacy as an anaesthetic in this important ornamental species. The general quadratic equation revealed that concentrations of eugenol and fish size along with their interactive effects have significantly contributed to the model, with concentration recording the highest beta value in all models (β = ?0.809, ?0.818 and ?0.909, P = 0.000). According to the results, minimum eugenol concentration to induce anaesthesia in less than 3 min was 50 mg L?1.  相似文献

Soil water retention characteristic is required for modeling of water and substance movement in unsaturated soils and need to be estimated using indirect methods. Point pedotransfer functions (PTFs) for prediction of soil water content at matric suctions of 1, 5, 25, 50, and 1500 kPa were developed and validated using a data-set of 148 soil samples from Hamedan and Guilan provinces, Iran, by multiobjective group method of data handling (mGMDH). In addition to textural and structural properties, fractal parameters of the power-law fractal models for both particles and aggregates distributions were also included as predictors. Their inclusion significantly improved the PTFs’ accuracy and reliability. The aggregate size distribution fractal parameters ranked next to the particle size distribution (PSD) in terms of prediction accuracy. The mGMDH-derived PTFs were significantly more reliable than those by artificial neural networks but their accuracies were practically the same. Similarity between the fractal behavior of particle and void size distributions may contribute to the improvement of the derived PTFs using PSD fractal parameters. It means that both distributions of the pore and particle size represent the fractal behavior and can be described by fractal models.  相似文献

This study reports on the phenotypic and genetic (co)variance components for reproductive traits in Zandi sheep, using between 1,859 and 2,588 records obtained from 577 ewes. The data were collected from the Khojir Breeding Station of Zandi sheep in Tehran, Iran from 1994 to 2008. The basic traits were litter size at birth (LSB), litter size at weaning (LSW), litter mean weight per lamb born (LMWLB), and litter mean weight per lamb weaned (LMWLW), and the composite traits were total litter weight at birth (TLWB) and total litter weight at weaning (TLWW). Genetic analyses were carried out using the restricted maximum likelihood method that was explored by fitting the additive direct genetic effects and permanent environmental effects of the ewes as random effects and the ewe age at lambing and lambing year as fixed effects for all of the investigated traits. Akaike’s information criterion was used to choose the most appropriate model. LSB, LSW, LMWLB, LMWLW, TLWB, and TLWW direct heritability estimates were 0.07, 0.05, 0.12, 0.10, 0.08, and 0.14, respectively. The estimated fractions of variance due to the permanent environmental effects of the ewe ranged from 0.03 for LMWLB to 0.08 for LMWLW and TLWW. Corresponding repeatability estimates ranged from 0.10 for LSW to 0.22 for TLWW. Direct genetic correlations varied from ?0.61 for LSB–LMWLB to 0.88 for LSB–LSW and LSB–TLWB. Results indicate that genetic change depends not only on the heritability of traits, but also on the observed phenotypic variation; therefore, improvement of non-genetic factors should be included in the breeding programs.  相似文献

The saturated hydraulic conductivity (\(K_{\text{s}}\)) is one of the important soil hydraulic properties which plays a significant role in developing flow transport models and irrigation and drainage practices. In this research, artificial neural networks approaches, group method of data handling (GMDH) model and a hybrid intelligent model based on combination of GMDH and harmony search (HS) model (GMDH-HS) were developed to estimate \(K_{\text{s}}\) based on 151 field samples collected from the northeast of Iran. Eleven topsoil properties were used as input parameters to estimate \(K_{\text{s}}\). The five quantitative standard statistical performance evaluation measures, i.e., coefficient of efficiency, root-mean-square error, mean square relative error, mean absolute percentage error and relative bias, were employed to evaluate the performance of various developed models. Statistical results indicated that the best performance can be obtained by GMDH-HS in terms of different evaluated criteria during the training and testing datasets for \(K_{\text{s}}\) estimation.  相似文献

Walnut (Juglans regia) is economically important for both its wood and nut nutritional value, but it is susceptible to diseases such as walnut bacterial blight, caused by Xanthomonas arboricola pv. juglandis (Xaj). Walnuts contain many phenolic compounds, providing a good model on which to study polyphenol oxidase (PPO). We inoculated the detached walnut fruits of cultivars Ford, Chandler, Franquette, Robert Livermore, and Payne with Xaj and measured the induction of PPO activity in infected sites and adjacent to infected sites. Compared to infected and uninfected sites, PPO activity was induced significantly in areas adjacent to infected sites in all cultivars except Ford. Ford and Franquette, presenting the lowest and highest PPO activity, showed the largest and smallest mean diameter spots in response to Xaj, respectively. Polyacrylamide gel electrophoresis confirmed monophenol oxidase activity of walnut PPO in the assessed tissues. Then, we revealed the antipathogenic potential of walnut PPO through Agrobacterium tumefaciens-mediated walnut JrPPO1 gene transfer into tobacco (Nicotiana tabacum). Two transformed tobacco lines overexpressing the JrPPO1 gene were regenerated successfully and challenged with Pseudomonas syringae pv. tabaci. Transgenic lines showed significantly higher PPO activity and lower disease severity to the pathogen compared to the control. However, a significant difference in disease severity and PPO activity level was observed between the two transgenic lines. Our results demonstrate a potential defence-related role of PPO in transgenic tobacco and its induction in areas adjacent to infection sites in walnut cultivars treated with Xaj.  相似文献

For the first time, the current study reports the genetic and phenotypic correlations between growth and reproductive traits in Zandi sheep. The data were comprised of 4,309 records of lamb growth traits from 1,378 dams and 273 sires plus 2,588 records of reproductive traits from 577 ewes. These data were extracted from available performance records at Khojir Breeding Station of Zandi sheep in Tehran, Iran, from 1993 to 2008. Correlations were estimated from two animal models in a bivariate analysis using restricted maximum likelihood procedure between lamb growth traits [birth weight (BW), weaning weight at 3 months of age (WW), as well as six-month weight (6 MW)] and ewe reproductive traits [litter size at birth (LSB), litter size at weaning (LSW), total litter weight at birth (TLWB), and total litter weight at weaning (TLWW)]. The genetic correlations between BW and reproductive traits varied from low to high ranges from 0.10 for BW–LSB to 0.86 for BW–TLWB. WW was moderately (0.37) to highly (0.96) correlated with all the reproductive traits. Moreover, the genetic correlations were observed between 6 MW and reproductive traits, varied from 0.19 to 0.95. Relationships between growth and reproductive traits ranged from 0.01 for BW–LSW to 0.28 for BW–TLWB in phenotypic effects. Results indicated that selection to improve WW would have high effect on genetic response in TLWW, and also, these results could be effective for all of the reproductive traits in Zandi sheep.  相似文献

The present study aimed to determine hemolysis interference in measurement of plasma biochemical parameters in fish specimens. For this purpose, blood samples were harvested from 24 Huso huso juveniles. After centrifugation, each plasma sample was divided into seven portions to make seven levels of hemolysis. Hemolysis was induced by addition of different percentages of the corresponding whole blood [0 (non-hemolyzed control group), 1, 3, 5, 10, 15, and 20% of whole blood]. Albumin, total protein, calcium, phosphorus, glucose, sodium, potassium, chloride, alkaline phosphatase (ALP), aspartate transaminase (AST), and alanine transaminase (ALT) were measured in different samples. Results showed that plasma albumin, ALP, potassium, and AST significantly increased (more than 200% at the highest hemolysis level) in the hemolyzed samples. Also, plasma total protein and phosphorus showed significant elevation in the hemolyzed samples (more than 70% at the highest hemolysis level). Plasma glucose, calcium, chloride, and ALT showed narrow but significant increase in hemolyzed samples (11.8–35.2% at the highest hemolysis level). Plasma sodium showed no significant changes in the hemolyzed samples. In conclusion, the present results show that hemolysis markedly affects plasma parameters levels, which interferes with plasma results interpretation. Therefore, analysis of hemolyzed samples should be avoided or the results should be interpreted with caution. This study encourages further investigations to develop methods for omitting the effects of hemolysis by sample blanking and/or presenting correction coefficients for measurement of plasma parameters in samples with different levels of hemolysis.  相似文献
