The objective of this study was to determine the effects of dietary supplementation of Nigella sativa L. seeds, Rosmarinus officinalis L. leaves and their combination on rumen metabolism, nutrient intake and digestibility, growth performance, immune response and blood metabolites in Dorper lambs. Twenty-four entire male Dorper lambs (18.68?±?0.6 kg, 4–5 months old) were randomly assigned to a concentrate mixture containing on a dry matter basis either, no supplement (control, T1), 1% R. officinalis leaves (T2), 1% N. sativa seeds (T3) or 1% R. officinalis leaves +1% N. sativa seeds (T4). The lambs had ad libitum access to urea-treated rice straw (UTRS) and were raised for 90 days. Supplemented lambs had greater (P?< 0.05) intake of DM and UTRS than the control lambs. The T4 lambs had lower (P?< 0.05) nutrient digestibility than those fed other treatments. Total and daily weight gain was greater (P?< 0.05) in T2 lambs than those fed other diets. The T3 and T4 lambs had greater (P?< 0.05) ruminal pH than the T1 and T2 lambs. Supplemented lambs had lower (P?<?0.05) ruminal total volatile fatty acids, acetate, propionate, NH3-N and C18:0 than the control lambs. The T4 lambs had lower (P?< 0.05) population of Fibrobacter succinogenes, Ruminococcus albus, methanogens and total protozoa compared with those fed other diets. Supplemented lambs had lower (P?< 0.05) neutrophils, basophils and serum urea and greater (P?<?0.05) serum IgA and IgG compared with the control lambs. The current results emphasised the variation in the efficacy of medicinal plants in ruminant nutrition.  相似文献

Bacground: Evidence from several lines of investigations suggests that Toll-like receptor 4 (TLR4) is involved in atherosclerosis as a bridge between innate and acquired immunity. Percutaneous coronary intervention (PCI) can trigger inflammation through activation of human TLR4 (hTLR4) on monocytes. Hydrocortisone as an anti-inflammatory and immuno-suppressant agent has multiple mechanisms of action. In this study, we aimed at assessing the effects of hydrocortisone on monocyte expression and activity of hTLR4 in patients underwent PCI. Methods: Blood samples were taken from a total of 71 patients with chronic stable angina who were scheduled for a PCI, before the intervention. Thirty patients received 100 mg hydrocortisone prior to the procedure. Control group was composed of 41 patients underwent PCI without receiving hydrocortisone. Blood collection was repeated 2 and 4 h after PCI. The expression of hTLR4 on the surface of CD14+ monocytes and the serum levels of TNF-α and IL-1β were measured using flowcytometry and Sandwich ELISA. Results: Compared with controls, hydrocortisone significantly reduced monocyte expression of hTLR4 in test group (P<0.01). In addition, it had a significant effect on reduction of serum concentrations of TNF-α and IL-1β in test group in a time-dependent manner (P<0.01). Conclusion: In this study, hydrocortisone was able to reduce the hTLR4/CD14 positive monocytes and its related pro-inflammatory cytokines, thus it can decrease inflammatory responses following PCI. This study was performed to investigate the effect of dietary supplementation with Aloe vera (AV) powder and synbiotic as growth promoter agents on performance, gut morphology, immune responses, hematology and serum biochemistry in broilers. A total of 240‐day‐old male broiler chicks (Ross 308) were randomly assigned to six treatments with four replicates. Birds were offered either a corn‐soybean meal basal diet (control) or the basal diet supplemented with 200 mg/kg virginiamycin (VM), 1 g/kg synbiotic (Syn), 2.5 g/kg AV (AV1), 5.0 g/kg AV (AV2) or 7.5 g/kg AV (AV3). Chickens fed any of the diets, except diet AV1, exhibited better feed conversion ratios at the 14–28 day period and higher average daily gain and duodenal villus height/crypt depth ratio at 42 days than those fed the control diet. Synbiotic supplementation caused a marked increase in the serum antibody titer against infectious bursal disease and infectious bronchitis vaccines. Feeding diet AV3 significantly increased red blood cell count and hemoglobin concentration, and decreased serum triglyceride level compared to the control group. The results suggested that dietary inclusion of 5 and 7.5 g/kg AV, similar with synbiotic supplementation, can be applied as effective alternatives to in‐feed antibiotics for broiler diets.  相似文献

  • 1. The lack of information about marine biodiversity is problematic for the selection of conservation reserves that aim to protect representative samples of biodiversity. A number of surrogate measures for biodiversity have been suggested as a potential solution to this problem.
  • 2. The present study tested the effectiveness of using higher taxa of macroinvertebrates as a surrogate for species‐level identification to depict spatial variation in species richness and assemblage variation and to select conservation reserves in one estuary in south‐east Australia.
  • 3. Spatial patterns of richness and assemblage variation for species were significantly correlated with patterns defined from genera, families, orders, classes, and phyla with a decline in the magnitude of correlation coefficients from finer to coarser resolutions. A network of reserves selected to include representatives of all phyla, classes, orders, families and genera coincidentally included 54%, 61.7%, 75%, 92.6%, 98.8% species in 8.3%, 13.9%, 17.7%, 44.4% and 58.3% of grid cells, respectively. However, only reserves selected for genera, families and orders performed significantly better than random selection.
  • 4. Percentage of species represented by orders, families and genera in a realistic level of available grid cells for conservation (i.e. 13.9%) were very close ranging between 70 and 73.5%. A factor diminishing the performance of order as surrogate for species richness was related to the difficulty of identifying many macroinvertebrates to the order level. Therefore, it is concluded that genus‐ and family‐level identification is an effective surrogate for species‐level identification for conservation planning in estuaries.

Objective : To describe the clinical, mycological, histopathological and molecular findings in green iguanas (Iguana iguana) affected with severe dermatophytosis in selected flocks near Tehran, Iran. Materials and Methods : Samples were collected from the scales of skin lesions and tested with standard mycological methods and dermatophyte‐specific PCR amplification analysis using the primer pair for the chitin synthase 1(CHS1) gene. Results : All iguanas were definitively diagnosed with dermatophytosis using both traditional and molecular diagnostic methods. PCR fingerprinting profiles using simple repetitive (GACA)4 primers showed that all diagnosed iguanas had the same pattern profile. Intraspecific variability was not observed for these isolates. Oligonucleotide sequencing of CHS1 gene PCR products confirmed Trichophyton mentagrophytes var. interdigitale as the infectious agent. Clinical Significance : These results suggest that (GACA)4‐based PCR has utility both as a simple and rapid method for identification of dermatophyte species and for differentiation of T. mentagrophytes variants.  相似文献

Syngnathids (Pisces, Syngnathidae: seahorses and pipefish) were investigated for their use as a flagship group to evaluate the conservation value of estuarine seagrass beds in estuaries in south‐east Australia. Some species of syngnathids are listed internationally as vulnerable or endangered, and they are a charismatic group of fish that attracts a high level of public support and sympathy. Syngnathids are also protected in several states of Australia. Conservation of syngnathids might provide coincidental benefits to other species that share their habitats.
  • 2. The effectiveness of syngnathids as a flagship group was assessed by (1) testing for correlations with other fish in species richness, density, assemblage variation, and summed irreplaceability value, and (2) determining the number of species of all other fish coincidentally captured in marine protected areas (MPAs) selected for syngnathids. The study was undertaken in a single estuary (scale: tens of square kilometres) and across multiple estuaries (scale: hundreds of square kilometres).
  • 3. Densities of syngnathids and other fish were correlated only at the scale of multiple estuaries. Species richness and summed irreplaceability of syngnathids and other fish were not spatially correlated. Spatial variations in assemblages of syngnathids and other fish were correlated. MPAs selected for syngnathids included more non‐syngnathid species than a random selection of locations.
  • 4. This study provides evidence that ranking the conservation value of seagrass beds on the basis of the density and assemblage variation of syngnathids, and selecting MPAs to represent syngnathid species, will simultaneously benefit other fish. Synganthids are therefore regarded as a useful flagship group for conservation planning.

INTRODUCTION: High levels of rheumatoid factors (RF) are detectable in serum of the majority of patients with rheumatoid arthritis (RA), but 5-10% of patients remain seronegative (SN). Despite clinical and genetic similarities between these two subsets of RA, it has been proposed that they may be regarded as distinct clinical entities. METHODS: In the present study a panel of monoclonal antibodies (mAb) recognizing RF-associated cross-reactive idiotypes (CRI) linked to the VH1 (G8), VH4 (LC1), VK3b (17-109) and a mAb recognizing the VK3 subgroup (C7) of immunoglobulin variable region (IgV) gene products were used to quantitate the level of expression of these gene products in serum and synovial fluid of 35 seropositive (SP) and 8 SN RA patients by capture ELISA. RESULTS: While the concentration and relative proportion of the IgV are recognized by the mAb G8, 17-109 and C7 were significantly higher in serum and synovial fluid of the SP RA, compared to the SN-RA patients (G8, p = 0.009; 17-109, p = 0.0001; C7, p = 0.001). The CRI recognized by the mAb LC1 was highly represented in serum and synovial fluid of the SN-RA patients. There have been no significant differences in the level of expression of these IgV gene products (other than the product recognized by C7 mAb in SP patients) between serum and synovial fluid of either group of patients. CONCLUSION: Our results suggest that the expressed repertoire of Ig VH and VK genes in these two subsets of RA is differentially regulated and may be influenced by selective mechanisms leading to positive or negative selection of certain genes.  相似文献
