The susceptibility of sheep to scrapie is under the control of the host’s prion protein (PrP) gene and is also influenced by the strain of the agent. PrP polymorphisms at codons 136 (A/V), 154 (R/H) and 171 (Q/R/H) are the main determinants of susceptibility/resistance of sheep to classical scrapie. They are combined in four main variants of the wild-type ARQ allele: VRQ, AHQ, ARH and ARR. Breeding programmes have been undertaken on this basis in the European Union and the USA to increase the frequency of the resistant ARR allele in sheep populations. Herein, we report the results of a multi-flock study showing the protective effect of polymorphisms other than those at codons 136, 154 and 171 in Sarda breed sheep. All ARQ/ARQ affected sheep (n = 154) and 378 negative ARQ/ARQ controls from four scrapie outbreaks were submitted to sequencing of the PrP gene. The distribution of variations other than those at the standard three codons, between scrapie cases and negative controls, was statistically different in all flocks. In particular, the AT137RQ and ARQK176 alleles showed a clear protective effect. This is the first study demonstrating a protective influence of alleles other than ARR under field conditions. If further investigations in other sheep breeds and with other scrapie sources confirm these findings, the availability of various protective alleles in breeding programmes of sheep for scrapie resistance could be useful in breeds with a low frequency of the ARR allele and would allow maintaining a wider variability of the PrP gene. 相似文献

Molecular weight and size distributions of two glutenin polymers were determined by a multi-angle laser light scattering (MALLS) photometer on-line to a size exclusion chromatography (SEC) system. Two glutenin polymers extracted, sonicated and purified from wheat flours of different cultivars, i.e. Cheyenne and Chinese Spring, were accurately fractionated by SEC using three buffers (pH 2.6, 4.0 and 6.9) and two column sets. Both molecular weight distribution (MWD) and radius of gyration distribution (RGD) could be used to differentiate the two cultivars. MWD of glutenin polymers is a complex mixture of high- and low-molecular weight fractions and the relative percentage was found to be very different. The two cultivars were found to be different; in particular, the Chinese Spring polymer showed more compact conformation than the Cheyenne polymer. The slope of the conformation plot for glutenin was about 0.37, close to the theoretical value for compact spheres. Determination of glutenin MWD and RGD was difficult and depended on the buffer used and the SEC columns. 相似文献

Effects of land use and landscape on spatial distribution and morphological features of gullies in an agropastoral area in Sardinia (Italy) 总被引:3,自引:0,他引:3
According to the United Nations Convention to Combat Desertification (UNCCD), soil erosion is one of the main causes of desertification in the Northern Mediterranean. Recently, the role of channelled erosion in sediment delivery has been underlined by several studies. The aim of this paper is to describe a simplified experimental methodology settled up for studying the relationships between gully erosion and land use.The experimental site, which is wide about 720 ha, is located in a hilly area threatened by desertification in Central Eastern Sardinia (Italy). This area is characterised by irregular morphology with steep slopes, where agropastoral activities, particularly tillage with heavy machinery and overgrazing, are causing severe soil erosion and compromising agricultural productivity. Previous studies carried out in the same area highlighted the need to quantify and monitor soil erosion in order to outline management guidelines for farmers. In this study, an integrated methodology that uses aerial photographs and field surveys was applied to inventory the gullies of the area. The degree of activity and morphological characteristics of gullies were assessed in the field by measuring the position and shape of gully heads, and their width and depth. Total gully volume was also estimated. Relationships among distribution, density and morphological characteristics of gullies, and geomorphological features, land use and land use historical dynamics of the area have been studied. The results show the very significant impact of agricultural activities on gully activity and density. 相似文献
