The changes in water proton transverse relaxation behavior induced by aggregation of whey proteins are explained in terms of the simple molecular processes of diffusion and chemical exchange. The water self-diffusion coefficient was measured in whey protein solutions and gels by the pulsed field gradient NMR method. As expected, water self-diffusion was reduced with increased protein concentrations. Whatever the concentration, the water molecules were free to diffuse over distances varying from 15 to 47 mum. Water diffusion was constant over these distances, demonstrating that no restrictions were found to explain the water hindrance. The modification in protein structure by gelation induced a decrease in water diffusion. The effects of protein concentration on water diffusion are discussed and modeled. Two approaches were compared, the obstruction effect induced by a spherical particle and the cell model, which considered two water compartments with specific self-diffusion coefficients.  相似文献

Environmental pollution due to arsenic (As) has been well studied and most investigations have focused on inorganic As coming from industrial areas. However, the natural rock-borne contamination of this metalloid has been underestimated and few studies focusing on annelids have addressed the question of geogenic As distribution in earthworm biogenic structures. Our study concentrated on the earthworm drilosphere and investigated the distribution of As in surface-casts and burrow-linings. In this context, we assumed that the concentration of geogenic As should differ in biogenic structures (faeces, burrow-linings) compared to non-ingested soil. To test our hypothesis, microcosms in controlled conditions were filled with soil containing 157 mg As kg?1 from a geogenic origin. The soil was collected from a meadow in the Jura Mountains. The earthworm Aporrectodea giardi was selected because of its natural presence in the meadow and its anecic behaviour.We found that while earthworm surface-casts were enriched in carbon, nitrogen and available phosphorus compared to non-ingested soil, no enrichment was found for geogenic arsenic. The coarse sand fraction contained 3.5 times more As than silt and clay fractions but no significant difference existed between surface-casts and non-ingested soil. Iron was shown to be the main mineral bearing phase for As and predominated in the coarse sand fraction. No retention effect of organic matter on geogenic arsenic was observed. In conclusion, it appeared that geogenic As did not affect biological components and environmental aspects. These findings are particularly encouraging considering that most of the environments at an altitude of 1330 m in the Jura Mountains have exposed rocks, especially on meadows that are regularly grazed by cattle.  相似文献

Different radiographic findings may be observed during double contrast cystography due to patient positioning affecting the distribution of positive and negative contrast media. A mass lesion was created in the urinary bladder of a canine cadaver to allow evaluation of the effect of patient positioning on the appearance of a mass during double contrast cystography. The mass appeared as a filling defect only on those views where positive contrast medium surrounded the mass. Otherwise, the mass appeared as a summation. Additionally, a patient is described illustrating the effect of patient positioning on detecting mural filling defects during double contrast cystography.  相似文献

This study examines the recovery of the microbial compartment following active restoration of a North American ombrotrophic peatland extracted for horticultural peat-based substrates and restored by the Sphagnum moss transfer method. We used phospholipid fatty acids (PLFAs) to portrait the microbial community structure and Community Level Physiological Profiles (CLPP) to describe the functional diversity of the microbial communities. Our results indicate that the PLFA profiles were different between the beginning and the end of the growing season, but that it was impossible to distinguish five different vegetation classes found along the disturbance-recovery gradient on the basis of the microbial community structure. The pH, the cover of mosses, Ledum groenlandicum and Eriophorum vaginatum var. spissum were the best environmental predictors for the PLFA composition. The newly formed peat found in aerobic conditions beneath restored Sphagnum carpets had the highest decomposition capacity, whereas the lowest rates were found in the surface samples of non-restored conditions or in the deepest horizons of the natural samples. A large proportion of the variation in the physiological profiles was explained with variables related to the vegetation cover, the physicochemical environment and the microbial structure of the community, which is very promising for future monitoring studies. Overall, this study demonstrates that the recovery of particular plant groups, namely mosses and shrubs in restored peatlands might be the driver of changes occurring in the structure of the microbial communities in restored peatlands.  相似文献

  1. Conservation of riverine fish often aims to improve access to spawning grounds and restore longitudinal connectivity by removing migration barriers, and involves substantial investments. However, these investments also enable non‐native predators to invade upstream into spawning areas and potentially adversely affect the recruitment of threatened freshwater fish through egg or fry predation.
  2. Detecting egg predation is often challenging. Visual inspections of fish gut contents may underestimate predation of soft materials such as eggs and fry, which limits the discovery of predators preying upon these life‐stages. DNA‐based detection assays may offer a more sensitive tool to assess predation of soft materials.
  3. A conservation issue was confirmed by developing and applying a species‐specific DNA‐based detection assay: invasive round goby (Neogobius melanostomus) prey on the eggs or fry of the threatened common nase (Chondrostoma nasus) in Switzerland.
  4. DNA‐based detection assays were also developed for five other valuable native fish species, including endangered salmonid and cyprinid river spawners. The applicability of the assays was confirmed in a series of laboratory and field feeding experiments involving eggs and fish tissue. In addition, this work provides a guiding framework for conservation managers regarding the use and applicability of different DNA‐based detection approaches for gut content analysis.
  5. The results of this study could inform local conservation measures – such as temporary reductions in the density of round goby at spawning sites prior to spawning – and demonstrate how targeted application of species‐specific molecular markers may advance freshwater fish management.

Phlorotannins are polyphenols occurring exclusively in some species of brown algae, known for numerous biological activities, e.g., antioxidant, antiproliferative, antidiabetic, and antiallergic properties. Their effects on the response of human lung cells to benzo[a]pyrene (B[a]P) has not been characterized. Our objective was to in vitro evaluate the effects of a phlorotannin-rich extract obtained from the brown algae Ascophyllum nodosum and Fucus vesiculosus on B[a]P cytotoxic effects. The A549 cell line was incubated with B[a]P for 48 and 72 h in the presence or absence of the brown algae extract. Cytochrome P450 activity, activation of P2X7 receptor, F-actin disorganization, and loss of E-cadherin expression were assessed using microplate cytometry and fluorescence microscopy. Relative to control, incubation with the brown algae extract was associated with lower B[a]P-induced CYP1 activity, lower P2X7 receptor activation, and lower reactive oxygen species production. The brown algae extract inhibited the alterations of F-actin arrangement and the downregulation of E-cadherin expression. We identified a phlorotannins-rich extract that could be deeper investigated as a cancer chemopreventive agent to block B[a]P-mediated carcinogenesis.  相似文献

The Bois-des-Bel Sphagnum peatland (Rivière du Loup, QC) was restored in 1999 after 20 years of abandonment. Restoration work included not only the blockage of drainage ditches, but also the reintroduction of plant material including Sphagnum remains. Following restoration, the physicochemical and microbial characteristics (biomass, activity and composition) of the peat were analysed. The goal was to investigate the functional status of the restored ecosystem. The high N:P (>20) and N:K (>15) ratios indicated possible K and P deficiencies in the restored and the cutover sites, which is mainly associated with the intense leaching and the high degree of decomposition of the peat in these sites. The concentrations of , P and K in the top layer of the restored site were closer to those of the natural site, which indicated a possible effect of restoration on the physicochemistry of the restored site. Microbial biomass values derived from the FE technique followed a gradient natural>restored>cutover through the profile, which was not the case with the SIR technique. Values from SIR varied overall between 0.19 and 4.88 mg C g−1 and were significantly higher in the natural site. The natural peatland site had significantly (P<0.05) greater cumulative C-CO2 production (surface aerobic: 4.5-8.7 μg C-CO2 g−1 h−1). The poor organic matter quality was the main explanation for the low respiration rates of the surface layer in the restored and the cutover site. All CO2 respiration data were plotted against time and the resulting curves were successfully fitted to a global kinetic model. Methane production was detected at low but measurable rates in the natural and the restored samples, but not in the cutover peat. Overall, the results confirmed the existence of a lag between the positive response of vegetation to restoration and that of the microbial compartment. This study also pointed out that some physicochemical dysfunctions remained even after three growing seasons following restoration in the subsurface horizons studied.  相似文献

Pulsed field gradient NMR spectroscopy was used to determine the poly(ethylene glycol) (PEG) self-diffusion coefficient (D(PEG)) as a function of NaCl concentration (C(NaCl)) and denaturation time (t(D)) in whey protein solutions and gels. D(PEG) in the gel decreased with increasing C(NaCl) concentrations and increased with increasing t(D); the increase ceased for all PEGs when the gel was fixed. This increase was more pronounced for the 82250 g/mol PEG than the 1080 g/mol PEG. Moreover, the diffusion coefficient of nonaggregated whey protein was measured and an increase for longer t(D) was also observed. Scanning electron microscopy images and (1)H spectra demonstrated that D(PEG) were related to the structure changes and to the percentage of beta-lactoglobulin denaturation.  相似文献
