The system of rice intensification (SRI) has generated considerable debate globally, particularly with regard to its potential to raise rice yields. Proponents of SRI have reported that the average rice yield with SRI is double the current average yield and can be increased to the level of three to four times. Opponents say the reported high yields are due to measurement error and that usual information expected in support of these fantastic yields is missing. The number of SRI adopters has increased in India in recent years. We evaluate the impact of adoption of SRI practices on rice yields, the economics of paddy cultivation and labour inputs based on field research conducted in Purulia, West Bengal, India. Paddy yields with SRI were higher than those under conventional paddy cultivation by 32% and net returns were higher by 67%. Labour input was reduced by 8%. SRI adoption enabled farmers consistently to enhance paddy yields, increase returns and save labour; and enhance productivity with respect to the key inputs in terms of paddy output per unit of seed, fertilizer and labour-day. SRI promises to be a significant alternative for not only raising paddy yields, but also for managing paddy based farming in resource-starved regions.  相似文献

The VP 28 gene encoding a structural envelope protein of the white spot syndrome virus (WSSV) was cloned into a pET32a(+) expression vector for the production of the recombinant VP28 protein. A purified recombinant protein of 39.9 kDa size was used for polyclonal antibody production in rabbit. Specific immunoreactivity of the rabbit anti rVP28 antiserum to the viral antigen was confirmed by a Western blot. The specificity of this polyclonal anti‐rVP28 antiserum to detect the presence of the virus in WSSV‐infected Penaeus monodon was verified using a immunodot blot assay. Immunodot blot showed a positive reaction in infected shrimp tissues with prominent colour development using 3,3′,5,5′‐tetramethylbenzidine (TMB) as a chromogenic substrate when compared with 3–3′ diaminobenzidine tetrahydrochloride (DAB). Highest signal intensities of the immunodots were observed in infected shrimp pleopod extracts and haemolymph. On comparison with polymerase chain reaction (PCR), immunodot blot could detect 76% of PCR‐positive WSSV‐infected shrimp samples. Immunodot blot was found to be equivalent to first‐step PCR sensitivity to detect WSSV particles estimated to contain 1.0 × 105 viral DNA copies.  相似文献

The objective of this study was to compare the variation in the soil seed banks and the aboveground vegetation in relation to three habitats, i.e., swamp forests, grassland and sand dunes within the Sundarbans mangrove forests of Bangladesh. We collected vegetation data (species and their percentage cover) by using quadrat sampling: 10 m × 10 m for swamp forests and 5 m × 5 m for grassland and sand dunes. We estimated the density of viable seeds of species in the seed bank by counting germinants from soil cores in a germination chamber. Species richness and composition of both aboveground vegetation and the soil seed banks differed significantly among habitats. We identified a total of 23 species from the soil seed bank. Of these, two were true mangrove species and the remaining were non-mangrove species, including halophytic grasses, herbs and mangrove associate species. Our results confirm that mangrove species do not possess a persistent soil seed bank. The presence of high-density non-mangrove and associated mangrove species in the soil seed bank implies that after frequent catastrophic disturbances which limit incoming propagules from adjacent forest stand, large canopy gaps can easily become invaded by non-mangrove and mangrove associate species. This would result in the formation of a cover of non-mangrove species and cryptic ecological degradation in mangrove habitats. We suggest that forest managers should actively consider gap plantations with mangrove species in the large canopy gaps created after catastrophic disturbances to counteract the invasion of non-mangrove species and cryptic ecological degradation.  相似文献

The present study evaluated the role of recombinant RNA‐dependent RNA polymerase (RdRp) protein of Macrobrachium rosenbergii nodavirus (MrNV) in modulating the immune response and in reducing MrNV load in infected prawn. In the first experiment, prawns (25–30 g) were injected with recombinant RdRp protein (RP) at a concentration of 0, 1.0 and 10 μg, and immune parameters and expression of some immune‐related genes were measured up to 14 days post injection (p.i.). In the second experiment, early juveniles were injected with a similar dose of RdRp and animals were challenged by immersion with MrNV. The infection status was detected in muscles by nested RT‐PCR up to 21 days post challenge. Prawn injected with higher concentration of RP showed significantly higher total haemocyte count at different period post injection. Significant up‐regulation of immune‐related genes was observed within 24 h in prawn treated with lower dose of RP and after 7 days p.i. at higher level of RP injection compared with adult control. Most of the tested samples (63%) were found to be RT‐PCR positive for MrNV at 48 h of post‐immersion challenge. After 14 days, MrNV was detected only in control prawn, while both RP‐injected groups were MrNV negative. This study elucidated the potential viral load reduction role played by RdRP in MrNV‐infected prawn.  相似文献

The oomycete Phytophthora colocasiae that causes taro leaf blight is the most devastating disease of taro and is widely distributed worldwide. Molecular and phenotypic techniques were employed for assessing and exploiting the genetic variability among four populations of P. colocasiae obtained from a fine spatial scale (multiple leaf blight lesions on single taro leaf). Phenotypic characters such as virulence, morphology and mating type showed no variation. ITS characterization revealed detectable polymorphism among isolates of P. colocasiae. The mean number of haplotypes (H), haplotype diversity (HD), nucleotide diversity (π), and nucleotide substitution rate (θ) among analyzed sequences were 6.75, 1.00, 0.069, and 0.088 respectively. High levels of inter and intra specific variation were detected by random amplified polymorphic DNA (RAPD) assays. Moderate genetic diversity (H?=?0.2651) was observed among populations of P. colocasiae. Analysis of molecular variance (AMOVA) confirmed that most of the genetic variability was confined to within a population (63.54 %). The coefficient of genetic differentiation among populations (G ST ) was 0.2007 and estimates of gene flow (Nm) among populations was 1.991 migrants per generation. Cluster analysis using UPGMA revealed that individuals from the same population failed to cluster in one distinct group. The results of the study reveal considerable genetic diversity among and within populations of P. colocasiae obtained from fine spatial scale. The possible mechanisms and implications of this genetic variation are discussed.  相似文献

Plant growth-promoting bacteria (PGPB) Pseudomonas lurida-NPRp15 and Pseudomonas putida-PGRs4 possessing multiple plant growth-promoting traits were isolated from rhizoplane of pea and rhizosphere of garlic, respectively. The effects of individuals and combinations of Pseudomonas spp. with effective root nodulating symbiotic nitrogen fixing Rhizobium leguminosarum-FB1 on plant growth, nutrient uptake and yield of the rajmash plant were studied under greenhouse conditions. Bacterial inoculation resulted in significantly higher values for plant dry biomass, N, P, K, Zn and Fe contents as compared to the uninoculated control. Furthermore, dual inoculation of P. lurida-NPRp15 with R. leguminosarum-FB1 significantly increased root and shoot dry weight, nodulation, nutrient uptake, pod yield, and nutrient content of pods of rajmash VL63 compared to controls, single and triple inoculation. The results of the study indicate the potential of harnessing the benefit of plant growth-promoting and nitrogen-fixing microorganisms to improve the growth and yield of rajmash.  相似文献

Russet Burbank potato plants grown in the greenhouse were given14C as14CO2 and as uniformly labelled sucrose. Labelled sucrose was applied; (1) by injection into the main stem, (2) injection with a hypodermic needle into the debladed petiole and (3) uptake of labelled sucrose kept in a vial fastened to the main stem using thread as a wick. Plants harvested 6 hours later were analyzed for radioactivity in the foliage, roots, and tubers. The greatest proportion of radioactivity was found in the foliage. Tubers were analyzed for radioactivity in the basic (amino acids), acidic (organic acids and sugar phosphates), neutral (sugars) and alcohol insoluble solid (starch) fractions. Most of the radioactivity was found in the sugar component of the chemical fraction. Plants treated with147CO2 had a higher percentage of radioactivity in the acidic and starch fractions than plants treated with sucrose. Sucrose applied by petiole injection not only showed higher total incorporation but also exhibited more translocation of labelled material into the tubers than the other methods.  相似文献

Context

Ecological processes that shape diversity and spatial pattern of ecological communities are often altered by disturbance. Spatial patterns (spatial autocorrelation) in species diversity are thus expected to change with disturbance.

Objective

When examining spatial patterns, ecologists traditionally lump positive and negative spatial autocorrelation into the overall spatial autocorrelation. By contrast, here we aim to understand disturbance effects on both positive and negative spatial autocorrelation of species richness and evenness, which may be related to environmental filtering and restricted dispersal, and to competition, respectively.

Methods

For 8 years, we monitored the spatial autocorrelation in species richness and evenness of riparian plant communities in both uncut control and experimentally clearcut sites in the boreal forest of Alberta, Canada. The overall spatial autocorrelation for each of these two indices of diversity was separately decomposed into the components of positive and negative spatial autocorrelations through eigendecomposition of the spatial weighting matrix.

Results

Negative spatial autocorrelation in richness and evenness were more pronounced in the clearcut than uncut sites, although positive spatial autocorrelations in all indices of diversity remained unchanged. Effect of disturbance was not detected on the overall spatial autocorrelation.

Conclusions

Disturbance increases negative spatial autocorrelation in species richness and evenness, with a stronger increase in evenness than richness, which underscores the importance of competition in structuring post-disturbance riparian communities. Our results also highlight the need for assessing positive and negative spatial autocorrelation and different aspects of diversity separately in understanding disturbance effects on the spatial pattern, or identifying processes from patterns.

The metacommunity concept provides a spatial perspective on community dynamics, and the landscape provides the physical template for a metacommunity. Several aspects of landscape heterogeneity, such as landscape diversity and composition, and characteristics of the matrix between habitat patches such as habitat connectivity, and geometry of habitat patches, may moderate metacommunity processes. These aspects of landscape heterogeneity are rarely considered explicitly in the metacommunity discussion, however. We propose landscape contrast (i.e., the average dissimilarity in habitat quality between neighboring patches) as a key dimension of landscape heterogeneity. The concept of landscape contrast unifies discrete and continuous landscape representations (homogeneous, gradient, mosaic and binary) and offers a means to integrate landscape heterogeneity in the metacommunity concept. Landscape contrast as perceived by the organisms affects several fundamental metacommunity processes and may thus constrain which metacommunity models may be observed. In a review of empirical metacommunity studies (n = 123), only 22 % of studies were explicit about their underlying landscape model assumptions, with striking differences among taxonomic groups. The assumed landscape model constrained, but did not determine, metacommunity models. Integration and explicit investigation of landscape contrast effects in metacommunity studies are likely to advance ecological theory and facilitate its application to real-world conservation problems.  相似文献
