Background

Canine mammary tumors (CMTs) are one of the most common malignancies in dogs and are associated with significant mortality. Serum tumor markers and non-coding microRNAs have gained widespread popularity in human oncology studies. The present study has two aims, first one is to investigate the miR-21 expression compared with changes in serum tumor markers (CEA and CA15-3) in CMT. The second aim is to detect the immunohistochemistry markers as vimentin, P63, and -SMA in CMT.

Methods

This study enrolled 17 female dogs: 10 with mammary tumors and seven controls without tumors. Blood samples were collected to measure miR-21, CEA, and CA 15-3, and histological samples were prepared for histological grading and immunohistochemistry.

Results

CA 15-3 was elevated in all animals, whereas CEA levels showed no change compared with controls. miR-21 was upregulated 12.84-fold in animals with CMT. The most frequently recorded CMT was the mixed type. Myoepithelial cells were identified by P63 immunoreactivity, but not SMA. High expression of miR-21 was observed with positive vimentin immunoreactivity, indicating the mesenchymal origin of the tumor cells.

Conclusion

The present study showed that miR-21 was elevated to a greater extent than CA 15-3 (12.84-fold vs. threefold). Tumors that was positive for vimentin immunoreactivity was also associated with an elevation in the levels of miR-21, showing that miR-21 is released from mesenchymal cells. These findings support the hypothesis that miR-21 may be a more sensitive, noninvasive indicator for CMT.

Two sponge-derived actinomycetes, Actinokineospora sp. EG49 and Nocardiopsis sp. RV163, were grown in co-culture and the presence of induced metabolites monitored by 1H NMR. Ten known compounds, including angucycline, diketopiperazine and β-carboline derivatives 1–10, were isolated from the EtOAc extracts of Actinokineospora sp. EG49 and Nocardiopsis sp. RV163. Co-cultivation of Actinokineospora sp. EG49 and Nocardiopsis sp. RV163 induced the biosynthesis of three natural products that were not detected in the single culture of either microorganism, namely N-(2-hydroxyphenyl)-acetamide (11), 1,6-dihydroxyphenazine (12) and 5a,6,11a,12-tetrahydro-5a,11a-dimethyl[1,4]benzoxazino[3,2-b][1,4]benzoxazine (13a). When tested for biological activity against a range of bacteria and parasites, only the phenazine 12 was active against Bacillus sp. P25, Trypanosoma brucei and interestingly, against Actinokineospora sp. EG49. These findings highlight the co-cultivation approach as an effective strategy to access the bioactive secondary metabolites hidden in the genomes of marine actinomycetes.  相似文献

Marine invertebrate-associated symbiotic bacteria produce a plethora of novel secondary metabolites which may be structurally unique with interesting pharmacological properties. Selection of strains usually relies on literature searching, genetic screening and bioactivity results, often without considering the chemical novelty and abundance of secondary metabolites being produced by the microorganism until the time-consuming bioassay-guided isolation stages. To fast track the selection process, metabolomic tools were used to aid strain selection by investigating differences in the chemical profiles of 77 bacterial extracts isolated from cold water marine invertebrates from Orkney, Scotland using liquid chromatography-high resolution mass spectrometry (LC-HRMS) and nuclear magnetic resonance (NMR) spectroscopy. Following mass spectrometric analysis and dereplication using an Excel macro developed in-house, principal component analysis (PCA) was employed to differentiate the bacterial strains based on their chemical profiles. NMR 1H and correlation spectroscopy (COSY) were also employed to obtain a chemical fingerprint of each bacterial strain and to confirm the presence of functional groups and spin systems. These results were then combined with taxonomic identification and bioassay screening data to identify three bacterial strains, namely Bacillus sp. 4117, Rhodococcus sp. ZS402 and Vibrio splendidus strain LGP32, to prioritize for scale-up based on their chemically interesting secondary metabolomes, established through dereplication and interesting bioactivities, determined from bioassay screening.  相似文献

The effect of N levels and sources on the antioxidant components in vegetable is not completely understood. Therefore, greenhouse experiments were conducted to determine the changes in antioxidant capacity diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP), total phenolics, ascorbic acid, total carotene and lycopene contents in edible portions of lettuce, onion and tomato due to different nitrogen levels and sources. The experiment included three N levels (20, 40 and 60 kg/ha for lettuce trial and 60, 120 and 180 kg/ha for either onion or tomato) and four nitrogen sources (calcium nitrate, ammonium sulphate, ammonium nitrate and urea). The antioxidant capacity and phenolic contents in lettuce was enhanced under low nitrogen level, while the antioxidant capacities, phenolics, carotenoids and lycopene in tomato fruits were not significantly affected by the nitrogen dose applied. Application of calcium nitrate caused 50% increase in DPPH and 20% in FRAP of lettuce compared to other nitrogen forms. Tomato treated with calcium nitrate or ammonium sulphate had higher phenolic and ascorbic acid contents than treated plants with ammonium nitrate or urea. Antioxidant tested parameters of onion were not significantly affected by the nitrogen treatments. Nitrogen concentration in tomato fruits significantly correlated with FRAP antioxidant capacity, phenolics, ascorbic acid, carotene and lycopene contents.  相似文献

Squamous cell carcinoma was recorded in 22 Nadji and 3 Maeimi sheep aged 3-6 years, of which 22 were females. Sixteen tumours involved the eyes, 7 occurred in the skin at different body sites, one developed in the ear and another in the rectum. In five cases secondary deposits were seen in the lymph nodes, nasal passages and upper lip. Surgical treatment was successful in the early stages of tumour development, but in advanced cases recurrences were noted. Histologically the tumours were typical for squamous cell carcinoma and most of them were well differentiated. Solar radiation was pointed out as a possible factor in the aetioloy of the tumour.  相似文献

Crude vegetable oils are usually oxidatively more stable than the corresponding refined oils. Tocopherols, phospholipids (PL), phytosterols, and phenols are the most important natural antioxidants in crude oils. Processing of vegetable oils, moreover, could induce the formation of antioxidants. Black cumin (Nigella sativa L.), coriander (Coriandrum sativum L.), and niger (Guizotia abyssinica Cass.) crude seed oils were extracted with n-hexane and the oils were further fractionated into neutral lipids (NL), glycolipids (GL), and PL. Crude oils and their fractions were investigated for their radical scavenging activity (RSA) toward the stable galvinoxyl radical by electron spin resonance (ESR) spectrometry and toward 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical by spectrophotometric method. Coriander seed oil and its fractions exhibited the strongest RSA compared to black cumin and niger seed oils. The data correlated well with the total content of polyunsaturated fatty acids, unsaponifiables, and PL, as well as the initial peroxide values of crude oils. In overall ranking, RSA of oil fractions showed similar patterns wherein the PL exhibited greater activity to scavenge both free radicals followed by GL and NL, respectively. The positive relationship observed between the RSA of crude oils and their color intensity suggests the Maillard reaction products may have contributed to the RSA of seed oils and their polar fractions. The results demonstrate the importance of minor components in crude seed oils on their oxidative stability, which will reflect on their food value and shelf life. As part of the effort to assess the potential of these seed oils, the information is also of importance in processing and utilizing the crude oils and their byproducts.  相似文献
