Effects of dietary selenium (Se) on immune competence of yellowtail kingfish, Seriola lalandi, were investigated. The fish were fed one of the three experimental diets including a control diet without Se supplementation and two diets supplemented with Se from Se‐yeast (Selplex) at 2 and 4 mg/kg. After feeding for 6 wk, the fish were challenged by injecting Vibrio anguillarum and observed for 2 wk. Dietary Se had no effect on feed intake, feed conversion ratio, and survival over the course of 6‐wk feeding; however, it significantly increased weight gain and Se content in muscle. Following the bacterial infection, the immune‐stimulating effects of Se were observed in antibody, lysozyme, and bactericidal responses, and there was a corresponding increase in survival and hematocrit by Se. Under infectious condition, antioxidant capacity of fish as measured in terms of resistance of red blood cells to peroxidation and glutathione peroxidase activity also increased by supplementation of Se. Liver necrosis and kidney melano‐macrophages were only seen in surviving fish fed the control diet after the challenge. Furthermore, there was evidence of myopathy in fish fed the diet without Se supplementation. This study suggests that Se, supplemented at 2 or 4 mg/kg, can improve growth and health of yellowtail kingfish.  相似文献

ABSTRACT:   Wild adult maturing and immature female Japanese flounder Paralichthys olivaceus were collected in June 2004 and January 2005, respectively, to clarify a possible role of gonadotropin-releasing hormones (GnRHs) in reproduction. Levels of salmon GnRH (sGnRH), chicken GnRH-II (cGnRH-II) and sea bream GnRH (sbGnRH) in the brain and pituitary were examined by time-resolved fluoroimmunoassay. Three forms of GnRHs were detected in the discrete brain at various levels. In the pituitary of both maturing and immature fish, sbGnRH was abundant together with a pronounced amount of sGnRH, whereas cGnRH-II was almost below the detectable limit. In maturing fish, levels of sbGnRH were high in the telencephalon, hypothalamus and pituitary, while levels of sbGnRH of immature fish were very low in these regions. These results indicate that sbGnRH is mainly responsible for gonadotropin secretion, and that sbGnRH in the anterior part of the brain is associated with gonadal maturation in the Japanese flounder.  相似文献

Somatic cell nuclear transfer (SCNT) technology has recently been used to generate animals with a common genetic composition. In this study, we report the derivation of a pluripotent embryonic stem (ES) cell line (SCNT-hES-1) from a cloned human blastocyst. The SCNT-hES-1 cells displayed typical ES cell morphology and cell surface markers and were capable of differentiating into embryoid bodies in vitro and of forming teratomas in vivo containing cell derivatives from all three embryonic germ layers in severe combined immunodeficient mice. After continuous proliferation for more than 70 passages, SCNT-hES-1 cells maintained normal karyotypes and were genetically identical to the somatic nuclear donor cells. Although we cannot completely exclude the possibility that the cells had a parthenogenetic origin, imprinting analyses support a SCNT origin of the derived human ES cells.  相似文献

The black‐lipped pearl oyster, Pinctada margaritifera, is the most important farmed species in French Polynesia and the basis of the most valuable export industry. Mass production of black pearls relies on a surgical operation requiring tissue from a donor pearl oyster to be grafted, together with a nucleus made of shell, into the gonad of a recipient oyster. Improving pearl size through family selection remains one of the main challenges for future aquaculture development. This study analyses the relative contribution of donor and recipient oysters to pearl size. To this end, hatchery‐produced donor oysters of two batches, large and small (based on shell height), were used to supply grafts for recipients, which were then monitored individually for their growth performance by recording shell height, width, and thickness, and total live weight (flesh + shells) every 6 months (four biometric measurement times) over 20 months of culture. Pearls issued from the two batches of donors showed no significant differences in nacre weight or thickness. In contrast, recipient oyster shell height and total weight were increasingly positively correlated with these pearl size parameters over the culture period, becoming significant at 8 months post‐grafting. Potential therefore exists to use shell height and oyster weight as phenotypic indicators for selective breeding of recipient oysters with high growth performance to increase pearl size in P. margaritifera.  相似文献

Sexual dimorphism is widely observed in almost all farmed aquatic animal species but giant freshwater pawn (GFP) is unique, with males characterized by three main morphotypes (blue claw, orange claw and small males) and females by different reproduction status (ovary, berried egg and already‐spawned females). There has been reported evidence that the effect of male morphotype may have masked genetic variation in growth‐related traits, as a result the heritability for male body weight was lower than that estimated in female. A pending question has arisen whether selection should be made in female only. To answer this question, we used an 8‐year data set from a long‐term selection programme (2008–2015) for high growth in this species comprising 106,756 individuals that were offspring of 515 sires and 810 dams. The body weight data of female and male GFP or of each morphotype was treated as a separate trait and a multi‐trait approach was used to estimate genetic correlations for homologous traits between sexes and between morphotypes. Our analysis showed that there were little differences in the heritability estimates between female and male. In female, mature ovary individual displayed higher heritability than berried egg and already‐spawned females. For male, the heritability for blue claw, orange claw and small males were 0.11, 0.06 and 0.00 respectively. Between‐sex genetic correlation was moderate (0.55 ± 0.11) for body weight, suggesting that the trait expressions in female and male may be genetically different. In female, the genetic correlations for body weight among three female types were close to one (0.91–0.94). In contrast, the genetic correlations for body weight between male morphotypes especially between blue claw or orange claw and small males were low (0.15–0.25). Furthermore, we estimated genetic gain as the difference in least square means (LSM) or estimated breeding values (EBV) between the selection line and control group. The genetic gain in body weight was smaller in females than in males. It is concluded that there is no need to run separate breeding programme for female and male GFP. A combined selection using both female and male data can achieve selection response for body weight as demonstrated in the present study.  相似文献

Cultured pearl nacre deposition rate in Pinctada margaritifera oysters was studied to examine its influence on Tahitian classification grade and colour and to further our understanding of using selected donor oysters produced in a hatchery system for the improvement of these traits. A large-scale grafting experiment using 150 farmed donor oysters was realised in Rangiroa atoll (French Polynesia). After 18 months of culture, 874 pearls were harvested and 7 pearls quality traits were recorded: (1) nacre deposition rate assessed by nacre thickness and weight, (2) classification grade, lustre and surface defects, and (3) colour assessed by pearl colour darkness level and visual colour categories. Nacre deposition rate significantly influence pearl grade, surface defects, darkness level and colour categories, but not the lustre. Grade A pearls had the thickest and heaviest nacre on average, compared with grade D pearls and rejects. In addition, pearls without any surface defect had thicker and heavier nacre. Pearls with low darkness were the thinnest and the lightest. Pearls of aubergine–peacock secondary colours were the thickest and the heaviest. These results highlight the importance of rapid nacre deposition rate for increasing the production of grade A pearls, with less surface defects and dominance of a black tone colour. Fast nacre deposition may, however, have a negative impact on other pearl quality traits, such as pearl lustre. This study demonstrates the importance of examining relationships between pearl quality traits in a way that provides solid information that can be directly used for breeding and genetic selection programmes.  相似文献

Cultured pearls produced with Pinctada margaritifera, using the surgreffe method (implantation of a second nucleus following pearl harvest) were studied for the first time to: (1) examine family effect on nacre thickness, nacre weight and nacre deposition speed and (2) compare variation in these three traits with that obtained from the cultured pearls previously harvested after the corresponding initial grafts. A surgreffe experiment using 783 recipient oysters was realized in Rangiroa atoll (French Polynesia). After 24 months of culture, 389 cultured pearls were harvested. Significant donor family effect was found for the harvested pearl rate from surgreffe (= 0.046). Highly significant donor family effect was recorded for nacre thickness (= 0.004). Very highly significant donor family effects were recorded for nacre weight and nacre deposition speed (< 0.0001). Comparison between surgreffe and initial graft showed: (1) no significant effect for the average cultured pearl rate harvested (= 0.052) and average cultured pearl nacre deposition speed (= 0.622) and (2) very highly significant differences (< 0.0001) for the average cultured pearl nacre thickness and nacre weight. This study highlighted three major implications for pearl industry management: (1) donor family effect was maintained from initial graft to surgreffe, for nacre thickness, weight and deposition speed, (2) the persistence of the pearl sac metabolic activity over three years of culture and (3) the relation between harvested pearl rate and the size of the nucleus inserted in the pearl sac.  相似文献

Genomic selection (GS) is a statistical and breeding methodology designed to improve genetic gain. It has proven to be successful in animal breeding; however, key points of difference have not been fully considered in the transfer of GS from animal to plant breeding. In plant breeding, individuals (varieties) are typically evaluated across a number of locations in multiple years (environments) in formally designed comparative experiments, called multi‐environment trials (METs). The design structure of individual trials can be complex and needs to be modelled appropriately. Another key feature of MET data sets is the presence of variety by environment interaction (VEI), that is the differential response of varieties to a change in environment. In this paper, a single‐step factor analytic linear mixed model is developed for plant breeding MET data sets that incorporates molecular marker data, appropriately accommodates non‐genetic sources of variation within trials and models VEI. A recently developed set of selection tools, which are natural derivatives of factor analytic models, are used to facilitate GS for a motivating data set from an Australian plant breeding company. The power and versatility of these tools is demonstrated for the variety by environment and marker by environment effects.  相似文献
