Tuna and mahi-mahi are two major fish species responsible for histamine poisoning. This research developed a rapid ultra-high performance liquid chromatography (UHPLC) method to determine amino acids, histamine, and other biogenic amines that can act as co-indicators of histamine poisoning in tuna and mahi-mahi. The modified UHPLC method could simultaneously determine four biogenic amines and 10 major free amino acids in mahi-mahi (Coryphaena hippurus) and yellowfin tuna (Thunnus albacares) within 17.5 min. This UHPLC method showed good linear response, sensitivity, resolution, recovery, repeatability, and number of theoretical plates. The concentrations of detected amino acids, biogenic amines in mahi-mahi and tuna, and their relationships with fish spoilage grade were determined by this UHPLC method. The developed UHPLC method is a rapid and accurate way to monitor quality changes of mahi-mahi and tuna by inspecting the changes of amino acids and biogenic amines.  相似文献

It has been stated that central injection of ghrelin is acting as an anorexigenic peptide in chicken. Ghrelin activity was studied through some neuronal pathways. The present study was designed in 4 experiments to examine the hypophagic response of ghrelin through the central serotonergic system in chicken. The guide cannula was surgically implanted in the right lateral ventricle of the chickens. In experiment 1, intacerebroventricular injection with PCPA (1.5 mg) performed followed by ghrelin (0.6 nmol). In experiments 2, 3 and 4 prior to ghrelin injection, chickens received fluoxetine (10 μg), 8-OH-DPAT (15.25 nmol), SB242084 (1.5 μg) respectively via guide cannula intacerebroventricularly. Cumulative food intake was determined at 3 h post injection. The results of this study showed that flouxetine pretreatment significantly amplified ghrelin hypophagia in chicken (p?<?0.05). The hypophagic effect of ghrelin was attenuated by pretreatment with PCPA and SB242084 (p?<?0.05) but 8-OH-DPAT had no effect. These results suggest that hypophagic effect of ghrelin probably is mediated by serotonergic mechanisms via 5-HT2C receptor.  相似文献

This study was conducted to investigate the effect of different methods of using probiotics Bacillus subtilis and Bacillus licheniformis in soya bean‐based diets on the physiology of beluga (Huso huso) (351 ± 4 g). Five experimental diets were formulated to feed beluga for 10 weeks. The results showed that there was no significant difference in weight gain between control (446 g) and 70%SBM‐SPRAY2 (409 g) treatments. Long‐chain polyunsaturated fatty acids in the muscle of beluga fed control (203.2 g/kg) and 70%SBM‐SPRAY2 (166.4 g/kg) diet did not change statistically. Blood performance as a newly introduced factor in individuals fed dietary control (64.06) and 70%SBM‐SPRAY2 (64.33) had higher value as compared with those fed dietary 70%SBM‐DIET2 (61.90) (p < 0.05). Also, total protein (1.68 g/dl) and RBC (0.74 × 106/mm3) in fish fed 70%SBM were statistically lower than other treatments (p < 0.05). Lysozyme, alternative complement activity and total viable counts in beluga fed dietary probiotics were significantly higher than others. In conclusion, feeding fish with the 70%SBM‐SPRAY2 diet improved fish growth and health.  相似文献

Tomato yellow leaf curl virus (TYLCV) can cause an economically relevant disease in the major tomato growing regions, and no resistance has been identified in commercial and wild tomato cultivars in Iran. In the present study, 34 common bean lines were screened for their reaction to TYLCV-Ir2 under greenhouse conditions. The experiment was conducted in a randomized complete block design with three replications. Bean plants were inoculated at the first trifoliate stage with viruliferous whiteflies (Bemisia tabaci biotype B) and were then sprayed with an insecticide and maintained in whitefly-proof greenhouse. The inoculated plants were monitored for the development of symptoms and the presence of viral DNA by polymerase chain reaction (PCR) four weeks after inoculation. Results revealed that out of the 34 lines, five were resistant. These lines exhibited either no symptoms or very mild symptoms and no viral DNA was detected in them by PCR. Two lines, which showed no typical disease symptoms but contained viral DNA, were identified as tolerant to TYLCV-Ir2. The vector feeding preference for common bean lines was assessed. Results indicated a significant difference in adult whitefly numbers among bean lines but there was no relationship between the number of whiteflies and disease symptom severity. The resistance to TYLCY-Ir2 expressed in common bean lines may be useful as source of resistance for the development of resistant commercial common bean cultivars.  相似文献
