The potential of Tetragenococcus halophilus as a starter culture for flavor improvement in fish sauce fermentation was elucidated. Four strains of T. halophilus isolated from fish sauce mashes were inoculated to anchovy mixed with 25% NaCl with an approximate cell count of 10(6) CFU/mL. The α-amino content of 6-month-old fish sauce samples inoculated with T. halophilus was 780-784 mM. The addition of T. halophilus MRC10-1-3 and T. halophilus MCD10-5-10 resulted in a reduction of histamine (P < 0.05). Fish sauce inoculated with T. halophilus showed high contents of total amino acids with predominantly high glutamic acid. Major volatile compounds in fish sauce were 2-methylpropanal, 2-methylbutanal, 3-methylbutanal, and benzaldehyde. T. halophilus-inoculated fish sauce samples demonstrated the ability to reduce dimethyl disulfide, a compound contributing to a fecal note. The use of T. halophilus for fish sauce fermentation improves amino acid profiles and volatile compounds as well as reduces biogenic amine content of a fish sauce product.  相似文献

The conditions of isolating protein from tilapia frame by-products (TFB) by a pH-shift process were investigated along with their physicochemical and gelling properties. The minimum solubility of TFB protein was observed at pH 5.5 and gradually increased at both acid and alkaline sides. The highest solubility was observed at pH 2 and 12. Protein solubility of TFB increased as the ratio of alkaline extraction medium and extraction time increased. The highest protein extraction was found at the ratio of ground sample to extraction medium of 1 to 9 and extraction time of 15 min. The maximum protein recovery was found at pH 12 with 19.19%. The color of all protein isolate samples obtained from either acid or alkaline extraction was dark brown. Whiteness of a TFB protein isolate extracted at pH 2 was the highest with a value of 14.69. An alkaline pH-shift process effectively removed fat as much as 95%. Breaking force and deformation of recovered protein gel from an alkaline pH-shift were higher than those from the acid counterpart.  相似文献

Although silver carp bones (SCB) are generated as waste, they are a natural source of calcium and have the potential to be a food ingredient. When the SCB had been soaked in hot sodium hydroxide (0.8%), autoclaved, and ground into SCB powder (SCBP), the total calcium content, analyzed by inductive coupled plasma-optical emission spectroscopy, was about 32%. X-ray absorption near-edge structure (XANES) spectroscopy specified hydroxyapatite as the major calcium component. The soluble calcium in SCBP was found to be 0.59% in de-ionized water. SCBP induced the cross-linking of proteins in minced fish after incubation at 40°C, as indicated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Fortification of SCBP at 0.0, 0.5, and 1.0% (w/w) in fish emulsion sausage resulted in a decrease of moisture content, while the fat and protein contents increased slightly. Total ash content in the sausage increased significantly, and total calcium content increased 15-fold in fortified SCBP. The hardness and gumminess values of sausage were improved with the addition of SCBP, while the microstructure was not altered by SCBP.  相似文献

Fish liver transglutaminase (FTG), a Ca(2+)-dependent enzyme, exhibits different characteristics from the Ca(2+)-independent microbial transglutaminase (MTG), leading to potential differences in their substrate specificity and reactivity. The ability of these enzymes to catalyze isopeptide bond formation by incorporating 5-(biotinamido)pentylamine (BPNH2) into peptides derived by tryptic digestion of threadfin bream (TB)-myosin was investigated to identify reaction sites and substrate specificity using a peptidomic strategy. BPNH2 was incorporated into TB-myosin peptides to a greater extent by MTG than FTG. Peptides derived from TB-myosin heavy chain (MHC) shared highest similarity to amberjack-MHC on the basis of a Mascot database search. Amino acid sequences and modification sites of BPNH2-tagged peptides were identified by tandem mass spectrometry based on the amberjack-MHC sequence. The BPNH2 modification sites catalyzed by both TGases were at the myosin rod. Most of the BPNH2 peptides contained charged amino acids (E, R, K) at the glutaminylamide site of reactive glutamine (Q*). The alpha-acrylamide site of Q* contained E, F, or L on peptides catalyzed by both enzymes, I, Q, or A on peptides catalyzed only by FTG, and V on a peptide catalyzed only by MTG. These results demonstrate the different structural requirements for glutaminyl substrates between these two enzymes.  相似文献

Upland rice production plays an important role in both household consumption and crop rotation. Until now, a blast resistant upland variety has not been released in Thailand. The bimodal pattern of rain distribution in Thailand’s upland rice production areas create a favorable environment for the outbreak of leaf blast when seedling-tillering, and neck blast within the heading stage. The use of genetically resistant cultivars has proven to be an effective way to cope with this problem. In this study, 256 indigenous upland rice plants were screened for blast resistance under greenhouse and field conditions. Ten indigenous upland rice varieties, ULR292, ULR242, ULR219, ULR162, ULR161, ULR134, ULR109, ULR098, ULR081, and ULR066, were identified as resistant to leaf blast disease in both natural infection and artificial inoculation, under greenhouse conditions. Additionally, six of the ten varieties, ULR162, ULR161, ULR134, ULR109, ULR098, and ULR081, were found to be resistant to neck blast under field conditions. These new sources of blast resistance identified from indigenous upland rice varieties proved more resistant than the check varieties, depicting their potential for further use in Thailand’s rice blast resistance improvement program.  相似文献

Small-scale mud carp (SC, Cirrhina microlepis) and common carp (CC, Cyprinus carpio) are underutilized freshwater fish. The objective of this study was to elucidate the gel-forming ability of mince and washed mince from these species in relation to washing cycles and CaCl2 addition. Protein loss of up to 67% was observed after three-washing cycles of both species, rendering relatively low yield. About 85–90% of total fat was removed from SC flesh after three-washing cycles, but only two-washing cycles were sufficient for fat removal in CC mince. Mince and washed mince of both species did not exhibit severe proteolysis with low autolytic activity at 65°C. SC exhibited superior gel-forming ability to CC. Two- and three-washing cycles in conjunction with a 40°C-preincubation resulted in the highest textural properties of SC and CC, respectively. CaCl2 increased the breaking force of CC and SC mince gels up to 65 and 95% at 0.3 and 0.5% CaCl2, respectively, as compared to without CaCl2. However, it showed negative effects on surimi gels of both species. Both SC and CC are potential freshwater fish resources for mince and washed mince production with reasonable good gel-forming ability.  相似文献

Exserohilum turcicum causes northern corn leaf blight (NCLB), an important disease occurring in maize producing areas throughout the world. Currently, the development of cultivars resistant to E. turcicum seems to be the most efficient method to control NCLB damage. Marker-assisted selection (MAS) enables breeders to improve selection efficiency. The objective of this work was to identify random amplified polymorphic DNA (RAPD) and sequence characterized amplified region (SCAR) markers associated with NCLB resistance. Bulked segregant analysis (BSA) was used to search for RAPD markers linked to NCLB resistance genes, using F2 segregating population obtained by crossing a susceptible inbred ‘209W’ line with a resistant inbred ‘241W’ line. Two hundred and twenty-two decamer primers were screened to identify four RAPD markers: OPA07521, OPA16457, OPB09520, and OPE20536 linked to NCLB resistance phenotype. These markers were converted into dominant SCAR markers: SCA07496, SCA16420, SCB09464, and SCE20429, respectively. The RAPD and SCAR markers were developed successfully to identify NCLB resistant genotypes in segregating progenies carrying NCLB resistant traits. Thus, the markers identified in this study should be applicable for MAS for the NCLB resistance in waxy corn breeding programs.  相似文献

The planting of upland rice is one cropping option in area with limited water availability and low soil fertility in North and Northeast Thailand.The varietal selection was determined by grain yield potential,wide adaptation,and good stability.This study was aimed at evaluation of indigenous upland rice germplasm for yield and yield stability in multi-locations.Thirty-six upland rice genotypes collected from six provinces of the North and Northeast Thailand and one check variety(Sewmaejan) were assessed under five locations in the rainy seasons of 2009 and 2010.The experiment was laid out in a randomized complete block design with three replications.The genotype grain yield was highly affected by location(59.90%),followed by genotypes(G)×location(L) interaction(12.80%) and genotype(6.79%).The most suitable location for the genotype evaluation was L3(Khon Kaen-KKU10) which associated with stability of grain yield for all genotypes.Furthermore,biplot and regression analysis indicated that genotype numbers 6(Jaowmong 1),10(Neawmong 1),18(Neawdum 1),19(Leamna),20(Prayaleamkang),32(Kunwang 2),and 33(Kunwang 3) showed great yield stability over five locations.The genotypes will be applicant for upland rice production area and parental base in breeding program.  相似文献

ABSTRACT

Changes in chemical parameters of tropical fish used for surimi production—namely, threadfin bream (TB; Nemipterus spp.), lizardfish (LZ; Saurida spp.), and goatfish (GF; Upeneus spp.)—during ice storage and their respective washed mince were investigated. Total volatile basic nitrogen (TVB-N), trimethylamine (TMA), hypoxanthine (Hx), and total biogenic amine (BA) content of mince increased with storage time. Crude protein of fish and their respective washed mince reduced as storage time of fish was prolonged. Principal component analysis (PCA) revealed that washed mince produced from fish stored in ice for 7–12 days showed chemical characteristics that were distinct from those of washed mince prepared from spoiled fish. The TMA, TVB-N, and Hx content of washed mince appeared to be sufficient raw material freshness indicators in these three species.  相似文献
