Large quantities of high protein fish meals are needed to sustain cultured species and thus the impact to marine ecosystem has been highly discussed. The aim of this study was to apply a PCR‐cloning methodology for a robust insight into the composition of commercial fish meals and feeds for farmed species of the Greek mariculture, assessing the risk posed by aquaculture to marine ecosystems but also the risk posed by commercial fish feeds to the increase in trophic level of species farmed in Greece. 89% of the sequences were identified to species level and only 11% to genus/family level. Overall, a total of 49 taxa were identified (44 fish species/taxon, five non‐fish species/taxon). Even though small pelagic fish like Engraulis sp. were the main portion, a wide range of species constituted the fish meals and feeds. Plant and animal species were also detected as an alternative protein source. Feed products employed in Greek mariculture still contain large portions of fish meals which increase the mean trophic level of farmed species causing a farming up trend. The results emphasize that such molecular methodologies are needed to certify aquafeeds allowing fish feed producers to demonstrate their commitment to sustainable aquaculture.  相似文献

A double-DNA approach was developed to discriminate the three Trachurus species that abide in European waters: T. trachurus, T. mediterraneus, and T. picturatus. The analysis aimed at both mitochondrial and nuclear loci. Polymerase Chain Reaction (PCR) amplification of the cytochrome b gene of mtDNA was followed by restriction analysis with three species-specific enzymes: NlaIII, NciI, and BsmAI. Digestion with these endonucleases yielded species-specific electrophoretic profiles. The universality of the results was verified by screening a large number of individuals from 12 geographical regions covering most of the distribution of the species. Additionally, the nuclear multicopy 5S rRNA gene was selected as an alternative candidate for the discrimination of the three Trachurusspecies. A simple agarose gel electrophoretic analysis of the amplicons proved to be capable of leading to unambiguous identification of the three Trachurus species. Thus, the double-DNA methodology presented here allows the accurate discrimination of Trachurus fish species and the detection of commercial fraud.  相似文献

A DNA methodology was developed to discriminate fry of six Mugilidae species found in the Mediterranean, namely Mugil cephalus, Mugil so‐iuy, Chelon labrosus, Liza aurata, Liza ramada and Liza saliens. Polymerase chain reaction (PCR) amplification of the 5S rDNA gene was used for the identification of the above six species. PCR products of two species showed different patterns on EtBr‐stained agarose gels; M. so‐iuy gave a pattern of three bands, while L. saliens gave a pattern of one band. Mugil cephalus, C. labrosus, L. aurata and L. ramada gave a pattern of two bands. Subsequent sequencing analyses revealed unique haplotypes for each of the remaining four species. This is a genetic technique that could be applied in hatcheries, for identification of fry mullet's species. Furthermore, 5S rDNA sequences of each of five of the studied species (i.e. M. cephalus, C. labrosus, L. aurata, L. ramada and L. saliens), as well as a sequence of Oedalechilus labeo that belongs to the Mugilidae family, were aligned and used for the examination of the phylogenetic relationships among them. Phylogenetic trees produced in our study are in general agreement with those presented in the literature. This is the first study to use a nuclear marker to elucidate the phylogeny of Mugilidae species.  相似文献

This study investigated the effects of dietary inclusion of soya bean oil combined with fish oil (SFO) on the activities of a) superoxide dismutase (SOD), glutathione reductase (GR), catalase (CAT) and glutathione transferase (GST) in blood plasma and b) SOD, GR, CAT and lactoperoxidase (LPO) in the milk of sheep and goats. Furthermore, the oxidative stress indicators for measuring total antioxidant activity and free radical scavenging activity [ferric reducing ability of plasma ( FRAP) and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulphonic acid) (ABTS) assays] and oxidative stress biomarkers [malondialdehyde (MDA) and protein carbonyl (PC)] were also determined in the blood plasma and milk of the animals. For this purpose, twelve dairy sheep and twelve dairy goats were assigned each to two homogenous subgroups. Treatments in both animal species involved a control diet without added oil and a diet supplemented with 5% soya bean oil and 1% fish oil. The results showed that the inclusion of SFO in the diets of sheep and goats increased significantly the activities of CAT and GR in their blood plasma. The same effect was observed for the activities of GST and FRAP in the blood plasma of goats. Moreover, the fact that the goats had significantly higher average daily PUFA intake (3.62 g/kg BW0.75) compared to sheep (2.51 g/kg BW0.75) resulted in an enhancement in the MDA content in their plasma. A significant increase in CAT activity in the milk in both animal species fed with SFO diets was also found. Finally, due to the higher apparent transfer rate of n‐3 FA from the diet to the milk in sheep, the PC concentrations were found to be enhanced in their plasma and milk. In conclusion, the impact of dietary SFO supplementation on the oxidative status of body and/or on the milk of small ruminants depends not only on the daily PUFA intake, but also on the amount of n‐3 FA that reach their milk.  相似文献

The use of UV absorbing films as greenhouse cover material is spreading out in protected cultivation. Although their effects on pest and disease management have received much attention, few studies focus on their effects on the crop. This study aims at assessing the consequences of UV absorbing film on the behaviour and production of an eggplant crop by comparing two different UV absorbing films (0 and 3% UV transmittance) to a standard polyethylene film (5% UV transmittance). Results show that the eggplants grown in the greenhouse with 0% transmission to UV light are about 21% taller and have about 17% higher leaf product (leaf length × width) than the plants grown in the greenhouse with 5% transmission to UV light. Finally, given that the production was slightly increased in quantity (20%) and quality (bigger fruits) in the greenhouse with absence of UV light compared to that with 5% transmission coefficient, it can be concluded that growing soilless eggplant under UV-absorbing material can be achieved with the same or better results as under standard covering material. Any other enhancement that the UV-absorbing film will bring (lower pest and disease impact on the crop, lower pesticide load and costs) will therefore be to the benefit of the grower.  相似文献

DNA analysis of hake products commercialized in southern European (Spanish and Greek) market chains have demonstrated more than 30% mislabeling, on the basis of species substitution. Tails and fillets were more mislabeled than other products, such as slices and whole pieces. African species were substitute species for products labeled as American and European species, and we suggest it is a case of deliberate economically profitable mislabeling because real market prices of European and American hake products are higher than those of African in Spanish market chains. The presented results suggest fraud detection that disadvantages African producers. Government-mandated genetic surveys of commercial hakes and the use of subsequent statements of fair trade on labels of seafood products could help to reduce fraud levels in a global market of increasingly conscious consumers sensitive to ethical issues.  相似文献

Several policy instruments have been proposed in order to minimize the negative impacts of local communities' activities on forests. The present paper presents an empirical survey conducted in a Greek forest area on the island of Lesvos, focusing on citizens' perceptions of alternative management scenarios along with the role of social factors on these perceptions. Our results demonstrate that citizens are supportive of policies which are not solely state-based. Furthermore, higher levels of local social capital are positively correlated with citizens' perceptions in favor of proposed co-management policies.  相似文献

Twenty‐four dairy sheep and goats, respectively, were assigned each to three homogenous subgroups per animal species and fed the same diet in quantities which met 70% (underfeeding), 100% (control) and 130% (overfeeding) of their energy and crude protein requirements. The results showed that the underfed sheep in comparison with the control had significantly lower glutathione reductase (GR), superoxide dismutase (SOD), catalase and glutathione peroxidase (GPX) activities and total antioxidant capacity (measured with Ferric Reducing Ability of Plasma [FRAP] assay) in their blood plasma. A significant increase in the glutathione transferase (GST) and GPX activities, malondialdehyde content and total antioxidant capacity (measured with 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulphonic acid) [ABTS] assay) in the blood plasma of underfed goats compared with controls was observed, while the opposite happened for the GR and SOD activities. The underfeeding in both animal species caused a significant increase in the protein carbonyls (PC) content of their blood plasma. The overfeeding, compared with the control, caused a significant decline in the GPX activity and total antioxidant capacity (measured with FRAP) in the blood plasma of sheep while the opposite happened for the GPX and GST activities in the case of goats. The overfed animals, of both species, compared with the respective controls, had higher PC content in their blood plasma. The feeding level had no noticeable impact on the antioxidants' enzymes activities of milk in both animal species. Moreover, the underfeeding in the blood plasma and the overfeeding in milk of both animal species resulted into a significant increase in the PC content. Finally, only in sheep milk, the underfeeding, compared with the respective control, and overfeeding reduced significantly the total antioxidant capacity (measured with ABTS). The feeding level caused oxidative stress in both organism and milk but the response was different in animal species and needs further investigation.  相似文献
