We have shown in vitro that mechanical stretch triggers activation of quiescent satellite cells of skeletal muscle to enter the cell cycle through an intracellular cascade of events including nitric oxide (NO) synthesis that results in the release of hepatocyte growth factor (HGF) from its extracellular association and its subsequent presentation to signaling receptors. In order to explore the activation mechanism in vivo, stretch experiments were conducted in the living animal using our suspension model developed. This system used the weight of the hind portion of rats to stretch the inside muscles of the left hind limb suspended for a period of 0.5–2.0 h. At the end of the stretch period, the rats received an intraperitoneal injection of bromodeoxyuridine followed by immunocytochemistry for its incorporation as an index of satellite cell activation in vivo. Depending on the period of stretch, bromodeoxyuridine labeling was increased significantly over the contralateral unstretched leg or control muscle from untreated rats. A stretched muscle extract prepared from the 2 h stretched tissue by incubating it in PBS, showed the active form of HGF as revealed by immunoblotting and it could stimulate the activation of unstretched satellite cells. Also, administering NO synthase inhibitor L‐NAME prior to muscle stretch abolished the stretch activation of satellite cells. Therefore, the results from these experiments demonstrate that stretching muscle triggers NO synthesis and HGF release, which could activate satellite cells in vivo.  相似文献

A novel Bacillus species of Calculus Bovis (cow gallstone) was isolated and identified in this study. Morphological features, bacterial fatty acid analysis using a microbial identification system, carbon source utilization profiles using Biolog system and 16S rDNA sequencing were employed to identify the isolated bacterium. This isolated bacterium was observed to be Gram‐negative, aerobic growing, rod‐shaped and short chain. The results of bacterial fatty acid analysis and physiological characteristics were not matched to the database. The main fatty acids found in the bacterium were 65.96% branched chain saturated fatty acids (iso C11:0, anteiso C11:0 and iso C13:0~anteiso C19:0). The bacterium oxidized 35 carbon sources and weakly responded with 49 of the 95 different carbon sources analyzed with the Biolog identification system. Based on 16S rDNA sequence analysis, this bacterium was classified as a novel Bacillus species.  相似文献

In order to produce angiotensin I‐converting enzyme (ACE) inhibitor for application in functional food, chicken bones were gathered from a meat processing factory and then hydrolyzed with Alcalase, pepsin and trypsin for 12 h. The hydrolysates were lyophilized, stored at ?80°C and tested experimentally every 2 h for pH value, peptide content, degree of hydrolysis (DH), electrophoresis and activity of ACE inhibitor. The hydrolysates of Alcalase had the highest peptide content and DH. The components of more than 66 kDa had disappeared in hydrolysates of Alcalase and trypsin after 2 h of hydrolysis. The hydrolysates of Alcalase were more active in inhibiting ACE, especially when hydrolyzed at 4 and 8 h, and also had low IC50 values of 1.960 and 0.945 mg/mL. According to the results of DH and electrophoresis, the higher activity of ACE inhibitor is assumed to be derived from the low molecular peptides in hydrolysates of Alcalase. Chicken leg bone has a high potential to be utilized to develop ACE inhibitory peptides as a potential ingredient of functional food intended to alleviate hypertension.  相似文献

The effects of organic acids on the microbial quality of Taiwanese‐style sausages were studied. Pork meat from a Taiwan retail market was decontaminated with various organic acids (citric, lactic and tartaric acid), then, the raw meat was used to make Taiwanese‐style sausages and stored from 0 to 40 days at 4°C. The total plate counts, lactic acid bacteria, Micrococcus, Listeria monocytogenes, Hunter‐L and Hunter‐a values were determined. The total plate counts of the control group were initially greater than those of the treated groups. Higher incidence rates of L. monocytogenes in the products were obtained from the control group, but were not detected in the treated groups during storage. Lactic acid bacteria counts following the lactic acid treatment were lower than those of the other groups. Micrococcus counts of the controls increased by 0.6–1.2 log10 colony‐forming unit (CFU)/g greater than those of treated groups throughout storage at 4°C. The light color (L‐value) of the control group gradually decreased during storage. Pork meat dipped in various organic acids was found to be suitable to extend the shelf‐life and improve the microbiological quality of Taiwanese‐style sausages.  相似文献

The purpose of the study was to investigate bioactive compounds of in vitro cultured Calculus Suis and natural Calculus Bovis obtained as valuable by‐products from animals used for meat production. The results showed that the components of natural Calculus Bovis were rich in bilirubin and biliverdin and had higher content of essential amino acids. The major amino acids of in vitro cultured Calculus Suis were identified as glycine, alanine, glutamic acid and aspartic acid, and those for natural Calculus Bovis were found to be glutamic acid, aspartic acid, proline, and arginine. The methionine and cysteine contents of precursors for glutathione in natural Calculus Bovis were significantly higher than those of in vitro cultured Calculus Suis. The mineral contents of zinc, iron and manganese of natural Calculus Bovis were significantly higher than those of in vitro cultured Calculus Suis. The major bile acids in both products were cholic acid and dehydrocholic acid, respectively. The chenodeoxycholic and ursodeoxycholic acid content of in vitro cultured Calculus Suis was significantly higher than that of natural Calculus Bovis.  相似文献

ABSTRACT:   In order to establish genetic signatures for intraspecific identification, parts of the genes rbcL , rbcS and psbA and their intergenic spacer (IGS) regions were amplified by polymerase chain reaction (PCR) from the chloroplast genomes of Heterosigma akashiwo strains isolated from Japan. A transfer RNA-Leu gene trnL and a hypothetical gene cfxQ , which is related to ribulose bisphosphate carboxylase/oxygenase expression, were found in the upstream region of the rbcL gene and in the rbcS-psbA IGS region, respectively. All the gene-coding regions and the IGS regions between rbcL and rbcS showed the same sequences among the strains tested. In contrast, the rbcL upstream regions and the rbcS-cfxQ IGS regions showed some differences such as nucleotide substitutions, duplications and inversions between NIES-5 and the other strains. Based on these sequence data, five genetic signatures were established and their simple and rapid detection by means of strain-specific PCR primers and PCR-restriction fragment length polymorphism techniques was examined. The results suggested the usefulness of these genetic signatures and techniques for the discrimination of H. akashiwo populations.  相似文献

Numerous attempts have been made to develop angiotensin I converting enzyme (ACE) inhibitors from various sources of food protein. Generally chicken leg bones are discarded after industrial chicken meat processing without any substantial benefit. In previous studies, chicken leg bone proteins were hydrolyzed by various enzymes and the results demonstrated that Alcalase hydrolysates have considerable ACE inhibiting activities. In this study, the best ACE inhibitory hydrolysate (A4) (which was derived from chicken leg bone protein by Alcalase after 4 h incubation) was orally administrated (50 mg/kg bw) in spontaneously hypertensive rats (SHRs) to investigate its antihypertensive effects. After oral administration of A4, a maximal reduction activity of about 26 mmHg was found at 4 h and maintained to 8 h. Moreover, SHRs treated with A4 (50 mg/kg bw/day) for eight weeks exhibited a reduction in systolic blood pressure, which is as significant as the effects of Captopril ( P  < 0.05). These results suggested that chicken leg bones have a high potential for utilization to develop ACE inhibitors as potential food ingredients intended to alleviate hypertension.  相似文献
