Bovine tuberculosis is a contagious and zoonotic disease of animals and humans. In Europe, the number of reported cases of tuberculosis has decreased. Equidae are relatively rarely infected even in endemic areas. The presented report describes a case of chronic Mycobacterium bovis tuberculosis in a 30-year-old female donkey. The donkey initially presented with persistent lymphadenopathy; however, as the disease progressed, weight loss became apparent. To the authors' knowledge, this is the second confirmed case of tuberculosis in a donkey in Europe.  相似文献

The current experiment was carried out to determine whether exogenous GnRH treatment in primiparous, lactating sows undergoing feed restriction would improve reproductive performance after weaning. Sows were allocated to one of three treatments: AA sows (n = 8) were fed to appetite throughout a 28-d lactation, AR (n = 12) and AR + GnRH (n = 12) sows were fed as AA sows from farrowing to d 21 of lactation, and feed intake was reduced to 50% of the ad libitum intakes from d 22 to 28. The AR + GnRH sows received 800 ng of GnRH i.v. every 6 h from d 22 to 28 of lactation, and AA and AR sows received saline. Sow weight, backfat, and litter weight were recorded weekly. Within 2 d after farrowing, litter size was standardized to 8 to 10. At d 17 of lactation, an indwelling jugular catheter was surgically implanted in each sow. Blood samples were taken for characterization of plasma LH, FSH, insulin, IGF-I, and leptin by RIA at d 21 and before and after weaning on d 28 of lactation. After weaning, all sows were given ad libitum access to feed, checked for onset of standing estrus twice daily with mature vasectomized boars, and inseminated 12 and 24 h after onset of standing estrus with pooled semen from the same fertile boars (3 x 10(9) sperm/AI). After breeding, feed allowance was reduced to NRC (1988) requirements for gestation. At d 28 +/- 3 of gestation, sows were killed and ovulation rate and embryo survival were determined. Restricted sows lost more weight during lactation than AA sows (P < .02). During the period of feed restriction, plasma IGF-I and postprandial insulin and leptin in AR and AR + GnRH sows, and LH pulse frequency in AR sows, were lower than those in AA sows (P < .04). Associations (P < .004) between plasma insulin and leptin and between leptin and mean LH concentrations were established. The LH pulse frequency in AR + GnRH sows did not differ from that in AA sows before weaning. After weaning, maximum, mean, and minimum LH concentrations in the AA and AR sows, and FSH concentrations in AR sows, increased (P < .05) in response to weaning. Paradoxically, GnRH treatment in lactation seemed to suppress the expected LH and FSH responses to weaning. Ovulation rate and embryo survival were not different among the three groups. In conclusion, although exogenous GnRH therapy restored LH secretion in feed-restricted sows, it did not improve overall reproductive performance.  相似文献

Root respiration may account for as much as 60% of total soil respiration. Therefore, factors that regulate the metabolic activity of roots and associated microbes are an important component of terrestrial carbon budgets. Root systems are often sampled by diameter and depth classes to enable researchers to process samples in a systematic and timely fashion. We recently discovered that small, lateral roots at the distal end of the root system have much greater tissue N concentrations than larger roots, and this led to the hypothesis that the smallest roots have significantly higher rates of respiration than larger roots. This study was designed to determine if root respiration is related to root diameter or the location of roots in the soil profile. We examined relationships among root respiration rates and N concentration in four diameter classes from three soil depths in two sugar maple (Acer saccharum Marsh.) forests in Michigan. Root respiration declined as root diameter increased and was lower at deeper soil depths than at the soil surface. Surface roots (0-10 cm depth) respired at rates up to 40% greater than deeper roots, and respiration rates for roots < 0.5 mm in diameter were 2.4 to 3.4 times higher than those for roots in larger diameter classes. Root N concentration explained 70% of the observed variation in respiration across sites and size and depth classes. Differences in respiration among root diameter classes and soil depths appeared to be consistent with hypothesized effects of variation in root function on metabolic activity. Among roots, very fine roots in zones of high nutrient availability had the highest respiration rates. Large roots and roots from depths of low nutrient availability had low respiration rates consistent with structural and transport functions rather than with active nutrient uptake and assimilation. These results suggest that broadly defined root classes, e.g., fine roots are equivalent to all roots < 2.0 mm in diameter, do not accurately reflect the functional categories typically associated with fine roots. Tissue N concentration or N content (mass x concentration N) may be a better indicator of root function than root diameter.  相似文献

The impact of feeding, fish size (body weight from 18.5 to 56.5 g) and water temperature (20 and 23 °C) on oxygen consumption (OC, mg O2 kg–1 h–1) and ammonia excretion (AE, mg TAN kg–1 h–1) was studied in Eurasian perch held in recirculation systems. OC for both fed and feed-deprived (3 days) fish was higher at 23 °C (278.5 and 150.1 mg O2 kg–1 h–1) than at 20 °C (249.3 and 135.0 mg O2 kg–1 h–1; P < 0.01). AEs for both fed and feed-deprived fish were also significantly higher at 23 °C than at 20 °C (P < 0.001). Water temperature and fish size had a significant impact on the oxygen:feed ratio (OFR, kg O2 kg–1 feed fed day–1) and ammonia:feed ratio (AFR, kg TAN kg–1 feed fed day–1; P < 0.001). Their average values at temperatures of 20 and 23 °C were 0.17 and 0.19 kg O2 kg–1 feed fed day–1 and 0.009 and 0.011 kg TAN kg–1 feed fed day–1, respectively.  相似文献

An important problem of treating patients with endotoxemia is to find drugs to reduce the negative effects of endotoxin on the organism. We tested fucoidan (sulfated polysaccharide) from the brown alga Fucus evanescens as a potential drug in a mouse model of endotoxemia inducted by lipopolysaccharide (LPS). The survival time of mice injected with LPS increased under fucoidan treatment compared with the group of mice injected with LPS only. The preventive administration of fucoidan to mice with endotoxemia resulted in inhibition of increased levels of proinflammatory cytokines (TNFα and IL-6), as well as decreasing of the processes of hypercoagulability. The parenteral or per os administration of fucoidan resulted in decreasing the degree of microcirculatory disorders and secondary dystrophic-destructive changes in parenchymal organs of mice with endotoxemia. Taken together, these results demonstrate that fucoidan prevents endotoxin-induced damage in a mouse model of endotoxemia and increases the mice’s resistance to LPS.  相似文献

A series of eight green, eight oolong, and 17 black teas have been analyzed for polyphenol content by absorbance at 272 nm and cyclic voltammetry response at an inert carbon electrode, a new method developed to provide a rapid measure of easily oxidizable polyphenols in beverages. The chain-breaking antioxidant activity of the teas has also been determined during the chain oxidation of methyl linoleate in a pH 7.4 micellar solution, for which realistic kinetic parameters have been derived. While higher mean values were obtained for green teas than for oolong and black teas, the differences were not large, and the spread of values within each type was considerable. The absorbance at 272 nm correlated well with the cyclic voltammetry response only for green teas and black teas taken on their own. The cyclic voltammetry measure and the antioxidant activity correlated well only for the green teas, where the polyphenol content is dominated by epigallocatechin gallate.  相似文献

Deposition of anthropogenic nitrogen (N) alters the decomposition of organic matter in forest ecosystems by changing the expression of key microbial enzymes. We investigated the effects of experimental N deposition on dissolved organic matter (DOM) in soils of three forest ecosystems representative of the upper Great Lakes region: the sugar maple/basswood (SMBW), sugar maple/red oak (SMRO) and white oak/black oak (WOBO) ecosystems. Mineral soil samples were collected on five dates from ambient and N-amended plots (80 kg N ha−1 yr−1) in three replicate stands of each forest type. DOM was extracted (2:1, water:soil) from each soil sample and analyzed for dissolved organic carbon (DOC). DOC concentration was significantly greater in the N-amended soils (on average: 24% higher for SMBW, 9% for SMRO, and 40% for BOWO). In June and October 2002, bioassays were performed to assess N treatment effects on the composition of DOM and its interacting bacterial community. Within each site, DOM extracts from the ambient and N-amended plots were reciprocally inoculated with bacteria from each plot. After a 48 h incubation at 20 °C, community activity in each microcosm was profiled by measuring 10 extracellular enzyme activities (EEA). MANOVA showed that ecosystem type, sampling date, DOM source (ambient or N-amended plot) and inoculum source (ambient or N-amended plot) all had significant effects on bioassay EEA. Post hoc tests (Tukey's HSD) found significant reductions in oxidative enzyme activity as a result of the N treatment. In general, the bioassay results corroborated a previous report describing losses in soil oxidative enzyme activity in response to N saturation. However, it is not clear whether increased DOC concentration is the direct result of reduced oxidative activity.  相似文献

Critical loads of acid deposition for forest soils, ground and surface water resources are calculated utilising a variety of mathematical models. The estimation of the predictive uncertainty inherent in these models is important since the model predictions constitute the cornerstone of the development of emissions abatement policy decisions in Europe and the United Kingdom. The Generalised Likelihood Uncertainty Estimation (GLUE) approach is presented here as a tool for estimating the predictive uncertainty of PROFILE, a steady-state geochemical model that is widely applied within the critical loads community. GLUE is based on Monte Carlo simulation and explicitly recognises the possible equifinality of parameter sets. With this methodology it is possible to make an assessment of the likelihood of a parameter set being an acceptable simulator of a system when model predictions are compared to observed field data. The methodology is applied to a small catchment at Plynlimon, Mid-Wales. The results highlight that there is a large amount of predictive uncertainty associated with the model at the site: three of the six chosen field characteristics lie within the predicted distribution. The study also demonstrates that a wide range of parameter sets exist that give acceptable simulations of the site characteristics as well as a broad distribution of critical load values that are consistent with the site data. Additionally, a sensitivity analysis of model parameters is presented.  相似文献

Human activity has increased the amount of N entering terrestrial ecosystems from atmospheric NO3 deposition. High levels of inorganic N are known to suppress the expression of phenol oxidase, an important lignin-degrading enzyme produced by white-rot fungi. We hypothesized that chronic NO3 additions would decrease the flow of C through the heterotrophic soil food web by inhibiting phenol oxidase and the depolymerization of lignocellulose. This would likely reduce the availability of C from lignocellulose for metabolism by the microbial community. We tested this hypothesis in a mature northern hardwood forest in northern Michigan, which has received experimental atmospheric N deposition (30 kg NO3-N ha−1 y−1) for nine years. In a laboratory study, we amended soils with 13C-labeled vanillin, a monophenolic product of lignin depolymerization, and 13C-labeled cellobiose, a disaccharide product of cellulose degradation. We then traced the flow of 13C through the microbial community and into soil organic carbon (SOC), dissolved organic carbon (DOC), and microbial respiration. We simultaneously measured the activity of enzymes responsible for lignin (phenol oxidase and peroxidase) and cellobiose (β-glucosidase) degradation. Nitrogen deposition reduced phenol oxidase activity by 83% and peroxidase activity by 74% when compared to control soils. In addition, soil C increased by 76%, whereas microbial biomass decreased by 68% in NO3 amended soils. 13C cellobiose in bacterial or fungal PLFAs was unaffected by NO3 deposition; however, the incorporation of 13C vanillin in fungal PLFAs extracted from NO3 amended soil was 82% higher than in the control treatment. The recovery of 13C vanillin and 13C cellobiose in SOC, DOC, microbial biomass, and respiration was not different between control and NO3 amended treatments. Chronic NO3 deposition has stemmed the flow of C through the heterotrophic soil food web by inhibiting the activity of ligninolytic enzymes, but it increased the assimilation of vanillin into fungal PLFAs.  相似文献
