The antifungal properties of 14 major oleoresin‐constitutive terpenoids of Norway spruce (Picea abies) against Heterobasidion parviporum were evaluated in vitro at three gradient concentrations, 0.1, 0.2 and 0.4 μmol/cm2, on potato dextrose agar medium. Eight monoterpene hydrocarbons (+)‐ and (?)‐α‐pinene, (?)‐β‐pinene, (+)‐3‐carene, myrcene, (+)‐ and (?)‐limonene, terpinolene; four oxygenated monoterpenes α‐terpineol, terpinen‐4‐ol, 1,8‐cineole, bornyl acetate; and two resin acids abietic acid and dehydroabietic acid were selected. Abietic and dehydroabietic acids showed the highest antifungal activities at all concentrations. Among oxygenated monoterpenes, bornyl acetate and α‐terpineol showed antifungal activity at the highest concentration. Among monoterpene hydrocarbons, (+)‐α‐pinene showed similar activity to terpinen‐4‐ol and 1,8‐cineole at the highest concentration; however, it was lower than α‐terpineol and bornyl acetate. Other monoterpene hydrocarbons inhibited mycelial growth by <10%. Re‐extraction of medium surfaces after the test period revealed that most α‐terpineol and terpinen‐4‐ol remained unchanged on the surface but monoterpene hydrocarbons completely disappeared. However, notable fungal transformed products were observed on surfaces applied with 1,8‐cineole and bornyl acetate. Thus, mycelial growth inhibition of monoterpenoids might be caused by complex functions such as biodegradation and/or detoxification by interaction between mycelium and compound. These results provide a basis for future studies considering the role of chemodiversity in the comprehensive chemical defence of P. abies.  相似文献

Interannual variability of the ecosystem of the Kii Channel, productive shelf water on the Pacific side of south‐western Japan, was analysed based on physicochemical environmental variables and abundance of major zooplankton taxa collected monthly for 12 yr from 1987 to 1999. The Kii Channel experienced both short‐term (i.e. 3–4 yr) cyclical changes and a long‐term (i.e. decadal) environmental trend. The short‐term variability was primarily associated with year‐to‐year differences in intrusion of subsurface, cold nutrient‐rich water along the bottom of the Kii Channel from the Pacific Ocean. When this bottom intrusion was intense, the Kii Channel experienced a cold, new production‐dominated ecosystem. The bottom intrusion, however, has become less intense in recent years, because of the closer proximity of the Kuroshio flow axis to the Kii Channel. Hence, there was a consistent trend towards warming and a regenerated production‐dominated ecosystem. In accordance with such environmental changes, the zooplankton community shifted towards more oceanic conditions; major herbivorous calanoids (i.e. Calanus sinicus, Paracalanus parvus (s.l.), Clausocalanus spp. and Acartia omorii) decreased, while carnivores (i.e. hydromedusae and Sagitta spp.) increased. The amount of total fish catch also decreased, while the catch of subtropical species increased. These findings lead us to conclude that the bottom intrusion from the Pacific Ocean plays a key role in determining the biological production in the Kii Channel.  相似文献

Parasporal inclusion proteins produced by Bacillus thuringiensis strain A1470 exhibit strong cytotoxicity against human leukemic T cells when activated by protease treatment. One of the cytotoxic proteins was separated by anion exchange chromatography and gel filtration chromatography and designated Cry45Aa. Its gene was then expressed in recombinant Escherichia coli, in which the Cry45Aa precursor was accumulated in an inclusion body. It was solubilized in sodium carbonate buffer and processed with proteinase K, and cytotoxic activities of the protein against various mammalian cell lines were evaluated using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H tetrazolium bromide assay. The protein exhibited high cytotoxic activity against CACO-2, Sawano, MOLT-4, TCS, and HL60 cells and moderate activity against U-937 DE-4, PC12, and HepG2 cells. On the other hand, the EC50 values against Jurkat, K562, HeLa, A549, Vero, COS-7, NIH3T3, CHO, and four normal tissue cells (human primary hepatocyte cells, UtSMC, MRC-5, and normal T cells) were >2 microg/mL.  相似文献

In our ongoing efforts to develop new uses for wood-based waste streams, the growth inhibition activities of extracts obtained from Sugi (Cryptomeria japonica) bark were examined against Heterosigma akashiwo, otherwise known as red tide plankton. The Sugi bark was separated into its outer and inner barks and then extracted sequentially with hexane, ethyl acetate, and methanol. Strong inhibitory activities against H. akashiwo were observed in the tests involving the hexane extract from the inner and outer barks, as well as the ethyl acetate extract from the inner bark. Gas chromatography mass spectroscopy (GC–MS) analysis revealed that cubebol, phyllocladanol, 6,7-dehydroferruginol, ferruginol, and sugiol were the main components in the active extracts. These components themselves were then tested for their growth inhibition activities against H. akashiwo. Cubebol and ferruginol showed potent inhibitory activities, whereas phyllocladanol, 6,7-dehydroferruginol, and sugiol were only weakly active. Taken together, these results suggested that the Sugi bark extracts could be used as inhibition reagents against red tide plankton.  相似文献

Annual amounts of litterfall and nitrogen input by litterfall were measured in a subtropical evergreen broad-leaved forest to examine the contribution of a liana species, Mucuna macrocarpa Wall., to the spatial heterogeneity of litterfall production and nitrogen input. The total litterfall in the study plot was 7.1 t ha−1 year−1. The amount of litterfall varied with topography and was greatest at the valley bottom and decreased toward the ridges. Macuna macrocarpa litterfall was absent on the ridges although it accounted for the largest percentage, 32%, of total leaf litter production in the valley. Nitrogen input by litterfall was 69 kg ha−1 year−1 in the plot. Nitrogen input by litterfall was also largest at the valley bottom and decreased toward the ridges. Leaf litter of M. macrocarpa had approximately twice the nitrogen concentration of litterfall of other species. Macuna macrocarpa accounted for 42% of nitrogen input by leaf litter in the valley. The abundance and the high nitrogen concentration of M. macrocarpa intensified differences in the amount of litterfall and nitrogen input by litterfall between valleys and ridges. It was concluded that a liana species, M. macrocarpa, can contribute to the spatial heterogeneity of litterfall and may subsequently affect nutrient cycling in a subtropical evergreen broad-leaved forest on Okinawa Island.  相似文献

Germination stimulants for root holoparasitic weeds broomrapes ( Orobanche and Phelipanche spp.) produced by tobacco ( Nicotiana tabacum L.) were purified and characterized. The root exudates of tobacco contained at least five different stimulants, and LC-MS/MS analyses revealed that four of them were strigolactones; a tetradehydrostrigol isomer, a didehydrostrigol isomer, and two strigol isomers. The two isomers of strigol were identified as (+)-orobanchol and its 2'-epimer by comparison of NMR and GC- and LC-MS data with those of synthetic standards. The structure of the tetradehydrostrigol isomer, the major stimulant of the bright yellow tobacco cultivars, was determined as 4-alpha-hydroxy-5,8-dimethyl-GR24 [( E)-4-alpha-hydroxy-5,8-dimethyl-3-(4-methyl-5-oxo-2,5-dihydrofuran-2-yloxy)methylene)-3a,4-dihydro-3 H-indeno[1,2- b]furan-2(8b H)-one] and named solanacol. 2'-Epi-orobanchol and solanacol are the first natural strigolactones having a 2'-epi stereochemistry and a benzene ring, respectively.  相似文献

To examine the effects of salicylic acid (SA), the ethylene precursor 1‐aminocyclopropan‐1‐carboxylic acid (ACC) and methyl jasmonate (MJ), which induce defence responses against pathogen in plants, on foliar endophytic fungi, we performed exogenous treatments of Quercus serrata leaves with these chemicals and subsequently isolated endophytic fungi. SA and ACC significantly reduced the isolation frequency of the most dominant fungus, a Xylaria species as identified by sequence analysis of the internal transcribed spacer (ITS) region of the ribosomal DNA. In contrast, the frequency of the second most frequent fungus increased by SA and ACC. A few other fungi were also affected positively or negatively. Shannon diversity indices decreased by all treatments because of the uneven isolation frequencies of the endophytes. Our results imply that host defence responses induced by the chemical treatments reduced the number of infections of some endophytes while the frequency of some other endophytes increased benefiting from the reduction of competitors.  相似文献

In order to find new utilization method of woody wastes, we examined the bioactivities of extracts from branch heartwood of Chamaecyparis obtusa (hinoki) and compared to extracts from trunk heartwood. The bioactivities examined were antifungal activities against four fungi (Trametes versicolor, Fomitopsis palustris, Trichoderma virens, Rhizopus oryzae), and bioassay with brine shrimp (Artemia salina) which shows any allelopathic activities not measurable with fungi. Antifungal activities were observed in the hexane and ethyl acetate extracts of branch and trunk heartwood. The hexane and ethyl acetate extracts of branch and hexane extract of trunk showed strong lethality against brine shrimp. The yields of the active extracts of branch were much more than that of trunk. The identified compounds in the active extracts of branch were germacra-1-(10),5-dien-4??-ol, t-cadinol, t-muurolol, hinokiresinol, and hinokinin. Hinokiresinol and t-muurolol showed strong antifungal activities. Hinokiresinol showed bioactivities against T. virens, R. oryzae and brine shrimp. Germacra-1-(10),5-dien-4??-ol was lethal to brine shrimp. Germacra-1-(10),5-dien-4??-ol and hinokiresinol were minor components in trunk heartwood, but major components in branch. These qualitative and quantitative results suggest that the branch heartwood could be a valuable chemical resource because it contains large amounts of antifungal and allelopathic compounds.  相似文献

Interleukin-18 (IL-18) is a proinflammatory cytokine expressed in female reproductive organs in humans, rats and mice. The physiological roles of uterine IL-18 and the regulatory mechanisms of IL-18 gene expression are unclear. The present study aimed to clarify the effects of estradiol-17beta (E2) and progesterone (P4) on IL-18 mRNA expression in the mouse uterus. Distribution and expression levels of IL-18 mRNA were studied using an RNase protection assay. Expression of IL-18 mRNA was observed in all organs studied, including testes, ovaries and uteri. The uterine IL-18 mRNA level of estrous mice was higher than that of diestrous mice. E2 treatment (1, 5, 25 or 250 ng/mouse) decreased uterine IL-18 mRNA levels in ovariectomized mice dose-dependently. E2 treatment acutely decreased IL-18 mRNA levels 3 h after injection, but these levels returned to the initial level after 48 h. P4 treatment (1 mg/mouse) decreased uterine IL-18 mRNA levels after 12 h, but levels returned to the initial level after 48 h. Both uterine IL-18 and IL-18Ralpha mRNAs were detected in cultured endometrial epithelial and stromal cells. These results suggest that uterine IL-18 expression is reduced by sex steroid hormones and that IL-18 acts on endometrial cells in a paracrine or autocrine manner.  相似文献

Japanese oak wilt (JOW) has been prevalent in Japan since the late 1980s. Infections of the fungus, Raffaelea quercivora Kubono et Shin. Ito, which is transmitted by an ambrosia beetle [Platypus quercivorus (Murayama)], can cause JOW. Although R. quercivora, P. quercivorus, and oak trees are distributed in other Asia–Pacific countries, the incidence of JOW has not been reported outside Japan. In this study, we collected R. quercivora isolates from 5 Asian countries, including Japan, and compared their ability to induce sapwood discoloration by inoculating Q. serrata logs. The tangential widths of the discoloration in sapwoods inoculated with non-Japanese isolates were equivalent to or greater than those of the 2 Japanese isolates. This indicates that a lack of JOW incidence outside Japan is not because of the lowered ability of R. quercivora to spread discoloration compared with the Japanese isolates. Statistical analyses of the relationship between discoloration and phylogeny based on DNA sequences of actin and chitin synthase showed that the discoloration width was independent of phylogenetic relatedness among the isolates. To discuss why the occurrence of JOW has not been reported outside Japan, further studies (e.g., on host susceptibility and P. quercivorus aggression) throughout Asia are needed.  相似文献
