Development of new semen cryopreservation techniques improving sperm survival and ensuring availability of viable spermatozoa for a prolonged time‐period after AI is promising tools to reduce sensitivity of timing of AI and enhance overall fertility. The SpermVital® technology utilizes immobilization of bull spermatozoa in a solid network of alginate gel prior to freezing, which will provide a gradual release of spermatozoa after AI. The objective of this study was to compare post‐thaw sperm quality and in vitro sperm survival over time of Norwegian Red bull semen processed by the SpermVital® (SV) technology, the first commercialized production line of SpermVital® (C) and by conventional procedure applying Biladyl® extender (B). Post‐thaw sperm motility was not significantly different between SV, C and B semen (p > .05). However, sperm viability and acrosome intactness were higher for SV than C and B semen (p < .05). Small differences in DNA quality were observed (p < .05). Sperm viability after storage in uterus ex vivo was higher for SV than for C semen (p < .05). Furthermore, sperm survival in vitro over time at physiological temperature was significantly higher for SV semen than C semen as well as B semen during the incubation period of 48 hr (p < .05). In conclusion, the SpermVital® technology is improved and is more efficient in conserving post‐thaw sperm quality and results in higher sperm viability over time in vitro for SV than for C and B semen.  相似文献

Stimulation of the non-specific defence enhances the disease resistance and growth, and has good potentials as a measure for increased microbial control in juvenile production of marine fish and shellfish. So far, the most commonly used immunostimulants are β-(1 → 3, 1 → 6)-glucans, and in this study the stimulatory potential of a β-(1 → 3, 1 → 6)-glucan of marine origin, the storage polysaccharide from the marine diatom Chaetoceros mülleri, was examined. The glucan (chrysolaminaran) was extracted from cultures of C. mülleri, and used as a dietary supplement in two first feeding experiments with larvae of Atlantic cod Gadus morhua L. In one experiment the microalgal glucan was compared to the commercial yeast-glucan product MacroGard®, and in the other to an alginate with a high content of mannuronic acid (High-M alginate) isolated from Durvillaea antarctica. The stimulants were given via rotifers, and weaning to formulated feed was initiated at day 17 or 18 after hatching. The survival ± SEM at day 27 after hatching was 24.5 ± 2.0%, 14.8 ± 4.5% and 13.1 ± 1.4% for the groups fed C. mülleri-glucan, yeast glucan and for the control, respectively, in the first experiment. The group fed C. mülleri-glucan group had higher survival compared to the control (< 0.05) group, whereas the yeast glucan had no positive effect on the survival (p > 0.05). The dry weights of the groups at day 27 were low, with 203.2 ± 52.2, 165.2 ± 43.4 and 198.5 ± 58.1 μg per larva for the C. mülleri-glucan, yeast glucan and control groups, respectively. In the second experiment the survival in the period of feeding formulated feed (days 18-30) were 44.6 ± 4.3%, 44.7 ± 1.3%, and 33.8 ± 4.1% survival for the C. mülleri-glucan, High-M alginate and control group, respectively. The cod larvae fed C. mülleri-glucan reached an average weight of 531.6 ± 17.2 μg at day 30, which was significantly higher (p < 0.05) than the control group that had an average of 473.6 ± 3.5 μg. The larvae fed High-M alginate had an average weight of 470.3 ± 31.6 μg per larva at day 30, and not significantly different from the control (p > 0.05). The early weaning to formulated diet had detrimental effect on the growth of the larvae. In both experiments the C. mülleri-glucan group was the only group showing a positive growth rate in the period of weaning to dry feed. The microbial conditions in larval gut and water were monitored with respect to total colony forming units on Marine agar, and Vibrio- and Pseudomonas-like species on selective agars (TCBS and marine Pseudomonas Agar with CFC-supplement). The larvae were rapidly colonised after hatching, but no or weak effects of the stimulants were observed on the colonisation rates or the composition. The total CFU varied from 101 to 102 CFU per μg larva after initiation of the first feeding. The percentages of Pseudomonas-like bacteria increased throughout the period, whereas the levels of Vibrio-like bacteria were low and stable. The chrysolaminaran from the diatom C. mülleri was shown to be a promising candidate for use as an immunostimulatory feed additive, and which should be further explored.  相似文献

Two transgenic Bt rice lines, KMD1 and KMD2, both containing a synthetic cry1Ab gene from Bt, were crossed with conventional rice varieties. The inheritance of resistance to SSB of KMD1 and KMD2was investigated through LSB and field examination of their progenies, e.g. F1, BC1 and F2 populations. In LSBs, 100.0% of newly hatched SSB larvae died on the second day after feeding on leaf tissues of F1 and GUS positive BC1 plants, of which the area of leaf tissues consumed by SSB is also similar to that of transgenic parents. These results imply that the resistance of Bt rice to SSB is dominantly controlled and could be easily exploited in hybrid rice production. Field evaluation showed that segregation ratios for SSB resistance to susceptibility in BC1 populations fit the ratio of 1:1, which was also confirmed by LSBs. However, in F2 populations, the ratio was significantly smaller than 3:1 for resistant to susceptible plants in all 6 indica × japonica (KMD1 and KMD2) crosses, though it fitted 3:1 in all 4 japonica × japonica crosses. The results implied that the resistance of Bt rice to SSB was controlled by a dominant gene which was present in a homozygous condition in both KMD1 and KMD2, but the inheritance could be affected by other factors. Assays for Cry1Ab protein showed that, in most crosses, the content of Cry1Ab is significantly higher in leaves of GUS positive F1, BC1 and F2 plants than that in transgenic Bt parent plants, which accounts for the high resistance observed in these plants to SSB. Bolting and flowering of Aster novi-belgii seedlings as well as correlations between characteristics within seedling populations and between seedlings and their vegetatively propagated offspring were investigated in order to improve breeding efficiency. In seedlings of A. novi-belgii, flower induction was observed without vernalization. Seedlings initially formed a rosette, but bolting was a prerequisite for development of a normal flowering shoot. Bolting was promoted by long photoperiods as compared to short days. The shortest time from sowing to anthesis was recorded when plants were grown under long photoperiods until the main stem was at least 5 cm long, and then moved to short photoperiods. A large variation in time to anthesis, plant height, branching ability, and flower size and color was found among the seedlings within all crosses. Magnitude and significance of correlations between some characteristics varied among crosses. The more consistent correlations seem mainly to be due to natural courses of plant growth and development. High correlations between seedling characteristics and their vegetatively propagated offspring were only found for flower hue. Lower, but significant correlations were found for flower color intensity, plant height and branching ability. There were no significant correlations for the time-to-anthesis between the two groups of plants. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The oxidation in aqueous dispersions of egg yolk powder and the influence of addition of the proposed antioxidants ascorbic acid and ascorbic acid 6-palmitate indicate that both ascorbic acid and ascorbic acid 6-palmitate propagated the oxidation of egg yolk powder dispersions. Ascorbic acid 6-palmitate was found to be more prooxidative than ascorbic acid. Moreover, it was found that addition of ascorbic acid or ascorbic acid 6-palmitate gave rise to an increase in the amount of free iron Fe(II) in the egg yolk dispersions. It is proposed that ascorbic acid and ascorbic acid 6-palmitate react with the phosvitin-Fe(III) complex found in egg yolk and release Fe(II), which subsequently propagates lipid oxidation. It appears that less oxidation occurs in egg yolk dispersions exposed to high concentrations of peroxy radicals with added ascorbic acid than egg yolk dispersions with added ascorbic acid without exposure to peroxy radicals.  相似文献

Studies of earthworm species and their activity, expressed as channels, on cultivated loamy (Humic Cryaquept) and clayey soils (Typic Endoaqualf and Typic Cryaquept) were conducted in southern and central Norway before conversion from conventional to organic cropping systems. At all the three study areas: Landvik (Grimstad), Voll (Ås) and Kvithamar (Stjørdal), the earthworm species Aporrectodea caliginosa, Aporrectodea rosea, Allolobophora chlorotica and Lumbricus rubellus were found. At Landvik, where the deep-burrowing species Aporrectodea longa, A. caliginosa and Lumbricus terrestris dominated, medium (4–6 mm) and coarse (>6 mm) earthworm channels were most numerous below the plough layer. Almost no coarse pores were found at Voll and Kvithamar. The volume of earthworm channels below the plough layer was 0.6–0.8% of total soil volume at Landvik and Voll and 0.3–0.4% at Kvithamar. Earthworm channels >6 mm below the plough layer were useful to identify present and previous activity of deep-burrowing earthworms such as L. terrestris. Below the plough layer, roots were almost entirely restricted to earthworm channels and interaggregate spaces.  相似文献

Analyses of the spatial and temporal variations in the natural abundance of 13C are frequently employed to study transformations of plant residues and soil organic matter turnover on sites where long continued vegetation with the C3-type photosynthesis pathway has been replaced with a C4-type vegetation (or vice versa). One controversial issue associated with such analyses is the significance of isotopic fractionation during the microbial turnovers of C in complex substrates. To evaluate this issue, C3-soil and quartz sand were amended with maize residues and with faeces from sheep feed exclusively on maize silage. The samples were incubated at 15 °C for 117 days (maize residues) or 224 days (sheep faeces). CO2 evolved during incubation was trapped in NaOH and analysed for C isotopic contents. At the end of incubation, 63 and 50% of the maize C was evolved as CO2 in the soil and sand, respectively, while 32% of the faeces C incubated with soil and with sand was recovered as CO2. Maize and faeces showed a similar decomposition pattern but maize decomposed twice as fast as faeces. The δ13C of faeces was 0.3‰ lower than that of the maize residue (δ13C −13.4‰), while the δ13C of the C3-soil used for incubation was −31.6‰. The δ13C value of the CO2 recovered from unamended C3-soil was similar or slightly lower (up to −1.5‰) than that of the C3-soil itself except for an initial flush of 13C enriched CO2. The δ13C values of the CO2 from sand-based incubations typically ranged −15‰ to −17‰, i.e. around −3‰ lower than the δ13C measured for maize and faeces. Our study clearly demonstrates that the decomposition of complex substrates is associated with isotopic fractionation, causing evolved CO2 to be depleted in 13C relative to substrates. Consequently the microbial products retained in the soil must be enriched in 13C.  相似文献
