In this study, the interrelation between nitrogen availability and prokaryotic diversity are studied using a well-characterised system from a long-term field experiment on a loamy sandy soil. The prokaryotic potential functional diversity and community composition were assessed using community-level physiological profiling (CLPP), and their phylogenetic diversity was analysed using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) in combination with sequencing analysis. Highest prokaryotic potential functional diversity was measured in the control soil receiving no N fertilisation, indicating an efficient as well as versatile utilisation of the substrates in this soil. Both substrate utilisation richness and substrate utilisation evenness, the two constituents of the functional diversity, were decreased with increasing N supply. Furthermore, distinct prokaryotic community compositions were generated in N-enriched soils compared to unfertilised control soils. These differences suggest a dominance of populations adapted to utilising readily available substrates. We demonstrated that the shift in prokaryotic functional diversity was connected to a shift in the phylogenetic structure of the bacterial and archaeal communities. Taken together, our data clearly show that, for the sandy soil system, prokaryotic diversity and N availability were interrelated.  相似文献

In contrast with macroorganisms, whose geographical ranges are typically restricted, many microbial species appear to have cosmopolitan distributions. This observation has been explained as a consequence of ubiquitous dispersal caused by the enormous population sizes of microbial species. Recently, this "everything is everywhere, but, the environment selects" theory has been challenged by the detection of considerable regional genetic variability within microbial morphospecies. We demonstrate that, contrary to what is expected under ubiquitous dispersal, evidence of regional-scale metacommunity processes can be detected in microbial morphospecies. Our results imply that the microbial and macrobial world are structured by analogous processes.  相似文献

Variability in the availability of suitable prey (mainly nauplii stages of Calanus spp.) in the early life stages of cod is likely a significant contributor to the fluctuations in the year‐class strength in the Arcto Norwegian cod stock. We have investigated the origin of the Lofoten shelf population of Calanus using a particle tracking model approach based on velocity fields simulated from 2002 to 2012. By performing backwards simulations of trajectories from particles released on the Lofoten shelf in early spring, we assessed the relative contribution of potential Calanus sources, such as the Lofoten Basin, the local overwintering on the continental shelf and the adjacent fjords. This analysis revealed significant interannual differences in advection patterns mainly driven by changes in wind conditions and variable strength of the Norwegian Coastal Current. Most of the particles advected into our study area originated on the shelf, and contrary to expectations, only a few originated from the Lofoten Basin where Calanus abundances tend to be high. These results suggest that contribution from the Lofoten Basin to the early spring shelf Calanus population may be overestimated and that contribution from more local sources, especially from adjacent fjords, may play a more important role than what has been previously anticipated. Our results highlight the necessity to improve quantification of shelf‐fjord exchange processes, as these may contribute more to the regulation of spring zooplankton stocks on the northern Norwegian shelf, and, as a result, may affect the survival of cod larvae in this area.  相似文献

Two methods for the determination of DNA have been adapted for use on the bacterial fraction of an organic soil, obtained by fractionated centrifugation. The. soil contained about 1.1 × 1010 bacteria g?1 dry weight when counted by fluorescence microscopy. One method was based on the reaction with 3,5-diaminobenzoic acid 2HCl, and the other on the specific reaction of the antibiotic mithramycin with double-stranded DNA. For the first method, about 8 × 108 cells were required and for the second, about 4 × 109. The analytical results with the two methods agreed well. As an average of several determinations, an amount of 8.4 fg (10?15g) DNA per bacterial cell was found when the cells were counted by fluorescence microscopy. This is in the upper size range of bacterial genomes. showing that virtually all microscopically counted bacteria in this soil contain DNA. The total amount of DNA was about 90μg g?1 soil dry weight.  相似文献

The coastal heathlands of north-western Europe are endangered habitats of great conservation value. Invasion by bracken Pteridium aquilinum is a major challenge for conservation and restoration of these heathlands, including the under-studied northern regions. Today, the herbicide asulam is the most widely applied bracken control measure, but increasing focus on organic farming and nature conservation calls for alternative, preferably mechanical, approaches. In a 7-year replicated field experiment in western Norway, we investigated efficiencies of the four bracken control measures asulam, Gratil, annual cutting and biannual cutting, in restoring the characteristic heathland vegetation structure and species composition. We specifically tested herbicide effects on diversity and composition of non-target species. Effects of treatments over time were evaluated by repeated measures ANOVA, and for multivariate data, Principal Response Curves. Our results show that UK based control methods are largely applicable to bracken at its northern limit in the European heathland habitat. Asulam resulted in the fastest reduction in cover but cutting proved equally efficient long-term. Community compositions progressed towards desired heathland vegetation, but successional trajectories differed. Asulam had unintended effects on a number of heathland species not predictable by species characteristics or functional groups. Gratil failed to have any long-term effects. In summary, cutting is as efficient as herbicide application in reducing bracken, and more so in restoring northern heathland vegetation over time.  相似文献

There are probably millions of species in the microorganismal domains Bacteria and Archaea (the prokaryotes), and we are only just beginning to work out the basic principles governing their distribution and abundance in natural environments. One characteristic that has become clear is that prokaryote diversity in aquatic environments is orders of magnitude less than in sediments and soils. Hypotheses and models explaining such differences are under development and are beginning to offer promising insights into the mechanisms governing prokaryote diversity and ecosystem function.  相似文献

DNA has been isolated from the bacterial fraction of two soils. Numbers of bacteria, determined by fluorescence microscopy were 1.1 and 2.2·1010 cells g?1 dry wt. The total amounts of bacterial DNA in these soils were 90 and 187 μg g?1 dry wt respectively.A modification of Marmur's method was used to isolate DNA, but difficulties in separating DNA from humic substances gave low yields and impure DNA.DNA could be partly separated from humic material in the presence of 8 m urea by ion-exchange chromatography on DEAE-Sepharose CL-6B. Final purification was obtained by chromatography on a hydroxyapatite column. When lowering the EDTA concentration in the saline-EDTA solution used for lysis, the amount of humic substances in the cell-free lysate after centrifugation was considerably decreased. The lysate could then be chromatographed directly on hydroxyapatite. Quantities up to 1.5 mg DNA high purity was isolated from 90 g wet soil (37 g dry wt).The isolated DNA was characterized by treatment with DNAse and absorption spectra. No uncommon bases were revealed by thin layer chromatography of the DNA hydrolysates. Melting curves of the isolated DNA showed a relatively broad melting profile, with half maximum hyperchromicity (Tm) near 90°C. Sedimentation coefficients determined by analytical ultracentrifugation showed that the isolated DNA had a high molecular weight, ranging from 2.3 to 10.1·105 daltons.  相似文献

Pesticide effects on bacterial diversity in agricultural soils – a review   总被引:20,自引:0,他引:20  
According to guidelines for the approval of pesticides, side-effects on soil microorganisms should be determined by studying functional parameters such as carbon or nitrogen mineralisation. However, the microbial diversity may have been markedly changed following pesticide use despite unaltered metabolism, and such changes may affect soil fertility. This review evaluates new methods for measuring pesticide effects on bacterial diversity, and discusses how sampling should take temporal and spatial heterogeneity into account. Future research on pesticide approval protocols should establish the relationships between mineralisation assays and new and rapid bacterial diversity profiling methods, and should include the possible ecological implications of altered bacterial diversity for soil fertility.  相似文献

With three natural soils from Western and Central Norway it has been shown that by homogenization and fractionated centrifugations, two fractions could be obtained, one containing 50–80% of the bacteria, and the other all the fungi together with soil debris and the rest of the bacteria. When the separation was carried out in the cold and lasting less than 3–4 h, the fractions if used immediately had constant respiratory rates for 1–2 h. The sum of the rates agreed well with that of an unfractionated soil homogenate, making it possible to calculate the ratio between bacterial and fungal respiration. Assuming that the much higher respiration in a homogenate than in intact soil is due to inhibitory conditions in the latter and that these can be analysed by separate experiments, the technique will permit estimates of the ratio between bacterial and fungal respiration in in situ soil under given conditions.The soils used had very great differences between plate and microscopic count values for bacteria. When based on plate counts, the calculated bacterial respiratory intensities became impossibly high, while they were within biologically possible limits when microscopic count values were used. The respiratory intensities for fungi when calculated from hyphal lengths and diameter measurements, were biologically possible.  相似文献
