Membranes from house fly heads were tested for the presence of mucarinic acetylcholine receptors using as a probe [3H]quinuclidinyl benzilate ([3H]QNB). Based on the presence of saturable and reversible high-affinity binding of [3H]QNB, which is inhibited by muscarinic drugs, it is suggested that these sites may be muscarinic receptors. However, these putative muscarinic receptors differ in several characteristics from the ones in mammalian brain. They have lower affinities for muscarinic drugs and lower stereoselectivity, a relatively higher affinity for the nicotinic antagonist d-tubocurarine, a lower Hill coefficient for binding of muscarinic antagonists, and a lower concentration relative to α-bungarotoxin binding sites in the same membranes. Also, unlike mammalian muscarinic receptors, they are sensitive to treatments with N-ethylmaleimide and 5,5′-dithiobis(2-nitrobenzoic acid). The effect of reduction of disulfide bonds by dithiothreitol or mercaptoethanol suggests that only the insect receptor has one or more disulfide bonds which are important to binding. On the other hand, the putative muscarinic receptors of both insect and mammalian brains have important SH group(s), whose alkylation by p-chloromercuribenzoate inhibits binding. Also, chlorobenzilate is equally effective in inhibiting [3H]QNB binding to muscarinic putative receptors of house fly and bovine brains. 相似文献

In order to identify antigens that can help prevent camel tick infestations, three major glycoproteins (GLPs) about 97, 66 and 40 kDa in size were purified from adult and larval Egyptian ticks, Hyalomma (H.) dromedarii, using a single-step purification method with Con-A sepharose. The purified GLPs were evaluated as vaccines against camel tick infestation in rabbits. The rabbits received three intramuscular inoculations of GLPs (20 µg/animal) on days 0, 14, and 28. In the immunoblot analysis, Sera from the immunized rabbits recognized the native GLPs and other proteins from larval and adult H. dromedarii ticks along with those from other tick species such as Rhipicephalus sanguineus but not Ornithodoros moubata. The effects of immunity induced by these GLPs were determined by exposing rabbits to adult H. dromedarii ticks. These results demonstrated that GLP immunization led to a slightly decreased reproductive index and significantly reduced rates of egg hatchability. These results demonstrated that immunization with the purified GLPs can provide protection against infestation by H. dromedarii and some other tick species. Further studies are needed to confirm the effectiveness of immunization with GLPs against other tick species. 相似文献

[3H]Flunitrazepam ([3H]Flu) was used to identify benzodiazepine binding sites in house fly thorax muscle membranes using a filter assay. [3H]Flu bound to a finite number of sites in a concentration- and time-dependent manner, reaching equilibrium in 10 min. Scatchard plots of the binding indicated a high-affinity site at 0.2 pmol/mg protein (Kd 24.3 nM) and a low-affinity site at 8.2 pmol/mg protein (Kd994nM). Binding of [3H]Flu to the high-affinity binding site was inhibited by several benzodiazepine analogs, with Flu, diazepam, and Ro 5-4864 being more potent than β-CCE, Ro 5-3027, and Ro 5-2180. Clonazepam was least potent in inhibiting [3H]Flu binding. Thus, the drug specificity of these insect muscle benzodiazepine binding sites was quite different from both the mammalian central and peripheral benzodiazepine receptor sites, though closer to the peripheral ones. GABA (γ-aminobutyric acid) and its agonists enhanced the specific binding of [3H]Flu in a dose-dependent manner, and this effect was inhibited with the GABA antagonist bicuculline. The effect was biphasic since at high GABA concentrations this stimulation was reduced. The data suggest that house fly muscles have benzodiazepine receptors, which are coupled allosterically to GABA receptors, analogous to the GABA/benzodiazepine receptors of vertebrates, but with some differences in their drug specificities. 相似文献

A cross-sectional survey was conducted in Sudan to determine sero-prevalence and risk factors associated with Neospora caninum infection in non-vaccinated dairy herds and to assess importance of the disease. Blood samples were collected from a total
of 262 animals from 25 herds. Sera were tested for antibodies against N. caninum using ELISA test. The prevalence rates of N. caninum antibodies in cattle were high both at herd level (44%) and at individual animal level (10.7%). Herd level infection rates
were similar in Khartoum State (43.7%) and at Gazira States (44.4%). The overall prevalence rates were higher (16.1%) in Gazira
State than in Khartoum State (9%) but with no significant variation. The sero-prevalence at individual animal level was significantly
higher (p < 0.05) in animals with history of abortion (12.8%) than in apparently healthy animal (11.3%), animal with history of infertility
(8.1%), or neonatal death of calves (4.3%). In addition, significantly higher (P < 0.05) sero-prevalence was observed in samples collected during the rainy season (6.87%) than winter (3.05%) or summer (0.76%).
However, no significant differences in sero-prevalence due to locality, animal breed, sex, and age were observed (p > 0.05). This preliminary study reveals for the first time the existence of natural N. caninum infection in Sudan. Also, the findings of the present study indicated that this disease is highly prevalent in two major
areas of dairy production in the country, and this calls for control strategy to be implemented. 相似文献

The aim of the present study was to evaluate the validity of Theileria annulata surface protein (TaSP)-ELISA, in comparison with traditional microscopic test, for the diagnosis of T. annulata infection among Egyptian baladi cattle (Bos taurus) and water buffaloes (Bubalus bubalis). Molecular confirmation of infection using T. annulata merozoite surface (Tams-1) target amplification by PCR was used as a gold standard. A total of 76 clinically suspected animals including 64 baladi cattle and 12 water buffaloes were investigated in the current study by the three methods. Based on the PCR-confirmed results, the evaluation study revealed higher sensitivity of TaSP-ELISA (72.9% and 75%) as compared to microscopic examination (58.3% and 50%) among cattle and buffaloes, respectively. On the other hand, the specificity of TaSP-ELISA in diagnosis of T. annulata infection was higher (87.5%) in baladi cattle as compared to water buffaloes (37.5%). In conclusion, TaSP-ELISA was shown to be suitable for the diagnosis of T. annulata infection in cattle under field conditions. 相似文献
