Background
Reports worldwide indicate high prevalence of Chlamydophila spp. infection in cattle. To assess the prevalence in Sweden, 525 cows in 70 dairy herds with reproductive disorders was investigated.Methods
To detect antibodies two commercially available kits were used. Moreover, 107 specimens, including vaginal swabs, organ tissues and milk were analysed by Polymerase Chain Reaction (PCR).Results
Two (0.4%) cows were seropositive in the Pourquier Cp. abortus ELISA. The seroprevalence with the Chekit ELISA was 28% with no difference between cases and controls. Five specimens were positive in real-time PCR and further analysed by nested PCR. Cp. pecorum was confirmed by partial omp1 DNA sequencing of the nested PCR product of vaginal swabs from control cows.Conclusion
The results suggest that Cp. abortus infection is absent or rare in Swedish cows whereas Cp. pecorum is probably more spread. They also suggest that Chlamydophila spp. are not related to reproduction disorders in Swedish cattle. 相似文献2.

Background
Reproductive disorders associated with chlamydial infection have been reported worldwide in cattle and there are indications of potential venereal transmission.Methods
Semen samples from 21 dairy bulls and cauda epididymidis tissue samples from 43 beef bulls were analysed for chlamydial agent by real-time polymerase chain reaction (PCR) including an internal amplification control (mimic). Additionally, presence of antibodies against Chlamydophila (Cp.) abortus among the bulls was investigated with the commercial Pourquier® ELISA Cp. abortus serum verification kit.Results
No chlamydial agent was detected by PCR in either the semen samples or in the tissue samples. Additionally, no antibodies against Cp. abortus were detected.Conclusions
The results suggest that Cp. abortus is very rare, or absent in Swedish bulls and thus the risk for venereal transmission of chlamydial infection through their semen is low. However, because Chlamydophila spp. infection rates seem to differ throughout the world, it is essential to clarify the relative importance of transmission of the infection through semen on cattle fertility. 相似文献3.
Resistance to anticoagulant rodenticides in brown rats (Rattus norvegicus Berk.) is associated with pleiotropic effects, notably with an increased dietary vitamin K requirement. Owing to this disadvantage, resistance is believed to be selected against if anticoagulant selection is absent. In small experimental populations of wild brown rats, an investigation was carried out to establish whether tolerance to anticoagulant exposure changed over a period of 2 years. In the same populations, DNA microsatellite markers were used to infer parentage, and this made it possible to estimate reproductive success of sensitive and resistant rats and estimate effective population size, Ne. Even though there was evidence for a selection against resistant rats with high vitamin K requirement, anticoagulant tolerance was not seen to be significantly influenced in the absence of bromadiolone selection. As the population size under investigation was small, random genetic drift may have played a role in this. In the presence of bromadiolone selection, however, the tolerance was significantly increased, suggesting that continuous selection will increase the proportion of highly resistant rats in the population. It was found that, for both males and females, surprisingly few individuals contributed to the next generation with numerous offspring, and most breeders contributed with none or a single offspring. The expected higher reproductive success and consequent increase in proportional numbers of sensitive rats in the absence of anticoagulant selection could not be observed. Among the resistant rats, moderately resistant females were found to be better breeders than highly resistant breeders, but for resistant males the reverse was true. This could be explained by the fact that the increased vitamin K requirement results in sex differential selection; in highly resistant males the selection presumably takes place at the immature stage, whereas in females the vitamin K requirement becomes crucial at the reproductive stage, as vitamin K is not only essential for the blood clotting process but also for bone formation. 相似文献

Clubroot (Plasmodiophora brassicae) is a serious soil-borne disease in brassica crops world-wide. We report on a time series of soil samples from Swedish long-term fertility trials started in 1957, 1963 and 1966, which were analyzed for the amount of P. brassicae DNA. The crop rotations included a brassica crop every 4 or 6 years. All experimental sites with a 4-year rotation of oilseed rape, except one with calcium carbonate in the soil profile, showed high (>1000 fg DNA g?1 soil) levels of P. brassicae DNA after 9, 11 and 12 rotations. In contrast, detectable levels (>5 fg DNA g?1 soil) of P. brassicae were found only at one of five sites with a 6-year rotation of spring oilseed rape. In years with high levels of P. brassicae DNA, low yield was reported and a subsequent decline in P. brassicae DNA in soil was observed. Different NPK (nitrogen/phosphorus/potassium) fertiliser regimes resulted in similar P. brassicae DNA levels. The robustness and reliability of the method applied was verified by analyses of soil from individual plots compared with a mixture of plots and by repeated analyses of selected samples, which showed that P. brassicae DNA remained stable during dry storage. 相似文献

Resistance to warfarin has been connected to an increase in dietary requirement for vitamin K in British strains of the Norway rat, Rattus norvegicus (Berk). This study examines vitamin K requirement of Danish anticoagulant-resistant Norway rats using a vitamin K deficient feeding test. Wild bromadiolone-resistant rats sampled from different localities in Denmark and rats from bromadiolone-resistant and susceptible laboratory strains were fed on a vitamin K deficient diet over a maximum period of 15 days. Development of vitamin K deficiency, measured as reduced blood-clotting capacity, took place in 43% of the Danish resistant rats and was independent of sex, treatment with supplementary vitamin K3 and sampling locality. Development of deficiency was slower for resistant rats that were supplemented with vitamin K3 prior to the feeding test, suggesting storage of the vitamin K in a vitamin body pool. Intraperitoneal administration of vitamin K1 revealed that 80 microg vitamin K1 kg(-1) bodyweight was sufficient to restore normal blood clotting activity in deficient rats, while 60 microg vitamin K1 kg(-1) bodyweight was insufficient. We conclude that vitamin K requirement is moderately increased in Danish homozygous resistant rats whereas heterozygous resistant rats only have a minor increase in vitamin K requirement compared with susceptible rats. We found no indication of different resistance types being present in our test material since vitamin K requirement was not different between rats from separate sampling localities. 相似文献

The effect of a second mutant allele (V199I, here denoted rn*) at the PRKAG3 (RN) locus on carcass composition was determined in 334 pigs, entire males and females, from crosses between Swedish Hampshire (H) and Finnish Landrace (L) (H × LH; LH × H; LH × LH). Pigs were classified according to DNA test into the following PRKAG3 genotypes: RN−/RN− (23%), RN−/rn+ (24%), RN−/rn* (33%), rn+/rn+ (8%), rn+/rn* (9%) and rn*/rn* (2%). The pigs were slaughtered at a commercial slaughterhouse and assessed 24 h postmortem. Right sides were fabricated into primary wholesale cuts, then further processed into defatted hams and loins, and a subset of hams (n = 122) was dissected into the five major individual muscles. The genotype frequencies for the subsample were RN−/RN− (27%), RN−/rn+ (20%), RN−/rn* (35%), rn+/rn+ (9%), rn+/rn* (8%) and rn*/rn* (1%). Weights were recorded for meat and bone in ham and loin, fat in ham, back and shoulder and the individual dissected muscles. The genotype effect was significant (P < 0.05) for estimated lean meat content and the proportions of meat and bone and fat in ham and loin (of carcass weight). Also, the content of meat and bone in ham and loin, in proportion of whole ham and loin, respectively, differed significantly (P < 0.01) between genotypes. Estimated lean meat content was highest for RN−/RN− (63.0%) and RN−/rn+ (63.1%) and lowest in the combined group rn*/− (rn+/rn* and rn*/rn*, 61.7%); RN−/rn* (62.5%) and rn+/rn+(62.1%) were intermediate. The same results were found for meat and bone in ham and loin, as a proportion of whole ham and loin, respectively. RN−/RN− and RN−/rn+ did not differ in any trait; however, they produced carcasses with the lowest proportions of fat within loin and the major wholesale cuts (ham, loin and shoulder). The carcass percentage of meat and bone in ham was higher in the three RN−/ genotypes (RN−/RN−, RN−/rn+ and RN−/rn*, P < 0.05) than in the rn*/− group, whereas rn+/rn+ did not (P > 0.05) differ from any of the other genotypes. RN−/rn+ and RN−/rn* had higher (P < 0.05) proportion of meat and bone in loin compared to the rn*/− group. We conclude that the second mutant allele found at the PRKAG3 (RN) locus, rn*, decreased the lean meat content compared with the two other alleles (RN−, rn+). The RN−/RN− and RN−/rn+ genotypes were leanest, followed by RN−/rn* and rn+/rn+, and rn+/rn* and rn*/rn* were the fattest. 相似文献

Measuring shear stress relaxation with an established research rheometer after slowly applying large strain proved useful for characterizing the effects of different chemical and enzymatic additives. Baking tests done with and without added ascorbic acid indicated that the method can be used for predicting effects of such additives on breadbaking quality. The relaxation process for dough is discussed in terms of two flow processes, one occurring at short time periods and the other at longer time periods. The slowly applied large strain shear stress relaxation method provides information about stress relaxation behavior during the second flow process. The effects of two enzymes (protease and glucose oxidase) and two chemical additives (ascorbic acid and potassium bromate) were studied. The results are presented as the stress relaxation curve with the corresponding rate plot and the initial value of the stress. Addition of the oxidizing agents increased the second flow process times and decreased the relaxation rate during the second process. Protease shortened the second flow process times and increased the relaxation rate during the second process. Glucose oxidase separated the two flow processes by increasing the relaxation time of the second process and decreasing the relaxation time of the first process. 相似文献
