The production of Random Amplified Polymorphic DNAs (RAPDs) by Polymerase Chain Reaction (PCR) was used to detect variation in the isolates of Heterobasidion annosum with various geographical origins. Specific RAPD products were detected for each of the intersterility groups: the European S, F, P and the North American S group. There was considerably more polymorphism found among European S than P isolates. The analysis of RAPDs was shown to be a simple and a fast way to generate DNA markers specific to the previously established intersterility groups and thus useful for diagnostic purposes. 相似文献

Urban derelict space can form a valuable complementary element to the formal green spaces of a city: wastelands are often biologically diverse and their unregulated status can provoke innovative spatial activities by a wide range of users. Using the case of an urban brownfield in Tallinn, Estonia, this paper examines how such a space is used and evaluates the magnitude of the impact of minor design interventions – so-called urban acupuncture – on the activities carried out by the users. The study used field observations and behaviour mapping to compare the spatial pattern of the users before and after small design interventions. Although there was widespread use beforehand there was both an increase in use and a different pattern afterwards, which was clearly detectable from the composite behaviour maps of both arrangements, where users follow the properties of the particular environment. The small improvements tested showed the largest effect on the female and the older user groups, raising the number of overall visits and increasing the occurrence of active behaviour almost five times. The research demonstrates that small, inexpensive and possibly temporary interventions can have a major positive effect. 相似文献

Background
The blaZ gene encoding penicillin resistance can be located either chromosomally or on plasmids. The aim of this study was to investigate the genetic relationships and to determine the location of the blaZ gene in S. aureus isolated in bovine mastitis in Finland and Sweden.Methods
Seventy-eight β-lactamase positive S. aureus isolates from bovine mastitis (34 from Finland and 44 from Sweden) were included in the study. The localization of blaZ gene was determined by Southern blotting. The blaZ genes of the isolates were sequenced and the sequences were translated to beta-lactamase proteins and further grouped as different protein signatures. The isolates and, as control, 33 Swedish and 36 Finnish beta-lactamase negative isolates were typed with pulsed-field gel electrophoresis (PFGE).Results
In 26 out of 34 Finnish isolates (76.5%) and in 25 out of 44 Swedish isolates (56.8%) the blaZ gene was localized on a plasmid. Six different protein signatures were found. One signature was found only in four Swedish isolates, but all other signatures were found both in Finnish and Swedish isolates. The PFGE results revealed a diversity of S. aureus clones. The protein signatures were not clearly associated with certain pulsotypes.Conclusions
The plasmid location of the blaZ gene was not statistically significantly more common in Finland than in Sweden, and hence does not explain the higher proportion of penicillin-resistant isolates of S. aureus causing bovine mastitis in Finland compared to Sweden. 相似文献4.

In recent years the potential for evolutionary change to drive ecological dynamics, and vice versa, has been widely recognized. However, the convincing examples of eco-evolutionary dynamics mainly stem from highly artificial experimental systems, with conspicuously few examples contributed by field systems. While rarely considered in the eco-evolutionary literature, the gene-for-gene hypothesis inherently recognizes the tight link between evolutionary and ecological dynamics. The boom-and-bust dynamics of some agricultural pathogens are an extreme demonstration of this. In this perspective, we place plant-pathogen systems in a spatial eco-evolutionary framework, which recognizes that ecology and evolution are tightly linked, take place at the same time scale and are strongly influenced by spatial structure. Specifically, we: i) exemplify how the ecological process of dispersal modifies rapid local coevolutionary dynamics and thereby shapes spatial variation in resistance, infectivity, and local adaptation; and ii) illustrate how the outcome of coevolution (spatial distribution in resistance, infectivity and local adaptation) drives ecological metapopulation processes. Overall, we conclude that both agricultural and wild pathosystems provide a unique illustration of the high relevance of spatial eco-evolutionary feedback in understanding species interactions. 相似文献

Background
Methicillin-resistant staphylococci (MRS) are increasingly being isolated in bovine mastitis. The aim of our study was to evaluate the occurrence of MRS in Finnish mastitis milk samples and characterize the MRS isolates using molecular methods.Results
Methicillin-resistant S. aureus (MRSA) was a rare finding in bovine mastitis in Finland. Only two out of 135 (1.5%) S. aureus isolates were positive for mec genes. One of these carried mecA and was of spa type t172, SCCmec type IV and ST375, and the other harboured mecC, being spa type t3256, and ST130. MRSA ST375 is common among human MRSA isolates in Finland, but this is the first report in the country of bovine mecC MRSA. In coagulase-negative staphylococci (CoNS) originating from bovine mastitis, methicillin resistance was more common. In the two CoNS collections studied, 5.2% (17/324) and 1.8% (2/110) of the isolates were mecA positive. Eighteen of these were methicillin-resistant S. epidermidis (MRSE), which were divided into 6 separate PFGE clusters. One pulsotype was detected in different parts of the country, indicating clonal spread. Most MRSE (13/18) were of SCCmec type IV, one was of type V and four were non-typeable. Comparison with a human staphylococcal database indicated that bovine MRSE strains were not closely related to human MRSE isolates.Conclusions
The occurrence of MRS, especially MRSA, in bovine mastitis in Finland was low. Most methicillin-resistant bovine CoNS are MRSE, and we found evidence of a bovine MRSE strain that may spread clonally. This is the first report of a Finnish bovine isolate of MRSAmecC ST130. The study provides a baseline for further MRS monitoring. 相似文献6.
The effectiveness of retention trees and patches in preserving diversity of nine epiphytic and epixylic old-growth forest lichens was studied in north boreal spruce forests in Finland. We compared (1) 7–8-year-old retention cuts, with at least 5–10 living or dead retention trees per hectare, (2) 10–12-year-old clear-cuts, with some scattered living and dead retention trees on the sites, (3) old-growth spruce forests, and (4) 7-8-year-old retention patches (0.06–0.45?ha) representing the original tree species composition of old-growth forests. The occurrence of indicator lichens was studied on 150 deciduous trees and snags in each forest category. The species richness was significantly higher in old-growth forests than in the clear-cuts and retention cuts, but did not differ between old-growth forests and retention patches. Only three species were found in clear-cuts and two in retention cuts. Foliose cyanolichens Leptogium saturninum and Nephroma bellum thrived on solitary retention trees, whereas humidity-requiring pin lichens from the genus Chaenotheca were found only in old-growth forests and retention patches. Our results suggest that the ability of epiphytic and epixylic species to survive on retained trees depends on several factors: (1) substrate quality (tree species, tree type and diameter of a tree), (2) environmental factors (e.g. humidity, slope exposition), and (3) morphological and physiological characteristics of species. Besides of substrate trees, the retained conifers (esp. spruce) seem to be important in retention patches to provide the shading necessary to maintain humidity. 相似文献

Species diversity of polypores (Basidiomycetes) was compared between woodland key habitats (WKHs) and old-growth forest controls in boreal forests in eastern Finland. WKHs, which were set aside for their rich vascular plant flora, turned out not to be hot spots for the species richness of polypores, nor did their species composition represent the overall species richness of the area. Differences in the total volume of CWD, tree species composition and several CWD qualities were reflected as differences in the species assemblages between the groups. The results indicated that only a fraction of the overall polypore diversity was represented in the small-size WKHs (<0.5 ha), and the protection of red-listed and indicator polypores in WKHs was random. However, rare old-growth forest indicators were found even in small-size WKHs, if the CWD quality was appropriate for them. Nevertheless, our study does not answer the question whether the occurrences of rare and red-listed species will survive in small-size WKHs surrounded by altered environments such as clear-cuts, because our study sites were located in undisturbed areas. The results stress the importance to protect typical old-growth forest patches with high CWD volume and quality in boreal forests. 相似文献

Antimicrobial susceptibility was determined for 805 domestic Campylobacter jejuni isolates obtained from broilers (n = 459), bovines (n = 120), human patients (n = 95), natural waters (n = 80), wild birds (n = 35) and zoo animals/enclosures (n = 16) with known multilocus sequence types (MLST) for 450 isolates. The minimum inhibitory concentration (MIC) values for erythromycin, tetracycline, streptomycin, gentamicin and the quinolones ciprofloxacin and nalidixic acid were determined with the VetMIC method. MICs were compared with MLST types to find possible associations between sequence type and resistance. The proportions of resistant isolates were 5% (broilers), 6.3% (natural waters), 11.4% (wild birds), 11.6% (human patients), 16.7% (bovines) and 31.3% (zoo). The most common resistance among the human and bovine isolates was quinolone resistance alone while resistance to streptomycin alone was most often detected among the broiler isolates and tetracycline resistance was most commonly observed in the wild bird, water and zoo isolates. No or negligible resistance to erythromycin or gentamicin was detected. In all data, 12/26 of the tetracycline‐resistant isolates were also resistant to streptomycin (P < 0.001) and the clonal complex (CC) ST‐1034 CC showed a high proportion of 75% (9/12) of tetracycline‐resistant isolates, most originating from the zoo and broilers with closely associated MLST types from these sources. No association between quinolone resistance and MLST type was seen. The low percentage of resistant isolates among the domestic Campylobacter infections is most probably due to the long‐term controlled use of antimicrobials. However, the higher percentage of tetracycline resistance observed among the zoo isolates could present a risk for zoo visitors of acquisition of resistant C. jejuni. The resistance pattern of tetracycline and streptomycin most often found in ST‐1034 CC could indicate a common resistance acquisition mechanism commonly present in this CC. Overall, MLST typing was found to be a useful method in recognition of potential genetic lineages associated with resistance. 相似文献

Background
The project "Antibiotic resistance in bacteria of animal origin – II" (ARBAO-II) was funded by the European Union (FAIR5-QLK2-2002-01146) for the period 2003–2005, with the aim to establish a continuous monitoring of antimicrobial susceptibility among veterinary laboratories in European countries based on validated and harmonised methodologies. Available summary data of the susceptibility testing of the bacterial pathogens from the different laboratories were collected.Method
Antimicrobial susceptibility data for several bovine pathogens were obtained over a three year period (2002–2004). Each year the participating laboratories were requested to fill in excel-file templates with national summary data on the occurrence of antimicrobial resistance from different bacterial species.A proficiency test (EQAS – external quality assurance system) for antimicrobial susceptibility testing was conducted each year to test the accuracy of antimicrobial susceptibility testing in the participating laboratories. The data from this testing demonstrated that for the species included in the EQAS the results are comparable between countries.Results
Data from 25,241 isolates were collected from 13 European countries. For Staphylococcus aureus from bovine mastitis major differences were apparent in the occurrence of resistance between countries and between the different antimicrobial agents tested. The highest frequency of resistance was observed for penicillin. For Mannheimia haemolytica resistance to ampicillin, tetracycline and trimethoprim/sulphonamide were observed in France, the Netherlands and Portugal. All isolates of Pasteurella multocida isolated in Finland and most of those from Denmark, England (and Wales), Italy and Sweden were susceptible to the majority of the antimicrobials. Streptococcus dysgalactiae and Streptococcus uberis isolates from Sweden were fully susceptible. For the other countries some resistance was observed to tetracycline, gentamicin and erythromycin. More resistance and variation of the resistance levels between countries were observed for Escherichia coli compared to the other bacterial species investigated.Conclusion
In general, isolates from Denmark, England (and Wales), the Netherlands, Norway, Sweden and Switzerland showed low frequencies of resistance, whereas many isolates from Belgium, France, Italy, Latvia and Spain were resistant to most antimicrobials tested. In the future, data on the prevalence of resistance should be used to develop guidelines for appropriate antimicrobial use in veterinary medicine. 相似文献10.
