The heat-induced denaturation curve of ovalbumin followed by the ellipticity at 222 nm in circular dichroism spectra was consistent with that monitored by fluorescence with thioflavin T, which is an indication of amyloid fibril formation, while other proteins such as lysozyme and ovotransferrin did not fluoresce with thioflavin T during heat denaturation. The amount of soluble aggregate formed during heat denaturation was proportional to the increase in fluorescence with thioflavin T. The binding of soluble aggregates with thioflavin T was greatly suppressed in heat-denatured ovalbumin in the presence of thioflavin T. The similar inhibition effect of thioflavin T on the gel formation of heat-induced ovalbumin was observed. These results suggest that the amyloidogenic intermolecular beta-structure is involved in the formation of soluble aggregate and gel of heat-induced ovalbumin. 相似文献

Alterations in the activities of enzymes related to energy metabolism in canine lymphoma cells were investigated. Cytosolic pyruvate kinase (PK) and mitochondrial malate dehydrogenase (MDH) activities in lymphoma cells were significantly higher than those in lymphocytes obtained from lymph nodes of healthy dogs, whereas cytosolic lactate dehydrogenase (LDH) activity was significantly lower in lymphoma cells. The cytosolic M/L ratio (MDH activity/LDH activity), which is considered to be a good indicator of energy metabolism related to glucose utilization in animal tissues, was significantly higher in lymphoma cells than in the normal lymphocytes. 相似文献

Seven novel cell lines from canine histiocytic sarcoma (HS), three of which were disseminated cutaneous HS and four of which were synovial HS, were established. All of the established cell lines had the same morphological (by light and electron microscopic findings), cytochemical (alpha-naphthyl butyrate esterase-positive), and immunohistochemical (vimentin- and lysozyme-positive, and cyto-keratin-negative) characteristics as the original HS tumor cells. All of the established cell lines injected into nude mice subcutaneously produced solid tumors. Because the established cell lines also showed phagocytic and processing activities, the HS tumor cells appear to originate from the mononuclear phagocytic system cells, despite their differences in locations or organs. 相似文献

Epidermal Langerhans cells (LC) express a high-affinity receptor for IgE (FcεRI), consisting of two chains (α and γ chains)
in humans that allows LC to perform Fc receptor-mediated uptake of allergens. We found that canine LC express α and γ chains
but not β chain of FcεRI, identical to human but not to mouse LC, which do not express functional FcεRI (only γ chain is expressed).
This finding indicates that canine LC have FcεRI-mediated function similar to or identical to human LC, raising the possibility
that canine species provides a better model than mouse to understand the pathogenesis of human atopic dermatitis and investigate

Hen egg white lysozyme was lipophilized with short and middle chain saturated fatty acids (caproic, capric, or myristic acid). The yield, bactericidal properties, and structural properties of lipophilized lysozymes were investigated. The yield of lipophilization of lysozyme greatly increased with the decrease in the chain length of fatty acid. Lipophilization broadened the bactericidal action of lysozyme to Gram-negative bacteria with little loss of enzymatic activity. The bactericidal activity increased in proportion to the number of bound short chain fatty acids. The thermal stability of lipophilized lysozyme decreased in proportion to the chain length and number of bound fatty acids. 相似文献

Chicken egg yolk phosvitin showed a remarkable antibacterial effect against Escherichia coli under thermal stress at 50 degrees C. E. coli cells (10(6)/mL) completely disappeared in 1 mL of L-broth coexisting with 0.l mg/mL phosvitin when incubated at 50 degrees C for 20 min, whereas a considerable amount of cells (10(5)/mL) survived at the same thermal stress without phosvitin. Blocking of the chelating effect of phosvitin by the addition of Ca(2+) ion displayed a protective effect against the bactericidal activity at 50 degrees C. In addition, the antibacterial activity of phosvitin was dramatically reduced by treatment with alpha-chymotrypsin, although the chelating effect remained. The surface properties, such as interfacial tension and emulsifying properties of phosvitin, which are an index of the affinity with the outer membrane, were greatly reduced by the alpha-chymotrypsin digestion. This indicates that the alpha-chymotrypsin-digested membrane-penetrating hydrophobic domains at the N- and C-terminal regions play an important role in antibacterial activity. These results suggest that a significant part of the bactericidal activity of phosvitin against E. coli resides in the synergistic effect of the high metal-chelating ability and the high surface activity under the influence of thermal stress. 相似文献

RAD51 forms a complex with BRCA2 and plays a central role in the DNA damage response pathway that is associated with homologous recombination. The structures of RAD51 and its homologues are highly conserved from prokaryotes to higher eukaryotes. Although a large number of BRCA2 mutations have been reported, there are only a few reports on the mutations of RAD51, which have been shown in humans and dogs. However, several mutations of canine RAD51 were identified from mammary gland tumour tissues in a recent study. Some of these mutations seem to have an influence on the homo‐oligomerization or interaction with “Partner and localizer of BRCA2” (PALB2). In this study, we cloned the canine PALB2 homologue and investigated the effect on its interaction with the RAD51 mutants to evaluate the alteration in the function of RAD51 mutants. The A209S and T225S mutants of RAD51 show an attenuation of the interaction between RAD51 and PALB2. These results indicate that the canine RAD51 mutations can potentially alter the homologous recombination pathways in response to DNA damage in dogs. 相似文献

Although androgen therapy resistance and poor clinical outcomes are seen in most canine prostate cancer cases, there are only a few tools for analysing canine prostate cancer by using a cell biological approach. Therefore, to evaluate androgen‐independent neoplastic cell growth, a new canine prostate cancer cell line (CHP‐1) was established in this study. CHP‐1 over‐expressed the co‐chaperone small glutamine‐rich tetratricopeptide repeat‐containing protein α (SGTA), which is over‐expressed in human androgen‐independent prostate cancer. The CHP‐1 xenograft also showed SGTA over‐expression. Although CHP‐1 shows poor androgen receptor (AR) signalling upon dihydrotestosterone stimulation, forced expression of AR enabled evaluation of AR signalling. Taken together, these results suggest that CHP‐1 will be a useful model for investigating the pathogenesis of androgen‐dependent and androgen‐independent canine prostate cancer. 相似文献
