Single phytotoxicity of two representative phthalate esters (PAEs), di-n-butyl phthalate (DnBP) and bis(2-ethylhexyl) phthalate (DEHP), was tested in mung bean (Vigna radiata) seedlings germinated for 72 h in soils spiked with varying concentrations (0-500 mg kg-1 soil) of DnBP or DEHP. PAEs added at up to 500 mg kg-1 soil exerted no significant effect on germination but both pollutants significantly inhibited root elongation (P 〈 0.01); DEHP inhibited shoot elongation (P 〈 0.01) and DnBP depressed biomass on a fresh weight basis (P 〈 0.05). Seedling shoot and root malondialdehyde (MDA) Contents tended to be stimulated by DnBP but inhibited by DEHP. However, increases in superoxide dismutase, peroxidase, ascorbate peroxidase and polyphenol oxidase activities, as well as glutathione (GSH) content, were induced at higher concentrations (e.g., 20 mg kg-1) of both compounds. Accumulation of proline in both roots and shoots and the storage compounds, such as free amino acids and total soluble sugars, in whole plant was induced under the stress exerted by both PAEs. The general responses of mung bean seedlings indicated higher toxicity of DnBP than DEHP on primary growth, during which root elongation was a more responsive index. MDA and GSH were more sensitive parameters in the roots than in the shoots and they might be recommended as physiologically sensitive parameters to assess the toxicity of PAE compounds in soils in future long-term studies. 相似文献

Pentachloronitrobenzene (PCNB) is an organochlorine fungicide that is mainly used in the prevention and control of diseases in crop seedlings. Microbial removal is used as a promising method for in-situ removal of many organic pesticides and pesticide residues. A short-term field experiment (1 year) was conducted to explore the potential role of a PCNB-degrading bacterial isolate, Cupriavidus sp. YNS-85, in the remediation of a PCNB-contaminated soil on which Panax notoginseng was grown. The following three treatments were used:i) control soil amended with wheat bran but without YNS-85, ii) soil with 0.15 kg m-2 of solid bacterial inoculum (A), and iii) soil with 0.30 kg m-2 of solid bacterial inoculum (B). The removal of soil PCNB during the microbial remediation was monitored using gas chromatography. Soil catalase and fluorescein diacetate (FDA) esterase activities were determined using spectrophotometry. In addition, cultivable bacteria, fungi, and actinomycetes were counted by plating serial dilutions, and the microbial biodiversity of the soil was analyzed using BIOLOG. After 1 year of in-situ remediation, the soil PCNB concentrations decreased significantly by 50.3% and 74.2% in treatments A and B, respectively, when compared with the uninoculated control. The soil catalase activity decreased in the presence of the bacterial isolate, the FDA esterase activity decreased in treatment A, but increased in treatment B. No significant changes in plant biomass, diversity of the soil microbial community, or physicochemical properties of the soil were observed between the control and inoculated groups (P<0.05). The results indicate that Cupriavidus sp. YNS-85 is a potential candidate for the remediation of PCNB-contaminated soils under P. notoginseng. 相似文献

Copper (Cu) mine tailings, because of their high content of heavy metals, are usually hostile to plant colonization. A pot experiment was conducted to determine the tolerance of four forage grasses to heavy metals in Cu mine tailings and to examine the variation in the microbial functional diversity of soils from the tailing sites in southern China. All the four grass species survived on Cu mine tailings and Cu mine tailing-soil mixture. However, on pure mine tailings, the growth was minimal, whereas the growth was maximum for the control without mine tailings. The tolerance of grasses to heavy metals followed the sequence: Paspalum notatum 〉 Festuea arundinaeea 〉 Lolium perenne 〉 Cynodon daetylon. The planting of forage grasses enhanced the soil microbial biomass. The Biolog data indicated that the soil microbial metabolic profile values (average well color development, community richness, and Shannon index) of the four forage grasses also followed the sequence: P. notatum 〉 F. arundinaeea 〉 L. perenne 〉 C. daetylon. Thus, P. notatum, under the experimental conditions of this study, may be considered as the preferred plant species for revegetation of Cu mine tailing areas. 相似文献

A pot experiment was carried out to study alleviation of soil acidity and Al toxicity by applying an alkaline-stabilised sewage sludge product (biosolids) to an acid clay sandy loam (pH 5.7) and a strongly acid sandy loam (pH 4.5). Barley (Hordeum vulgare L. cv. Forrester) was used as a test crop and was grown in the sewage sludge-amended (33.5 t sludge DM ha-1) and unamended soils. The results showed that the alkaline biosloids increased soil pH from 5.7 to 6.9 for the clay sandy loam and from 4.5 to 6.0 for the sandy loam. The sludge product decreased KCl-extractable Al from 0.1 to 0.0 cmol kg-1 for the former soil and from 4.0 to 0.1 cmol kg-1 for the latter soil. As a result, barley plants grew much better and grain yield increased greatly in the amended treatments compared with the unamended controls. These observations indicate that alkaline-stabilised biosolids can be used as a liming material for remedying Al phytotoxicity in strongly acid soils by increasing soil pH and lowering Al bioavailability. 相似文献

A lysimeter experiment with undisturbed soil profiles was carried out to study nitrogen cycling and losses in a paddy soll with applications of coated urea and urea under a rice-wheat rotation system in the Taihu Lake region from 2001 to 2003. Treatments for rice and wheat included urea at conventional, 300 (rice) and 250 (wheat) kg N ha^-1, and reduced levels, 150 (rice) and 125 (wheat) kg N ha^-1, coated urea at two levels, 100 (rice) and 75 (wheat) kg N ha^-1, and 150 (rice) and 125 (wheat) kg N ha^-1, and a control with no nitrogen arranged in a completely randomized design. The results under two rice-wheat rotations showed that N losses through both NH3 volatilization and runoff in the coated urea treatments were much lower than those in the urea treatments. In the urea treatments N runoff losses were significantly (P 〈 0.001) positively correlated (r = 0.851) with applied N. N concentration in surface water increased rapidly to maximum two days after urea application and then decreased quickly. However, if there was no heavy rain within five days of fertilizer application, the likelihood of N loss by runoff was not high. As the treatments showed little difference in N loss via percolation, nitrate N in the groundwater of the paddy fields was not directly related to N leaching. The total yield of the two rice-wheat rotations in the treatment of coated urea at 50% conventional level was higher than that in the treatment of urea at the conventional level. AIM: To detect the presence of bovine herpesvirus (BoHV) type 4 in New Zealand dairy cows with clinical metritis.METHODS: Serum samples taken from 92 dairy cows with clinical metritis, each from a different farm, were tested for the presence of antibodies against BoHV-4 using a commercially available, indirect ELISA. Peripheral blood mononuclear cells (PBMC) were collected from 10 BoHV-4 seropositive cows, and PBMC were examined by a pan-herpesvirus nested PCR to detect herpesvirus. PCR products were sequenced directly and a proportion of the PCR products were cloned and sequenced to identify the virus present.RESULTS: Antibodies to BoHV-4 were detected in 23/92 (25%) serum samples. The pan-herpesvirus PCR was positive in 8/10 PBMC samples. Cloning and sequencing identified that all of the eight PCR-positive PBMC contained bovine lymphotropic herpesvirus (BLHV); no BoHV-4 DNA was detected.CONCLUSIONS: This study reports the finding of the presence of apparent antibodies to BoHV-4, and BLHV DNA in New Zealand dairy cows affected by metritis.CLINICAL RELEVANCE: Bovine herpesvirus type 4 and BLHV are reported to have the potential to cause reproduction failure in cows. This is the first report of apparent BoHV-4 antibodies, and BLHV in New Zealand. The importance and epidemiology of these viruses in cattle in New Zealand requires further investigation. 相似文献
