We assessed the safety and efficacy of an experimental canarypox-vectored recombinant canine distemper virus (CDV) subunit vaccine in the Siberian polecat (Mustela eversmanni), a close relative of the black-footed ferret, (M. nigripes), an endangered species that is highly susceptible to the virus. Siberian polecats were randomized into six treatment groups. Recombinant canine distemper vaccine was administered s.c. at three dose levels (10(4.5), 10(5.0), and 10(5.5) plaque-forming units [PFU] per dose) and was administered orally by spraying the vaccine into the oropharnyx at two dose levels (10(5.5), 10(8.0) PFU per dose). The sixth group of control animals was not vaccinated. For both routes of administration, two 1-ml doses of reconstituted vaccine were delivered 4 wk apart, followed by live virus challenge 3 wk after the second vaccination. During the challenge, Synder Hill test strain CDV obtained from the National Veterinary Services Laboratory in Ames, Iowa, was administered i.p. Serial blood samples for CDV serology were collected immediately before vaccination and challenge, and 10, 15, and 20 days after challenge. Clinical signs and body weights were recorded up to 32 days after challenge. The survival rate in animals receiving vaccine at the highest oral dose (10(8.0) PFU per dose) was 83.3%. Survival rate was 50.0% in the high s.c. and 60.0% in the medium s.c. groups. All animals in the low-s.c. dose, low-oral dose, and control groups died after exposure. Vaccine dose overall (oral and s.c.) and dose in response to s.c. administration when considered alone were significant predictors of survival (P = 0.006 and P = 0.04, respectively). Among the polecats challenged with virulent virus, those that died became sick sooner than those that survived. Animals that died lost significantly more weight during the 10 days after challenge than did animals that survived (P = 0.02). Survival rates did not differ by sex, founder female status, or breeding pedigree in any of the treatment groups. Survival rates were higher in animals with increasing serum neutralization titers (P = 0.027). This study demonstrates the efficacy of oral delivery of a recombinant CDV vaccine in the Siberian polecat. Further studies are needed to evaluate the safety and efficacy of vectored recombinant vaccines in highly susceptible species and especially in those species in which vaccination with modified live CDV has led to disease.  相似文献

Provisioning and regulatory ecosystem services of Pacific geoduck clam (Panopea generosa) culture were simulated for an intertidal shellfish farm in Eld Inlet, South Puget Sound, Washington, USA. An individual geoduck clam growth model was developed, based on a well-established framework for modeling bivalve growth and environmental effects (AquaShell?). Geoduck growth performance was then validated and calibrated for the commercial farm. The individual model was incorporated into the Farm Aquaculture Resource Management (FARM) model to simulate the production cycle, economic performance, and environmental effects of intertidal geoduck culture. Both the individual and farm-scale models were implemented using object-oriented programming. The FARM model was then used to evaluate the test farm with respect to its role in reducing eutrophication symptoms, by applying the Assessment of Estuarine Trophic Status (ASSETS) model. Farm production of 17.3 t of clams per 5-year culture cycle is well reproduced by the model (14.4 t). At the current culture density of 21 ind m?2, geoduck farming at the Eld Inlet farm (area: 2684 m2) provides an annual ecosystem service corresponding to 45 Population-Equivalents (PEQ, i.e. loading from humans or equivalent loading from agriculture or industry) in top-down control of eutrophication symptoms. This represents a potential nutrient-credit trading value of over USD 1850 per year, which would add 1.48% to the annual profit (USD 124,900) from the clam sales (i.e. the provisioning service). A scaling exercise applied to the whole of Puget Sound estimated that cultured geoducks provide a significant ecosystem service, of the order of 11,462 PEQ per year (about USD 470,600) in removing primary symptoms of eutrophication, at the level of the whole water body. The modeling tools applied in this study can be used to address both the positive and negative externalities/impacts of shellfish aquaculture practices in the ecosystem and thus the trade-offs of the activity.  相似文献

It is well established that tumour cells have metabolic differences when compared with normal cells. This is particularly true for energy metabolism in which dogs with cancer have been reported to have higher blood insulin and lactate concentrations than control dogs. Moreover, some human and animal studies suggest that the insulin‐like growth factor 1 (IGF‐1) signalling pathway may play a role in tumorigenesis and tumour progression. At present, IGF‐1 has not been evaluated in dogs with multicentric lymphoma. In this prospective, cross‐sectional study, blood levels of IGF‐1, as well as other markers of energy metabolism—insulin, glucose, lactate, and β‐hydroxybutyrate—were measured in 16 dogs with histologically or cytologically confirmed treatment‐naïve lymphoma. These results were compared with 16 age‐, sex‐ and weight‐matched healthy controls. Dietary histories were collected, and protein, fat and carbohydrate intake were compared between groups. Results demonstrated that IGF‐1, insulin, glucose and insulin:glucose ratio were not different between groups. However, lactate and β‐hydroxybutyrate were higher in the dogs with lymphoma than that in the control dogs (1.74 ± 0.83 mmoL/L vs 1.08 ± 0.27 and 2.59 ± 0.59 mmol/L vs 0.77 ± 0.38 mmol/L, respectively). Median dietary protein, fat and carbohydrates did not differ between the groups. This preliminary study suggests that higher insulin and IGF‐1 levels relative to controls may not be a consistent finding in dogs with lymphoma. The significance of increased β‐hydroxybutyrate in dogs with lymphoma warrants further investigation in a larger prospective study.  相似文献

Heart base tumours (HBT) occur commonly in older, brachycephalic dogs. A presumptive diagnosis is made based on location and appearance of the tumour via echocardiogram. Effective treatment options are limited to surgery (when feasible) or radiation therapy. Benefit of medical management is presently unknown. The goal of this retrospective study was to assess the efficacy and tolerability of toceranib phosphate for dogs with HBT. Twenty‐eight dogs with histologically, cytologically confirmed or presumed HBT were evaluated retrospectively. Twenty‐seven dogs were treated with single agent toceranib. One dog received combination therapy with concurrent metronomic chemotherapy. This dog was not included in response or survival analysis. Factors assessed included clinical signs, hematologic/biochemical parameters and response to treatment. For the 27 dogs receiving single agent toceranib, an overall response rate of 10% was found. Overall median survival time was 823 days (range, 68‐1190 days). The overall response rate for the dogs presenting with metastasis was 28.5%, with a median survival time of 532 days (range, 77‐679 days). This was not significantly different than the median survival time of 796 days for dogs who did not present with metastasis. Of the dogs displaying clinical signs at the time of diagnosis, 90% had improvement and 81% had complete resolution of signs after starting toceranib. Toxicity was seen in 54% of dogs with gastrointestinal distress as the most common toxicity but dose reductions were infrequent required. Results demonstrate that toceranib phosphate is a well‐tolerated and effective treatment for inoperable canine heart base tumours including dogs with advanced or metastatic disease.  相似文献

Many of us who are guardians for companion animals were struck with fear when we heard about the massive recall of cat and dog foods during the spring of 2007. Even premium brands were being added to the recall list. Dogs and cats were becoming ill and many died from what appeared to be some sort of food contamination. Eventually, the cause of these pet illnesses and deaths was linked to the intentional adding of melamine to wheat gluten and rice protein imported from China. The melamine combined with cyanuric acid—a by-product of melamine and an additive in some animal feed—caused the formation of crystals in the kidneys of cats and dogs that consumed the tainted food (Dobson et al., 2008).

Shortly after the pet food recalls of 2007, Congress worked to promote better food safety, including the safety of pet food. Sen. Dick Durbin (D-IL) chaired hearings on the pet food recall (American Veterinary Medical Association, 2007). New pet food legislation was included and passed as part of the Food and Drug Administration Amendments Act of 2007, Public Law 110–85. Section 1002 of the Act deals with pet food safety, and this section was codified at 21 U.S.C. §2102. This provision requires the Secretary of Health and Human Services to work with the Association of American Feed Control Officials (AAFCO) and other stakeholders to establish regulations regarding ingredients, processing standards, and labeling for pet food. It also requires the establishment of “early warning surveillance systems and notification during pet food recalls” (21 U.S.C. §2102[b]).  相似文献
