The prevalence of two serotypes of Streptococcus parauberis isolated from the olive flounder, Paralichthys olivaceus, was evaluated in a total of 29 isolates between 2003 and 2010 in Korea. Streptococcus parauberis isolates were divided into two serologically distinct types (serotype 1 and serotype 2), except for one strain (S1091), using an agglutination assay with rabbit antiserum, and serotype 1 was identified as the dominant type (24 of 29 isolates) in this study. To identify the characteristics of the two serotypes of S. parauberis, we conducted a biochemical test using the API 20 Strep kit, a transmission electron microscopy (TEM) assay, sequence analysis of 16S‐23S rRNA intergenic spacer region (ISR) and a pathogenicity test. In TEM, both serotypes possessed polysaccharide capsule layers around the cell surface when bacterial cells were treated with a homologous serotype of rabbit antiserum. However, we were unable to discriminate serotype‐specific biochemical characteristics and genetic characteristics of 16S‐23S rRNA ISR between the two serotypes. In the pathogenicity test, the serotype 1 strains induced significantly higher mortality than the serotype 2 strains in olive flounder when experimentally inoculated via the intraperitoneal route.  相似文献

The antibiotic resistance of 16 Aeromonas (A.) salmonicida strains isolated from diseased fish and environmental samples in Korea from 2006 to 2009 were investigated in this study. Tetracycline or quinolone resistance was observed in eight and 16 of the isolates, respectively, based on the measured minimal inhibitory concentrations. Among the tetracycline-resistant strains, seven of the isolates harbored tetA gene and one isolate harbored tetE gene. Additionally, quinolone-resistance determining regions (QRDRs) consisting of the gyrA and parC genes were amplified and sequenced. Among the quinolone-resistant A. salmonicida strains, 15 harbored point mutations in the gyrA codon 83 which were responsible for the corresponding amino acid substitutions of Ser83→Arg83 or Ser83→Asn83. We detected no point mutations in other QRDRs, such as gyrA codons 87 and 92, and parC codons 80 and 84. Genetic similarity was assessed via pulsed-field gel electrophoresis, and the results indicated high clonality among the Korean antibiotic-resistant strains of A. salmonicida.  相似文献

ABSTRACT: A series of experiments were conducted to examine the effects of salinity, aeration and light intensity on oil globule absorption, feeding incidence, and growth and survival of early-stage Epinephelus coioides larvae. Newly hatched larvae were transferred to 40-L aquaria at a density of 1500 individuals/aquarium. Larvae were exposed to different levels of aeration (0 mL/min per L, 0.62 mL/min per L, 1.25 mL/min per L, 2.50 mL/min per L, or 3.75 mL/min per L); salinity (8 ppt, 16 ppt, 24 ppt, 32 ppt, or 40 ppt); and light intensity (0 lx, 120 lx, 230 lx, 500 lx, or 700 lx) for 4–6 days. Twenty larvae were sampled daily at 11:00 hours to measure for total length (TL), oil globule volume, and feeding incidence. Survival rates were determined by counting the total number of larvae remaining in each aquarium at the end of the experiment. Significantly higher survival rates ( P   <  0.05) were observed at aeration levels of 0.62 mL/min per L and 1.25 mL/min per L, at salinity levels of 16 ppt and 24 ppt, and at light intensities of 500 lx and 700 lx. The influence of aeration level, salinity and light intensity on oil globule absorption, feeding incidence, and growth and survival of early-stage grouper larvae are discussed.  相似文献

The antibacterial effect of the presence of Tilapia hornorum against luminous bacteria in shrimp culture has been reported. This study investigates how the presence of commercially valued marine species such as seabass, snapper and siganid affect the growth of luminous bacteria in shrimp culture water. Results showed that luminous bacterial count of water stocked with seabass, siganid and snapper are significantly lower than those without fish. Therefore this study has demonstrated that seabass, siganid and snapper are alternative species for culture with shrimp to control or inhibit the growth of luminous bacteria in shrimp ponds.  相似文献

Viral nervous necrosis (VNN) is a worldwide disease affecting several species of cultured marine fish. In Korea, VNN has been identified in several species of cultured marine fish. In this study, the authors present data of the amplified nested polymerase chain reaction product (420 bp) of 21 nodavirus strains from different species of apparently healthy wild marine fish on the southern coast of Korea. Phylogenetic analysis based on the partial nucleotide sequence (177 bases) of the RNA2 coat protein gene of 21 strains was highly homologous (93-100%) and closely related to that of the known betanodavirus, redspotted grouper nervous necrosis virus. These results indicate that betanodaviruses occur in large populations of wild marine fish in the southern part of the Korean peninsula, suggesting the importance of these subclinically infected fish as an inoculum source of betanodavirus that is horizontally transmitted to susceptible cultured fish species.  相似文献

Several studies have previously been conducted regarding cell cycle synchronization in mammalian somatic cells. However, limited work has been performed on the control of cell cycle stages in the somatic cells of fish. The aim of this study was to determine the cell cycle arresting effects of several dimethyl sulfoxide (DMSO) concentrations for different times on different cell cycle stages of goldfish caudal fin‐derived fibroblasts. Results demonstrated that the cycling cells or control group (68.29%) yields significantly higher (p < 0.05) arrest in G0/G1 phase compared with the group treated for 24 h with different concentrations (0.5%, 1.0% or 1.5%) of DMSO (64.88%, 65.70%, 64.22% respectively). The cell cycle synchronization in the treatment of cells with 1.0% DMSO at 48 h (81.14%) was significantly higher than that in the groups treated for 24 h (76.82%) and the control group (77.90%). Observations showed that treatment of DMSO resulted in an increase in the proportion of cells at G0/G1 phase for 48 h of culture. However, high levels of apoptotic cells can be detected after 48 h of culture treated with 1% concentration of DMSO.  相似文献

Streptococcosis cause severe losses for global tilapia farming, especially in developing countries. The aim of this study was to identify and characterize streptococci recovered from Nile tilapia farmed in the Philippines. Moribund and apparently healthy fish were sampled from grow-out cages, ponds and hatcheries. Clinical signs observed included exophthalmia, eye opacity, ascites, lethargy, erratic swimming and haemorrhages. Results showed that both Streptococcus iniae and Streptococcus agalactiae were associated with disease in these sites. Consistent with global reports, including those from South-East Asia, S. agalactiae was more widespread than S. iniae. Molecular serotyping of the S. agalactiae isolates identified the serotype Ia and serotype Ib. Histopathological findings were meningitis, meningoencephalitis and septicaemia. Identical virulence profiles were found for all strains of S. iniae, while S. agalactiae strains were separated into virulence profile I and profile II. All strains were susceptible to the tested antibiotics and resistant to oxolinic acid. Only S. agalactiae serotype Ib showed resistance to sulphamethoxazole–trimethoprim. This is the first study from the Philippines to characterize the streptococci involved in disease outbreaks in tilapia aquaculture. Outputs from this study will promote the development of efficacious disease control strategies in tilapia farming for the Philippines and South-East Asia.  相似文献
