The intestinal supply of amino acids (AA) in sheep fed alkaline hydrogen peroxide-treated wheat straw (AHPWS)-based diets supplemented with soybean meal (SBM) or corn grain plus combinations of corn gluten meal (CGM) and blood meal (BM) was measured in a 5 X 5 latin square. Sheep (avg wt 45 kg) with ruminal, duodenal and ileal cannulas were fed diets containing 65% AHPWS supplemented with the following protein sources: soybean meal (SBM), corn gluten meal (CGM), blood meal (BM), 2/3 CGM:1/3 BM and 1/3 CGM:2/3 BM. Total nitrogen (N) flow at the duodenum was not affected (P greater than .05) by protein source. Flows of bacterial N and AA increased (P less than .05) and flows of nonbacterial N and AA decreased (P less than .05) when wethers were fed SBM vs corn plus other protein sources. When diets contained SBM, quantities of total AA at the duodenum were lower (P less than .05) and the profile of AA supplied to the intestine was altered substantially. Total flows of AA at the duodenum and total quantities of AA disappearing from the small intestine were similar (P greater than .05) for all diets containing BM and CGM, but flows and disappearance of valine, histidine, lysine and arginine increased linearly (P less than .05), whereas flows and disappearance of leucine, isoleucine and methionine decreased linearly (P less than .05) as BM replaced CGM in the diets. Results suggest that quantities of individual AA flowing to the duodenum and disappearing from the intestine of wethers fed AHPWS-based diets can be altered by source of dietary protein. Furthermore, feeding protein sources resistant to ruminal degradation in combination may improve the profile of AA supplied to the intestine.  相似文献

ObjectiveTo describe simultaneous pharmacokinetics (PK) and thermal antinociception after intravenous (IV), intramuscular (IM) and subcutaneous (SC) buprenorphine in cats.Study designRandomized, prospective, blinded, three period crossover experiment.AnimalsSix healthy adult cats weighing 4.1 ± 0.5 kg.MethodsBuprenorphine (0.02 mg kg?1) was administered IV, IM or SC. Thermal threshold (TT) testing and blood collection were conducted simultaneously at baseline and at predetermined time points up to 24 hours after administration. Buprenorphine plasma concentrations were determined by liquid chromatography tandem mass spectrometry. TT was analyzed using anova (p < 0.05). A pharmacokinetic-pharmacodynamic (PK-PD) model of the IV data was described using a model combining biophase equilibration and receptor association-dissociation kinetics.ResultsTT increased above baseline from 15 to 480 minutes and at 30 and 60 minutes after IV and IM administration, respectively (p < 0.05). Maximum increase in TT (mean ± SD) was 9.3 ± 4.9 °C at 60 minutes (IV), 4.6 ± 2.8 °C at 45 minutes (IM) and 1.9 ± 1.9 °C at 60 minutes (SC). TT was significantly higher at 15, 60, 120 and 180 minutes, and at 15, 30, 45, 60 and 120 minutes after IV administration compared to IM and SC, respectively. IV and IM buprenorphine concentration-time data decreased curvilinearly. SC PK could not be modeled due to erratic absorption and disposition. IV buprenorphine disposition was similar to published data. The PK-PD model showed an onset delay mainly attributable to slow biophase equilibration (t1/2ke0 = 47.4 minutes) and receptor binding (kon = 0.011 mL ng?1 minute?1). Persistence of thermal antinociception was due to slow receptor dissociation (t1/2koff = 18.2 minutes).Conclusions and clinical relevanceIV and IM data followed classical disposition and elimination in most cats. Plasma concentrations after IV administration were associated with antinociceptive effect in a PK-PD model including negative hysteresis. At the doses administered, the IV route should be preferred over the IM and SC routes when buprenorphine is administered to cats.  相似文献

Seventy-three crossbred steers (initial BW = 170.5 +/- 5.5 kg) from The Ohio State University (Exp. 1) and 216 crossbred steers (initial BW 135.4 +/- 4.4 kg) from the University of Illinois (Exp. 2) were used to determine the effect of source of energy and rate of growth on performance, carcass characteristics, and glucose and insulin profiles on early-weaned steers. Effects of the diets used in Exp. 1 and 2 on ruminal pH and VFA concentrations were quantified using ruminally fistulated steers (Exp. 3). Cattle were weaned at an average age of 119 d in all experiments and were allotted by age, BW, and breed to one of four diets: high-concentrate, fed ad libitum (ALCONC), high-concentrate fed to achieve a gain of either 1.2 kg/d (1.2CONC) or 0.8 kg/d (0.8CONC), or high-fiber, fed ad libitum (ALFIBER). At 218 d of age, all steers were placed on the ALCONC diet until slaughter. Steers were implanted with Compudose at the initiation of all experiments and with Revalor-S when they were estimated to be 100 d from slaughter. When steers in Exp. 1 averaged 181 and 279 d of age, serum samples were collected to determine glucose and insulin concentrations. Steers were slaughtered when a fat thickness of 1.27 cm was reached (Exp. 1) or after 273 d on feed (Exp. 2). In Exp. 1, days in the feedlot (P < 0.01) and age at slaughter (P < 0.01) were lowest for ALCONC and ALFIBER steers, and greatest for 0.8CONC steers. Overall, ADG was greatest for ALCONC and lowest for 0.8CONC steers; feed efficiency was lowest (P < 0.01) for ALFIBER steers. Final BW did not differ (P > 0.57) among treatments. At 181 and 218 d of age, serum insulin was increased (P < 0.10) and intramuscular fat percentage was greatest (P < 0.07), respectively, for ALCONC steers. In Exp. 2, overall ADG (P < 0.06) and final BW (P < 0.04) were greatest for ALCONC and lowest for 1.2CONC and 0.8CONC steers. Overall feed efficiency was greatest for 0.8CONC and lowest for ALFIBER (P < 0.01). Growing phase diet did not affect marbling score at 218 d of age or at slaughter (P > 0.81). In Exp. 3, differences in ruminal pH after feeding may have been a consequence of increasing acetate (ALFIBER), propionate (ALCONC), or a combination of VFA (0.8CONC and 1.2CONC), respectively (diet x time after feeding, P < 0.10). Controlling growth by limit-feeding a high-concentrate diet for only 100 d does not extend the growth curve of early-weaned steers or enhance intramuscular fat deposition at slaughter compared to ad libitum intake of a high-concentrate or high-fiber diet.  相似文献

The persistence of foot-and-mouth disease virus on wool   总被引:1,自引:0,他引:1  
SUMMARY Five Suffolk sheep, held in a high-security isolation room, were exposed for 2 hours to the aerosol of 3 mature pigs that had been infected with foot-and-mouth disease virus (FMDV), strain O1-BFS. The fleeces of 3 of the sheep were contaminated with FMDV at 2 days post exposure (dpe), while at 5 dpe the fleeces of all 5 sheep were more extensively, and more heavily, contaminated. The persistence of FMDV on contaminated wool was examined in vitro using multiple 0.5 g samples of Merino wool that were each contaminated with one of 3 strains of FMDV in tissue-culture medium: O1-BFS, O-Morocco (O-MOR 9/91) or an Asia 1 strain (TAI 1/90). Wool samples were held at either 4°C, 18°C or 37°C, and decay curves were established for each virus at each temperature. These curves predicted that O1-BFS, O-MOR 9/91 and TAI 1/90 would fall below detect-able levels at 72, 70 and 48 days post contamination (pc), respectively, for wool stored at 4°C; at 11, 12 and 12 days pc, respectively, for wool stored at 18°C; and at 57, 68 and 33 hours pc, respectively, for wool stored at 37°C. For wool contaminated with O1-BFS-infected sheep faeces, urine or blood, or with O1-BFS-infected cattle saliva, decay curves predicted virus to persist for 5 to 11 days pc at 18°C. We demonstrated that the simulated scouring of FMDV-contaminated wool at 60° to 70°C would usually reduce virus to below detectable levels. The detergent component of the scouring process had little, if any, antiviral activity, and scouring at 20°C or 50°C had limited impact on FMDV titres . We recommend that either (1) simple storage of FMDV-contaminated wool for 4 weeks at temperatures of 18°C or higher, or (2) scouring of contaminated wool at 60° to 70°C would be sufficient to remove the threat of FMDV-contaminated wool being infectious to other animals .  相似文献

Four Simmental steers with ruminal, duodenal, and ileal cannulas were used to examine effects of dietary forage: concentrate ratio and supply of ruminally degradable true protein on site of nutrient digestion and net ruminal microbial protein synthesis. Steers (345 kg) were fed ammoniated corn cob (high forage; HF)- or corn cob/ground corn/cornstarch (low forage; LF)-based diets supplemented with soybean meal (SBM) or a combination of corn gluten meal and blood meal (CB). Diets were fed at 2-h intervals with average DM intake equal to 2.2% of BW. Feeding LF vs HF increased (P less than .05) OM digestion (percentage of intake) in the stomach, small intestine, and total tract. Efficiency of microbial CP synthesis (EMCP; g of N/kg of OM truly fermented) decreased (P less than .05) for LF vs HF (24.1 vs 26.8), but microbial N and total N flows to the small intestine were similar (P greater than .05) between energy levels (average 112 and 209 g/d, respectively). Total N flows to the small intestine were 13.1% greater (P less than .05) for CB than for SBM because of increased (P less than .05) passage of nonmicrobial N. Feeding SBM vs CB increased (P less than .05) EMCP (27.3 vs 23.3) and microbial N flow to the small intestine (127.5 vs 112.5 g/d), but these increases were not likely due to increased ruminal concentrations of ammonia N (NH3 N). Decreased (P less than .05) incorporation of NH3 N into bacterial N and slower turnover rates of ruminal NH3 N for SBM vs CB suggest that direct incorporation of preformed diet components into cell mass increased when SBM was fed. Results of this study suggest that the inclusion of ruminally degradable protein in the diet may increase the supply of products from proteolysis and that this can increase EMCP and microbial protein flow to the small intestine.  相似文献

Six steers (468 kg) with ruminal and duodenal cannulas were fed diets formulated for two levels of energy containing three crude protein (CP) sources in a 6 X 6 Latin square with a 2 X 3 factorial arrangement of treatments. Energy levels were 2.17 and 2.71 Mcal metabolizable energy (ME)/kg dry matter (DM) provided by hay-corn (H) and corn silage-corn (CS) diets, respectively. Soybean mean (SBM), corn gluten meal-urea (CGM) and urea (U) provided 33% of dietary CP in 12% CP diets. Apparent organic matter (OM) digested in the stomach was not affected (P greater than .05) by energy level or CP source, but OM truly digested in the stomach was greater (P less than .05) when steers were fed the CS compared with the H diet. Duodenal flow of non-NH3 N was greater (P less than .05) when steers were fed CS compared with H and when fed SBM or CGM compared with U. Efficiency of bacterial protein synthesis and duodenal bacterial N flow were increased (P less than .05) when steers were fed CS, but non-NH3, nonbacterial N flow to the duodenum was increased (P less than .05) when steers were fed H. When steers were fed CS rather than H, flows (g/d) of bacterial amino acids were greater (P less than .05), but flows of nonbacterial amino acids tended (P less than .08) to be less. Total amino acid flows were not affected (P greater than .05) by energy level. Duodenal flows of total amino acids tended (P less than .06) to be greater when steers were fed CGM compared with SBM or U, due mainly to an increased (P less than .05) flow of nonessential amino acids.  相似文献
