ABSTRACT A histological study was conducted to provide insights into the defense mechanisms of Pinus banksiana resistant to the European (EU) race of Gremmeniella abietina in naturally infected sites. At the time of sampling, the only apparent symptom was a blight induced at the tip of the shoots. The identity of G. abietina during microscopic examinations was confirmed by an immunogold labeling method. Once the fungus had succeeded in penetrating the bracts through stomata, it invaded the stem cortex and the phloem cells and attained the vascular cambium. The progression of the pathogen to the pith was possible principally through intense colonization of needle traces but also by the invasion of the rays. Ligno-suberized tissues confining the pathogen within the necrotic area were revealed by histochemical tests. Well-defined boundaries were initiated at the base of healthy needles and at the vascular cambium level. They regularly formed one continuous suberized barrier completely crossing the shoot from one needle to the other. A nonlamellar form of suberin was observed in transmission electron microscopy. Restoration of cambial activities and tissue regeneration following necrophylactic periderm formation were suggested as essential factors in the defense system of P. banksiana against the EU race of G. abietina. To our knowledge, this is the first demonstration of an anatomical defense mechanism of a conifer against Scleroderris canker.  相似文献

ABSTRACT The presence of the European (EU) race of Gremmeniella abietina var. abietina, the causal agent of Scleroderris canker of conifers, was first reported in North America in 1975 in the northeastern United States and subsequently in southern Quebec and Newfoundland during the late 1970s, where it quickly became established. We analyzed DNA profiles in samples from a historic collection of G. abietina var. abietina that included some of the first isolates of the EU race reported in the United States to test hypotheses concerning the G. abietina var. abietina epidemic in North America. Genetic diversity was partitioned by an analysis of molecular variance with haplotype frequencies and distances. Genetic differentiation was high between populations in continental North America and Newfoundland (between region differentiation, Phi(ct) = 0.665, P < 0.001). This result was not consistent with the hypothesis of a single introduction of the pathogen into the northeastern United States followed by secondary spread into northeastern Canada. In contrast, small levels of genetic differentiation were observed among continental North American populations (Phi(ct) = 0.047, P = 0.079), suggesting gene flow among these populations. A single haplotype of G. abietina var. abietina dominated the continental populations (80% of the isolates) but was absent from Newfoundland and Europe. Five haplotypes were found in the New-foundland population, all of which were either absent or very rare on the continent. Populations from continental North America clustered together and were distinct from a second cluster composed of European and Newfoundland populations. A phylogenetic analysis of the haplotypes indicated that some of the rare haplotypes may have derived from somatic mutations, whereas others probably occurred as the result of new introductions. The results are consistent with a scenario of distinct primary introductions of this pathogen into Newfoundland and continental eastern North America followed by secondary asexual propagation.  相似文献

Sixteen 10-wk-old, phosphorus (P)-depleted Holstein bull calves were fed for 6 wk a control diet containing .08% P or P-supplemented diets containing .14, .20 or .32% P with supplemental P from two sources (CDP and Dynafos). The diets contained .45, .56, .66 and .87% Ca. After 5 wk of the experiment, the calves were dosed orally with 65Zn, and daily total fecal collections were initiated. At the end of the experimental period, the calves were killed and tissue samples were taken for total Zn and 65Zn analyses. Growth, feed intake and feed efficiency improved with increasing dietary P levels. Level of dietary P and Ca had little or no effect (P greater than .05) on total Zn content of rib, tibia, liver, heart, kidney, muscle or blood. Likewise, 65Zn absorption and content in most tissues were not affected (P greater than .05). The results do not preclude the possibility of some minor effects of P levels on Zn metabolism. However, it is apparent that when adequate Zn is fed, any effects are likely to be of little or no practical importance.  相似文献

We demonstrate the protection of one bit of quantum information against all collective noise in three nuclear spins. Because no subspace of states offers this protection, the quantum bit was encoded in a proper noiseless subsystem. We therefore realize a general and efficient method for protecting quantum information. Robustness was verified for a full set of noise operators that do not distinguish the spins. Verification relied on the most complete exploration of engineered decoherence to date. The achieved fidelities show improved information storage for a large, noncommutative set of errors.  相似文献

Treatment of diabetes mellitus (DM) is a common disease, and nutrition has become an integral part of management. This article focuses on the role of nutrition and dietary ingredients in OA, evaluating current evidence for obesity management, omega-3 fatty acids, and chondromodulating agents. Feeding either a highly digestible, moderate-carbohydrate diet or a highly digestible, high-protein, low-carbohydrate diet resulted in significant (P < .05) improvements in fecal scores in 71% of cats with chronic, nonspecific diarrhea. Approximately 58% of the cats improved on either diet, with no significant differences between the two diets regarding the percentage of cats responding or the degree of response. These results suggest that dietary management may be helpful in cats with chronic diarrhea. If cats do not respond within 1 month, an alternative diet should be considered.  相似文献

The goal of this article was to review the evidence surrounding the risks posed by insufficient or excessive dietary protein. Dietary protein is required to provide essential amino acids and replenish protein reserves. When intake is deficient, protein turnover slows and lean body mass is gradually depleted. These changes lead to increased morbidity and mortality. Dogs can maintain nitrogen balance (typically used to define minimum requirements in adults), yet be in a protein-depleted state due to physiologic adaptations. Preservation of protein turnover and lean body mass requires about threefold more protein than nitrogen balance. The ability of excess dietary protein to induce renal pathology was studied in both dogs with chronic kidney failure and older dogs without kidney failure. Numerous studies have confirmed that protein does not adversely affect the kidneys. However, phosphorus- and protein-restricted diets are clinically beneficial in dogs with existing chronic kidney failure. Protein restriction for healthy older dogs is not only unnecessary, it can be detrimental. Protein requirements actually increase by about 50% in older dogs, while their energy requirements tend to decrease. When insufficient protein is provided, it can aggravate the age-associated loss of lean body mass and may contribute to earlier mortality. Older dogs should receive at least 25% of their calories from protein, typically provided by diets containing at least 7 g protein/100 Kcal ME.  相似文献

Elevated concentrations of serum phosphate are linked with progression and increased case fatality rate in animals and humans with chronic kidney disease. Elevated concentrations of serum phosphate can be a risk factor for development of renal and cardiovascular diseases or osteoporosis in previously healthy people. In rodents, an excess intake of dietary phosphorus combined with an inverse dietary calcium : phosphorus ratio (<1 : 1) contributes to renal calcification. Renal injury also has occured in cats fed experimental diets supplemented with highly soluble phosphate salts, especially in diets with inverse calcium : phosphorus ratios. However, not all phosphorus sources contribute similarly to this effect. This review, which focuses on cats, summarizes the published evidence regarding phosphorus metabolism and homeostasis, including the relative impact of different dietary phosphorus sources, and their impact on the kidneys. No data currently shows that commercial cat foods induce renal injury. However, some diets contain high amounts of phosphorus relative to recommendations and some have inverse Ca : P ratios and so could increase the risk for development of kidney disease. While limiting the use of highly soluble phosphates appears to be important, there are insufficient data to support a specific upper limit for phosphate intake. This review also proposes areas where additional research is needed in order to strengthen conclusions and recommendations regarding dietary phosphorus for cats.  相似文献

Twenty-four crossbred beef cows were used to investigate the concentration of alpha-tocopherol in plasma and tissues following oral administration of four tocopherol sources. Animals were allotted to the following treatments: DL-alpha-tocopherol, D-alpha-tocopherol, DL-tocopheryl acetate and D-alpha-tocopheryl acetate. Animals received a daily oral dose of 1,000 IU of the respective tocopherol treatment for 28 d and then were slaughtered. Blood samples were collected on d 0, 1, 7, 14 and 28 for tocopherol concentration assays, and samples from 10 different tissues were collected from slaughtered cows. Identification of alpha-tocopherol in tissues was confirmed by HPLC retention times and by comparison of mass spectra with that of alpha-tocopherol standards. The D-alpha-tocopherol and its acetate ester increased plasma tocopherol concentration faster than the racemic products, the greatest response occurring with D-alpha-tocopherol. Across all treatments, the highest alpha-tocopherol concentrations were noted in the adrenal gland and liver, the lowest in muscle and thyroid tissue. Tissue analyses confirmed that in adrenal gland, kidney, liver and lung, alpha-tocopherol concentrations were higher following D-alpha than DL-alpha-tocopherol supplementation.  相似文献
