Cloning through seeds has potential revolutionary applications in agriculture, because it would allow vigorous hybrids to be propagated indefinitely. However, asexual seed formation or apomixis, avoiding meiosis and fertilization, is not found in the major food crops. To develop de novo synthesis of apomixis, we crossed Arabidopsis MiMe and dyad mutants that produce diploid clonal gametes to a strain whose chromosomes are engineered to be eliminated after fertilization. Up to 34% of the progeny were clones of their parent, demonstrating the conversion of clonal female or male gametes into seeds. We also show that first-generation cloned plants can be cloned again. Clonal reproduction through seeds can therefore be achieved in a sexual plant by manipulating two to four conserved genes.  相似文献

We synthesized multimetal microrods intrinsically encoded with submicrometer stripes. Complex striping patterns are readily prepared by sequential electrochemical deposition of metal ions into templates with uniformly sized pores. The differential reflectivity of adjacent stripes enables identification of the striping patterns by conventional light microscopy. This readout mechanism does not interfere with the use of fluorescence for detection of analytes bound to particles by affinity capture, as demonstrated by DNA and protein bioassays.  相似文献

In low Ca soils, gypsum is applied at flowering of peanut (Arachis hypogaea L.) to insure adequate availability of Ca in the fruiting zone (0 to 8 cm soil depth) during pod development. Effects of 15.2 cm water applied as 1, 2, 4 or 12 split applications over 28-d period following the application of gypsum on the distribution of Ca and other cations in the fruiting zone and immediately below the fruiting zone (8 to 16 cm depth) were investigated on the Lakeland and Tifton soils under controlled conditions in rainout shelter plots. In Lakeland soil, the fruiting zone Ca concentration was greater for the high frequency (15.2 cm in 4 or 12 split applications) irrigation treatments than for the low frequency (15.2 cm in 1 or 2 split applications) treatments at 7 d. Those differences became non-significant after 14 d. In Tifton soil, leaching of Ca below the fruiting zone was significantly greater for the low frequency irrigation treatments than for the high frequency treatments throughout the duration of this e experiment. High frequency irrigation of moderate quantities should result in low Ca losses from the fruiting zone. Irrigation following gypsum application decreased K and Mg concentrations in the fruiting zone for both soils. The gypsum-induced leaching of K or Mg was lower in high frequency irrigation treatments than in low frequency treatments. High frequency irrigation following gypsum application resulted in a decrease in soil pH during the initial 7 d.  相似文献

We hypothesized that ruminants discriminate among different mineral supplements and modify their selection as a function of need. Forty lambs were offered four‐way choices among P, Ca, and Mg sources: monosodium phosphate (MSP), Magnesium phosphate (MGP), Monocalcium phosphate (MCP), and Dicalcium phosphate (DCP); and two‐way choices between DCP and the other mineral sources (Baseline). Subsequently, lambs were randomly assigned to four groups (10 lambs/group) and fed rations such that levels of Ca and P were low (LCa_LP), adequate (NCa_NP), low in Ca and adequate in P (LCa_NP), or adequate in Ca and low in P (NCa_LP). Four‐ and two‐way choice tests were repeated (Post‐deficiency). During Baseline, lambs ate and preferred MSP > MGP>DCP = MCP (p < .0001), showing that they discriminated among minerals. This pattern remained during four‐way Post‐deficiency tests, but lambs in LCa_LP, LCa_NP, and NCa_LP showed increased preference for MCP, MGP, and DCP, respectively, relative to other groups and Baseline (p < .05). Some mineral preference also changed in relation to serum concentration of P and Ca. Thus, lambs discriminated among minerals and some groups modified their preferences from Baseline to Post‐deficiency choice tests, likely influenced by mineral needs, which in turn contributed to rectify mineral imbalances.  相似文献

Context

Wild flowering plants and their wild insect visitors are of great importance for pollination. Montane meadows are biodiversity hotspots for flowering plants and pollinators, but they are contracting due to tree invasion.

Objectives

This study quantified flowering plants and their flower-visitor species in montane meadows in the western Cascade Range of Oregon. Species diversity in small, isolated meadows was expected to be lower and nested relative to large meadows. Alternatively, landform features may influence richness and spatial turnover.

Methods

Flowering plants and their visitors were sampled in summers of 2011–2017 in twelve montane meadows with varying soil moisture. All flowering plants and all flower-visitors were recorded during five to seven 15 min watches in ten 3?×?3 m plots in each meadow and year.

Results

A total of 178 flowering plant species, 688 flower-visitor species and 137,916 interactions were identified. Richness of flower-visitors was related to meadow patch size, but neither plant nor flower-visitor richness was related to isolation measured as meadow area within 1000 m. Species in small meadows were not nested subsets of those in large meadows. Species replacement accounted for more than 78% of dissimilarity between meadows and was positively related to differences in soil moisture.

Conclusions

Although larger meadows contained more species, landform features have influenced meadow configuration, persistence, and soil moisture, contributing to high plant and insect species diversity. Hence, conservation and restoration of a variety of meadow types may promote landscape diversity of wild plants and pollinators.
