A systematic immunohistochemical study of the ultimobranchial tubule (UBT) has been carried out in 45 Wistar rats of different ages (0, 5, 10, 15, 20, 25, 30, 60 and 120 days). The existence of calcitonin immunoreactive cells in the UBT wall has been demonstrated in a 5-days old rat. In addition, immunohistochemical studies for thyroglobulin revealed positive staining in follicular cells connected to the UBT and, occasionally, in isolated cells lying within solid clusters from the UBT. These last results together with the continued and repeated existence of numerous mitosis and PAS (+) microfollicles, apparently rising from the UBT, support the hypothesis that the ultimobranchial body (UBB) may contribute partially to the formation of a part of the follicular component.  相似文献

Lymph nodes, spleen, liver, lung and kidney obtained from pigs experimentally infected with two African Swine Fever Virus (ASFV) isolates of differing virulence were fixed by perfusion with glutaraldehyde and embedded in paraffin. An immunoperoxidase technique using a polyclonal anti-ASFV serum was performed on tissue sections in order to detect ASFV antigen. The distribution of ASFV antigen in such infected organs is shown and the differences between both infections compared and discussed. Monocytes, macrophages, hepatocytes, endothelial cells, neutrophils and epithelial cells were found to contain ASFV antigens.  相似文献

Viruses in the genus Mastrevirus (family Geminiviridae), including those infecting sugarcane, have natural geographical ranges almost exclusively restricted to Africa and the Indian Ocean islands off the African coast. Only sugarcane white streak virus (SWSV) in Barbados and sugarcane striate virus (SStrV) in Florida and Guadeloupe are known to infect a few sugarcane varieties in the Western Hemisphere. In this study, PCR assays were developed to detect these two viruses in sugarcane. Five hundred and seventy-one DNA samples from Saccharum species and interspecific hybrids from the Miami World Collection of sugarcane and related grasses were tested for the presence of SStrV and SWSV by PCR. No variety was found infected by SWSV but SStrV was detected in 19 varieties. PCR data were confirmed by sequencing amplified fragments (248 bp). These fragments shared 93%–100% nucleotide identity with SStrV sequences from the GenBank database. SStrV isolates were distributed in six phylogenetic groups, including the four strains of the virus. Most varieties infected by SStrV originated from Asia, thus confirming a previous hypothesis stating that this virus originated from this continent. Absence of SStrV in commercial sugarcane in Florida also suggested that this virus has not been spread in this location, while infected plants have been present for several decades.  相似文献

OBJECTIVE: To determine clinical characteristics and mode of inheritance of seizures in a family of Standard Poodles. DESIGN: Case series. ANIMALS: 90 Standard Poodles descended from the same maternal bloodline (30 with probable idiopathic epilepsy [PIE] and 60 without any history of seizures). PROCEDURES: Researchers contacted owners to determine whether dogs had ever had any seizures and, if so, the nature of any such seizures and any potential underlying causes. Dogs were considered to have PIE if they were between 6 months and 7.5 years old at the time of seizure onset and had no evidence of any underlying cause. To determine the mode of inheritance, segregation analyses were designed to allow the family to be analyzed as a whole, as opposed to as nuclear families. Competing models of inheritance were compared statistically for their ability to explain the data. RESULTS: Of the dogs with PIE, 28 (93%) had focal onset seizures with or without secondary generalization. Median age of onset was 3.7 years; 6 dogs were > 5 years old at the onset of seizures. Segregation analyses strongly suggested that PIE was inherited as a simple recessive autosomal trait with complete or almost complete penetrance. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that in this family of Standard Poodles, PIE was inherited as a simple recessive autosomal trait with complete or almost complete penetrance. Seizures often had focal, as opposed to generalized, onsets, and it was not uncommon for seizures to begin after 5 years of age.  相似文献

Two experiments were conducted to determine the effects of dietary Cr tripicolinate (CrPic) or Cr propionate (CrProp) on growth, carcass traits, plasma metabolites, glucose tolerance, and insulin sensitivity in pigs. In Exp. 1, 36 barrows (12 per treatment; initial and final BW were 20 and 38 kg) were allotted to the following treatments: 1) corn-soybean meal basal diet (control), 2) as 1 + 200 ppb Cr as CrPic, or 3) as 1 + 200 ppb Cr as CrProp. Growth performance data were collected for 28 d, and then 23 pigs (seven, eight, and eight pigs for treatments 1, 2, and 3, respectively) were fitted with jugular catheters and a glucose tolerance test (500 mg glucose/kg BW) and an insulin challenge test (0.1 IU of porcine insulin/kg BW) were conducted. Both CrPic and CrProp decreased (P < 0.05) ADG and ADFI but did not affect gain:feed (P > 0.10). Fasting plasma glucose, total cholesterol, urea N, insulin, and high-density lipoprotein cholesterol:total cholesterol concentrations were not affected (P > 0.10) by either Cr source. Pigs fed CrPic had lower (P < 0.02) fasting plasma NEFA concentrations than control pigs, but plasma NEFA concentrations of pigs fed CrProp were not affected (P > 0.10). During the glucose tolerance test, glucose and insulin kinetics were not affected by treatment (P > 0.10). During the insulin challenge test, glucose clearance was increased (P < 0.01) in pigs fed CrProp but not affected (P > 0.10) in pigs fed CrPic. Glucose half-life was decreased (P < 0.03) in pigs fed CrPic or CrProp, but insulin kinetics were not affected (P > 0.10). In Exp. 2, 48 barrows (four replicates of four pigs per replicate; initial and final BW were 23 and 115 kg) were allotted to the same dietary treatments in a growing-finishing study. Average daily gain, ADFI, and gain:feed were not affected (P > 0.10) by treatments. Carcass length tended (P = 0.10) to be greater in pigs fed CrPic than in pigs fed CrProp, but other carcass measurements were not affected (P > 0.10). Glucose kinetics from the insulin challenge test indicate that both CrPic and CrProp increase insulin sensitivity and that both Cr sources are bioavailable.  相似文献

ABSTRACT Fusarium oxysporum f. sp. canariensis causes Fusarium wilt disease on the Canary Island date palm (Phoenix canariensis). To facilitate disease management, a polymerase chain reaction diagnostic method has been developed to rapidly detect the pathogen. A partial genomic library of F. oxysporum f. sp. canariensis isolate 95-913 was used to identify a DNA sequence diagnostic for a lineage containing all tested isolates of F. oxysporum f. sp. canariensis. Two oligonucleotide primers were designed and used to amplify a 567-bp fragment with F. oxysporum f. sp. canariensis DNAs. DNA from 61 outgroup isolates did not amplify using these primers. Once the primers were shown to amplify a 0.567-kb fragment from DNA of all the F. oxysporum f. sp. canariensis isolates tested, a rapid DNA extraction procedure was developed that led to the correct identification of 98% of the tested F. oxysporum f. sp. canariensis isolates.  相似文献

ABSTRACT The role of NaCl in suppression of Fusarium crown and root rot of asparagus was investigated in split root culture so the direct effects of NaCl on the root and rhizosphere could be separated from effects that were translocated to the nontreated root side. One side of the root system was exposed to 100 ml of 0, 0.5, or 1.0% NaCl, while the other side received deionized water. Both sides of the root system were inoculated with conidial suspensions of the pathogens Fusarium oxysporum and F. proliferatum. When plants were harvested and assayed, root lesions and CFUs of F. oxysporum or F. proliferatum per centimeter of root from both exposed and nonexposed roots decreased as the NaCl rate increased to 1.0%, but the reduction relative to the control was significantly greater on roots that were directly exposed to NaCl (51% reduction in root lesions) than on adjacent nonexposed roots (31% reduction in root lesions). On both sides of the root systems, disease suppression with NaCl was associated with increases in the rhizosphere densities of fluorescent pseudomonads and Mn-reducing bacteria in the rhizosphere soil. In addition, as the NaCl rate increased, root tissues had marked reductions in malic acid and amino acids while concentrations of Cl and Mn increased in equal proportions on both sides of the root system. Chloride ions were absorbed in greater amounts than Na ions, and were more mobile in the plant than Na. Plants treated with 1% NaCl (171 meq of Cl(-) per liter) had soil leachates 1 week later of 47 meq of Cl(-) per liter from pots exposed to NaCl, but in the adjacent nonexposed pots, the amount of Cl in the leachates slowly increased over the course of the study to 20 meq/liter, presumably through the root exudation. These findings suggest that suppression of Fusarium crown and root rot of asparagus with NaCl may be due to multiple mechanisms. Maximum suppression occurs when NaCl is directly applied to roots, but suppression still occurs on distal non-treated roots resulting from systemic mechanisms. The latter mechanism may be associated with a root-mediated alteration in the rhizobacteria.  相似文献
