Holstein bull calves, 8 to 12 wk of age, were anesthetized with halothane gas. An approximate 20-cm section of small intestine, 60 to 90 cm proximal to the ileocecal junction was clamped to isolate blood circulation to a single set of arcuate vessels and to form an intestinal segment fitted for infusion and drainage. The vein was catheterized to allow total venous collection. Donor blood was transfused via jugular vein to replace venous drainage. This technique was evaluated in four calves by exposing the lumen to eight replications (12 or 20 min incubation, 30-min wash with 39 C saline) of 16 mM L-Met (14C-labeled). Time course appearance of Met in venous blood indicated similar rates and patterns of absorption for individual calves. There were no clinically significant alterations in jugular blood chemistry profiles across replications. Four calves were used to evaluate the effect of three isotonic perfusion media (saline, Krebs-Ringer bicarbonate and M-199 tissue culture media) on Lys and Met absorption. Venous flow rates and absorption of Lys were faster with Krebs buffer than with other media. Perfusate medium did not influence venous flow rates or absorption of Met. Effect of restricting venous flow on absorption of Lys and Met was evaluated in two calves. Flow was alternately controlled (6.5 ml/min) or allowed to flow freely (mean = 12.2 ml/min). Restricting flow decreased steady-state absorption. Light and scanning microscopy indicated maintenance of mucosal tissue integrity throughout 8 h of anesthesia. Results demonstrate validity of the in situ technique to study nutrient absorption in the young bovine.  相似文献

A 2 1/2-month-old, female German Shepherd Dog was examined because of incontinence since birth. Multiple urinary anomalies which included left renal agenesis, right hydronephrosis and hydroureter and urinary bladder dysfunction were found by excretory urography. The aim of this study was to compare three different enzyme-linked immunosorbant assays (recombinant major antigenic protein 2 (rMAP2)-ELISA, the Immunocomb (Biogal, Israel) and the Snap 3Dx assay (IDEXX Laboratories Inc., USA)) with the indirect immunofluorescent antibody test in detecting anti-Ehrlichia canis immunoglobulin-G (IgG) antibodies. Samples tested were collected from dogs suspected to be naturally infected with E. canis and from experimentally infected dogs.When qualitative results (positive/negative) were compared, there was an overall agreement of 81% (54/67) between the indirect immunofluorescence antibody (IFA) test and the rMAP2-ELISA. An overall agreement of 94% (63/67) was found between the IFA test and the Immunocomb, and an overall agreement of 91% (61/67) was found between the IFA test and the Snap 3Dx assay. In 50 of 67 (74.6%) samples tested, complete agreement in the qualitative results was found in all four tests. Sixteen of 17 samples with disagreement in the qualitative results were found to have IFA titers of 1:320 or less. The sensitivities and specificities of the tests were found to be 0.71 and 0.85 for the rMAP2-ELISA, 0.86 and 0.98 for the Immunocomb, and 0.71 and 1.00 for the Snap 3Dx assay.The tests performed in this study were found to be highly specific in detecting E. canis antibodies. Their sensitivity was found to be low with sera having IFA titers of < or =1:320, while high with sera having titers greater than 1:320. Repeating the serological tests 1-2 weeks after the first antibody assay may overcome the sensitivity problem with titers of < or =1:320.  相似文献

We contrasted the regeneration success of angiosperm canopy species with that of Podocarpus latifolius to test whether this conifer was sufficiently shade tolerant to be favoured on infrequently disturbed and well-shaded sites. Seedling and sapling population structures were measured in several habitats representing a light gradient in warm temperate forest in the Drakenberg mountains of South Africa. The angiosperm-dominated forest was well-shaded (∼5.5% PAR) and lacked ground vegetation. Seedlings of angiosperm canopy species were abundant but the absence of saplings indicated regeneration failure. In contrast, P. latifolius was represented by all size classes beneath the intact canopy indicating continuous regeneration in angiosperm-dominated forest. Angiosperm regeneration was similarly poor in forest gaps, which were dominated by grasses, ferns and vines. P. latifolius entered the advanced regeneration in gaps by establishing in shade before gap formation. The floor of the Podocarpus-dominated forest was less shaded (∼7.5% PAR) than angiosperm-dominated forest and dominated by grass, which suppressed most conifer regeneration. Gaps in Podocarpus forest were more than double the size of gaps in angiosperm forest and dominated by vines and understorey shrubs. There was no clear winner of the angiosperm–conifer contest in Podocarpus forest gaps. Consequently, gaps maintain limited angiosperm diversity in Podocarpus forest. Angiosperm canopy species regenerated continuously in the high light (∼11.5% PAR) thicket environment. Few seedlings of P. latifolius were recorded in the thicket environment. Our findings are consistent with the hypothesis that shaded and infrequently disturbed forest sites favour shade-tolerant conifers over relatively light-demanding angiosperm species. The outcome of the competitive interaction between angiosperm and conifer at the regeneration phase depends on the relative shade-tolerance of associated species.  相似文献

The purpose of the study was to determine the ability of Doppler ultrasound to detect changes in femoral blood flow during pharmacologic manipulation of arterial blood pressure. Doppler ultrasonography was performed in the femoral vessels of six halothane-anesthetized horses before and during administration of phenylephrine HCI and sodium nitroprusside. The time-averaged mean velocity and volumetric flow were calculated. The contour of the velocity waveform was assessed, and the early diastolic deceleration slope (EDDS) and pulsatility index (PI) were calculated. Administration of phenylephrine HCI resulted in increased mean aortic blood pressure (MABP) by 40% (29.3-53.0%). This caused significant decrease in cardiac output (26.8 to 13.5 l/min), femoral arterial velocity (left artery 7.20 to 4.00 cm/s; right artery 5.01 to 3.39 cm/s) and volumetric flow (left artery 556 to 221 ml/min; right artery 397 to 193 ml/min) in the femoral vessels and significant increase in systemic vascular resistance (163 to 433 dyn-s/cm5), EDDS (1a: 285 to 468: ra: 250 to 481) and PI (1a: 9.38 to 20.4; ra 17.1 to 29.1). Administration of sodium nitroprusside resulted in a decreased MABP of 27.2% (22.5-33%). This increased cardiac output (20.8 to 32.4 L/min), however, no significant changes were observed in femoral blood flow. Despite obvious changes in the waveform contour, no significant change occurred in EDDS or PI. These results suggest that Doppler ultrasound may be useful for measuring femoral blood flow in anesthetized horses. However, waveform analysis appears to be limited when multiple changes occur in central and peripheral haemodynamics.  相似文献

In order to evaluate whether infection with E. canis alters the expression of major histocompatibility complex (MHC) class I and/or MHC class II receptors, and by doing so alters the immune response to the organism, flow cytometry was performed on DH82 cells infected with Ehrlichia canis (90% infection) and on uninfected DH82 cells of the same passage, using anti-canine MHC class I and II antibodies. MHC class II expression was evident in 47.6 and 46.2% (mean 46.9%) of uninfected DH82 cells using two different anti-MHC class II antibodies, while no MHC class II expression was evident in DH82 cells infected with E. canis. The present results indicate that infection of DH82 macrophages with E. canis down-regulates their MHC class II receptors. These results suggest a possible mechanism by which E. canis evades the immune system.  相似文献
